A formaldehyde-free adhesive consisting of a corn flour/NaOH adhesive mixture and a mimosa tannin/hexamine intermediate component was developed and evaluated for application to wood panels such as particleboards. The main ingredients of this adhesive include corn flour, NaOH, mimosa tannin and hexamine. This study investigated the physical properties (rheological and thermal analysis) of corn flour/NaOH and mimosa tannin/hexamine adhesives as well as mechanical properties of particleboards produced with these adhesives. Thermomechanical (TMA) experiments indicate the best performance of the adhesives to be around the relative mass proportions of 50:50 between corn flour/NaOH and mimosa tannin/hexamine. Former NMR measurements on corn starch and mimosa tannin proved that the two components behave as a polymer blend rather than co-reacting. The laboratory results show that particleboards bonded with the adhesive at this optimal ratio show good mechanical properties. Moreover, the formaldehyde emission levels obtained from boards bonded with the optimal adhesive were considerably lower to those obtained from boards made with control urea formaldehyde.  相似文献

Cooling of equine semen obtained from some stallions results in lower seminal quality and viability when the seminal plasma (SP) is present. The objective of this study was to evaluate the effect of the removal of SP using a Sperm Filter on the viability of cooled stallion semen. For this purpose, 31 stallions were used. Their ejaculates were divided into three groups: CN, semen was diluted with an extender; FLT, SP was removed by filtration; and CT, SP was removed by centrifugation and cooled to 15°C for 24 hours. Sperm kinetics and plasma membrane integrity were evaluated immediately after collection (T0) and after 24 hours of refrigeration (T1). No difference (P > .05) was noted at T1 for total sperm motility (TM), progressive sperm motility, or plasma membrane integrity when semen samples from all the stallions were analyzed. However, when samples from stallions termed “bad coolers” were analyzed (TM = <30% at T1), a difference was observed in TM and progressive sperm motility for CN compared with FLT and CT at T1. Sperm recovery was greater when SP was removed using the filter (FLT) to that when the SP was removed by centrifugation (CN) (89% vs. 81%). Thus, we concluded that filtering with a Sperm Filter is an efficient and practical method for removal of SP from stallion ejaculates, with lower sperm loss than centrifugation. We also found that the presence of SP reduces the quality and viability of cooled semen from stallions whose semen is sensitive to the process of refrigeration.  相似文献

A halotolerant, gram positive, motile and rod, was isolated from the E1 Gol6a lake of Ghardaia, Alegria and screened for its antimicrobial potential. The strain showed an inhibitory effect to gram positive and gram negative bacteria against Cladosporium spp. and a slight inhibitory effect against C. albicans using chloroform as the extraction solvent and the nutrient broth as production medium. 16S rRNA sequencing showed that the isolate named LMB3981 is a new taxon in the family of Bacillaceae (with 96% similarity). The strain is close to Filobacillus milosensis and Bacillus haloalkaliphilum with 95% similarity. The phenotypic study showed differences between LMB3981 and two strains that are near and have confirmed the results of 16S rRNA sequencing by specific metabolic properties of the strain.  相似文献

Background

The aim of this study was to fabricate the poly caprolactone (PCL) aligned nanofiber scaffold and to evaluate the survival, adhesion, proliferation, and differentiation of rat hair follicle stem cells (HFSC) in the graft material using electrospun PCL nanofiber scaffold for tissue engineering applications.

Methods

The bulge region of rat whisker was isolated and cultured in DMEM: nutrient mixture F-12 supplemented with epidermal growth factor. The morphological and biological features of cultured bulge cells were observed by light microscopy using immunocytochemistry methods. Electrospinning was used for production of PCL nanofiber scaffolds. Scanning electron microscopy (SEM), 3-(4, 5-di-methylthiazol- 2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay, and histology analysis were used to investigate the cell morphology, viability, attachment and infiltration of the HFSC on the PCL nanofiber scaffolds.

Results

The results of the MTT assay showed cell viability and cell proliferation of the HFSC on PCL nanofiber scaffolds. SEM microscopy images indicated that HFSC are attached, proliferated and spread on PCL nanofiber scaffolds. Also, immunocytochemical analysis showed cell infiltration and cell differentiation on the scaffolds.

Conclusion

The results of this study reveal that PCL nanofiber scaffolds are suitable for cell culture, proliferation, differentiation and attachment. Furthermore, HFSC are attached and proliferated on PCL nanofiber scaffolds.Key Words: Nanofiber, Electrospinning, Stem cells, Tissue engineering  相似文献

We studied seed morphometric characters, oil content and azadirachtin content in a medicinally important and insect repellent tree species Azadirachta indica A. Juss. Ten populations were selected from two central Indian states of Madhya Pradesh and Chhattisgarh and twenty candidate plus trees were selected from each population based on tree attributes. Azadirachtin content was estimated in seeds, in vitro shoots and callus cultures through HPLC. Significant variation (p < 0.05) was observed in seed morphometric characters, oil content and azadirachtin content among the ten populations. Maximum value for 100 seed weight was obtained in seeds of Raigarh (30.51 g) which was on par with seeds of Bargi (28.37 g) and Chhatarpur (26.36 g). The ratio of highest kernel to seed was also obtained in seeds of Raigarh (60.11%). Kernels from Chhatarpur population recorded the highest oil content (24.20%), which was equal to that in Sihore (23.75%) and Katni (21.75%). We recorded highly significant variation in azadirachtin content in seed samples, in vitro shoot samples and callus cultures. Maximum azadirachtin was present in the seeds of Gwalior (0.882%) which was on par with azadirachtin content in seeds of Chhatarpur, Katni, Sihore, Shahdol and Chhindwara. Maximum azadirachtin in the in vitro shoots was present in the cultures of Gwalior (0.218%) which was statistically on par with Sihore (0.176%). Similarly, in callus cultures of nodal segments maximum azadirachtin was recorded in the Gwalior population (0.033%) which was statistically on par with Sihore (0.028%). The trees of Gwalior and Sihore populations can be produced in large numbers through vegetative propagation to obtain seeds with higher azadirachtin content.  相似文献

Purpose

The carbon sink function of peatlands is primarily driven by a higher production than decomposition of the litter Sphagnum mosses. The observed increase of vascular plants in peatlands could alter the decomposition rate and the carbon (C) cycle through a litter mixing effect, which is still poorly studied. Here, we examine the litter mixing effect of a peat moss (Sphagnum fallax) and two vascular plants (Pinus uncinata and Eriophorum vaginatum) in the field and laboratory-based experiment.

Materials and methods

During the laboratory incubation, mass loss, CO2 production, and dissolved organic carbon concentration were periodically monitored during 51 days. The collected data were then processed in a C dynamics model. The calculated enzymatic activity was correlated to the measured β-glucosidase activity in the litter. In the field experiment, mass loss and CO2 production from litter bags were annually measured for 3 years.

Results and discussion

Both laboratory and field experiments clearly show that the litter mixture, i.e., Sphagnum-Pinus-Eriophorum, had a synergistic effect on decomposition by enhancing the mass loss. Such enhanced mass loss increased the water extractable C and CO2 production in the litter mixture during the laboratory experiment. The synergistic effect was mainly controlled by the Sphagnum-Eriophorum mixture that significantly enhanced both mass loss and CO2 production. Although the β-glucosidase activity is often considered as a major driver of decomposition, mixing the litters did not cause any increase of the activity of this exo-enzyme in the laboratory experiment suggesting that other enzymes can play an important role in the observed effect.

Conclusions

Mixing litters of graminoid and Sphagnum species led to a synergistic effect on litter decomposition. In a context of vegetation dynamics in response to environmental change, such a mixing effect could alter the C dynamics at a larger scale. Identifying the key mechanisms responsible for the synergistic effect on litter decomposition, with a specific focus on the enzymatic activities, is crucial to better predict the capacity of peatlands to act as C sinks.

Tropospheric ozone (O3) is considered harmful to agriculture production and soil community. Wheat cultivars HD 2987 (O3 sensitive) and Kharchiya 65 (O3 tolerant) were grown under ambient and elevated (ambient + 30 ppb) levels of O3 (EO) using open top chambers, and microbial biomass and enzymatic activities were investigated in soil rhizosphere. Elevated O3 declined soil enzymatic activities related to carbon cycling viz. β-glucosidase, cellobiohydrolase, cellulase and amylase more in sensitive cultivar compared to tolerant. Enzymatic activities linked to nitrogen cycling like N-acetyl-glucosaminidase and urease decreased while protease and glycine aminopeptidase increased. Microbial biomass carbon and nitrogen declined more in sensitive cultivar by 11.1 and 21.2%, respectively. Root biomass reduced in wheat cultivars with an increase of their phenolics contents by 34.3 and 10.2% in HD 2987 and Kharchiya 65, respectively at 60 days after germination. Non-significant changes were observed in soil organic carbon and total nitrogen in both the cultivars. Redundancy analysis suggested that soil enzymatic activities were predominantly affected by O3 induced changes in microbial biomass carbon and root biomass. Study also showed that rhizosphere of sensitive cultivar HD 2987 was affected more under EO as compared to tolerant Kharchiya 65.  相似文献

A new Integrated Pest Management program based on the combination of synthetic pesticide with a GRAS(generally recognized as safe)-classified sanitizer for the control of Botrytis cinerea in field conditions was described. The aim behind this research was to determine whether the use of this mixture would enhance the efficiency of pesticides while decreasing the recommended dose. Naturally infected tomato plants, grown in the greenhouse, were treated with two commonly used fungicides SWITCH(Syngenta, Switzerland) and SIGNUM(BASF, Germany) each alone or combined with a commercially available organic sanitizer PERACLEAN?5(Evonik Industries, Germany). A total of 27 treatments were tested consisting of three concentrations of synthetic fungicide(×1, ×1/2 and ×1/4 of the recommended dose) either applied separately or combined with three concentrations of the tested sanitizer(0.5, 1 and 1.5%). The control efficacy achieved by the fungicides applied alone ranged between 0 and 66.7% while all fungicide-sanitizer mixtures resulted in up to 70% control of grey mould. The treatment that provides the maximum control of B. cinerea was the result mixture of ×1/4 of the recommended concentration of SWITCH(15 g L~(–1)) with 0.5% of PERACLEAN~?5. This combination suppressed 85% of grey mold infections while decreasing the usually used amount of this pesticide by 75%, reducing therefore the well known negative impacts of chemical pesticides on environment and consumers health.  相似文献

  • 1. Surveys were carried out between December 2004 and March 2006 on board tourism schooners to detect possible impacts due to tourist activities on estuarine dolphins (Sotalia guianensis) in the Estuarine Complex of Cananéia.
  • 2. The estuarine dolphins exhibited different reactions according to the different periods of time that the boats remained close to them: the longer the period, the estuarine dolphins reacted less negatively, and for periods between 2 and 30 min, more positive reactions were observed.
  • 3. Estuarine dolphin reactions also varied according to the different procedures followed by the skippers. The approach methods considered ‘correct’ resulted in a positive reaction from 97% of the animals encountered and 100% of occasions where there were no apparent reactions.
  • 4. Direct approaches to within less than 50 m were responsible for a large number of negative reactions from the estuarine dolphins. Procedures classified as ‘chasing’ and ‘direct approach with no intention of stopping’ had a 100% negative reaction. If the skippers used a correct approach, estuarine dolphins reacted positively or did not show any reaction.
