Insulin‐like growth factor‐1 (IGF1) plays an important role in muscle development in chickens. In this study, an F2 chicken population of 362 individuals, obtained from an intercross between high breast muscle yield line males and low breast muscle yield (LB) line females, was constructed for investigating the associations between IGF1 gene and breast muscle yields. The IGF1 sequence was investigated in the grandparents. There were no differences in the exon sequences. However, sequence analysis of the IGF1 promoter revealed a known single nucleotide polymorphism (g.570C > A) in LB line grandparents. PCR – restriction fragment length polymorphism was used for screening the F2 population, which was evaluated for body weight (BW), carcass weight (CW), breast muscle weight (BMW), and breast fillet weight (BFW). Significant associations with the polymorphism were detected for BMW, BFW, BMW% and BFW%, although there were no associations between the polymorphism and BW or CW. The allelic effect on BMW, BFW, BMW% and BFW% acted in additive and dominance modes. We confirmed that the g.570C > A polymorphism is significantly associated with breast muscle yields in the F2 population. Therefore, this polymorphism in the IGF1 gene may help improve breast muscle yields by marker‐assisted selection. 相似文献

The leptin receptor (LEPR) gene is considered a candidate gene for fatness traits. It is located on SSC 6 in a region in which quantitative trait loci (QTLs) for backfat thickness (BF), fat area ratios, and serum leptin concentration (LEPC) have previously been detected in a Duroc purebred population. The objectives of the present study were to identify porcine LEPR polymorphisms and examine the effects of LEPR polymorphisms on fatness traits in this same population. The Duroc pigs (226 to 953 pigs) were evaluated for BF, fat area ratios using image analysis, and LEPC. A total of seven single nucleotide polymorphisms (SNPs) in the full‐length LEPR coding region were identified in pigs from the base population. Four non‐synonymous SNPs of the LEPR gene and 15 microsatellite markers on SSC 6 were then genotyped in all pigs. During candidate gene analysis, we detected significant effects of the non‐synonymous SNP c.2002C>T in exon 14 on all traits. In fine mapping analysis, significant QTLs for BF, fat area ratios, and LEPC were detected near the LEPR gene in the same region. These results indicated that the c.2002C>T SNP of LEPR has a strong effect on BF, fat area ratios and LEPC. 相似文献

Analysis of L-type amino acid transport expression of hepatocellular carcinoma cells
(HCCs) of the dog was performed. The leucine transport activity of canine HCCs was 0.628 ±
0.018 nmol/mg protein/min. The inhibitor of LAT 2-aminobicyclo[2.2.1]heptane-2-carboxylic
acid (BCH) reduced 90% of the activity at 1 mM. The deduced amino acid sequences of canine
LAT2, LAT3 and LAT4 were well conserved in mammalians, exhibiting 89, 88 and 77% homology,
respectively. RT-PCR revealed distinct LAT1 expression compared with normal hepatocytes.
Western blotting analysis confirmed the potent LAT1 expression in canine HCCs but not
hepatocytes, and real-time RT-PCR analysis indicated that canine HCCs possessed 28 times
higher LAT1 expression than hepatocytes. These results indicated that the leucine
transport activity of canine HCCs was due to LAT1. 相似文献

Fifty-one Salmonella enterica serovar 4,[5],12:i:-
(S. 4, [5],12:i:-) isolates (14 human strains, 34 animal strains and 3
river water strains) which are assumed to be monophasic variants of S.
Typhimurium were analyzed using pulsed-field gel electrophoresis (PFGE) and multiple-locus
variable-number tandem repeat analysis (MLVA) in order to investigate their genetic
diversities and relationships. PFGE, MLVA and combination of them identified 28, 27 and 34
profiles (Simpson’s diversity indices [DI]=0.94, 0.96 and 0.97), respectively. No
correlations were detected between MLVA clustering and PFGE clustering or phage typing.
These results suggested that S. 4,[5],12:i:- originated from multiple
S. Typhimurium ancestors. Two cattle and one pig isolates showing
identical phage types as well as PFGE and MLVA profiles to human isolates
S. 4,[5],12:i:- suggested the existence of the links between human

ABSTRACT: Nucleoplasmin, first isolated from Xenopus laevis eggs, promotes nucleosome assembly. Hereby, we have purified a nucleoplasmin-like protein from carp ( Cyprinus carpio ) eggs using ion exchange and subsequent gel filtration columns. The protein was recognized by a polyclonal antiserum against Xenopus laevis nucleoplasmin and had an amino acid composition similar to other member of the nucleoplasmin family proteins. Partial amino acid sequences from the cyanogen bromide (CNBr)-cleaved fragments showed high homology with Xenopus nucleoplasmin. The protein was also found to form an oligomeric complex and to be phosphorylated. Moreover, this protein promoted sperm nuclear decondensation as well as that of nucleoplasmin from Xenopus laevis eggs. These results suggest that the fish protein isolated here is a member of nucleoplasmin family. 相似文献

It has been demonstrated that L-pipecolic acid (L-PA), a major metabolic intermediate of L-lysine (L-Lys) in the brain, is involved in the functioning of the γ-aminobutyric acid-ergic system. Previous study has shown that intracerebroventricular injection of L-PA suppressed feeding and induced sleep-like behavior in neonatal chicks. The present study examines whether the action of L-PA was induced by gavage in both chicks and mice. Oral administration of L-PA significantly inhibited food intake at 2 h after treatment in neonatal chicks, although no significant effect of L-Lys was detected. In mice, oral L-PA suppressed food intake compared to the control after 2 h of treatment. It was concluded that L-PA was effective for suppression of food intake after oral administration in both avian and mammalian species. 相似文献
