Accurate detection of oestrus is important for artificial insemination. The aim of this study was to identify oestrous‐specific bovine cervical mucus proteins that could be used to determine the optimal time for artificial insemination. Non‐oestrous and controlled internal drug release (CIDR)‐induced oestrous‐stage mucus proteins were purified and subjected to surface‐enhanced laser desorption/ionization time‐of‐flight mass spectrometry, sodium dodecyl sulphate polyacrylamide gel electrophoresis and MALDI‐TOF/TOF. Among differentially expressed proteins, lactoferrin (LF) and glutamate receptor‐interacting protein 1 (GRIP1) showed a twofold increase during the CIDR‐induced oestrous stage compared to the levels in non‐oestrous stage in bovine cervical mucus. The RT‐PCR, Western blotting and immunohistochemistry results showed that LF and GRIP1 expression was significantly increased during the oestrous stage in the uterus. This study demonstrated that bovine LF and GRIP1 exist during the oestrous stage, but not during the non‐oestrous stage, suggesting that cervical mucus LF and GRIP1 are useful oestrous detection markers in cattle. 相似文献

The relationship of Helicobacter felis, an organism that is observed in the stomachs of dogs, to gastric disease in dogs is unclear. The objective of this study was to determine if Helicobacter felis infection alters gastric morphology and gastric secretory function in dogs. Five specific-pathogen-free (SPF), Helicobacter-free Beagle dogs were examined before and for 26 weeks after inoculation with H. felis (ATCC 49179). Three SPF uninfected dogs served as controls. All five dogs became colonized by H. felis as determined by urease activity, histopathology, polymerase chain reaction, and transmission electron microscopic examination of serial gastric biopsies. The degree of colonization ranged from < 1 organism/400 x field to > 10 organisms/400 x field. The fundus, body, and cardia were most heavily colonized. Evaluation of gastric biopsies showed mild gastric inflammation and lymphoid follicles in both infected and uninfected dogs. There was no correlation between the number of organisms observed and the degree of gastric inflammation or number of lymphoid follicles. The gastric secretory axis, assessed by fasting and meal-stimulated plasma gastrin, mucosal gastrin and somatostatin immunoreactivity, fasting gastric pH, and pentagastrin-stimulated gastric acid secretion, was similar in both infected and uninfected dogs. Fasting gastric pH was not a reliable indicator of gastric secretory function. These findings suggest that H. felis may not be a gastric pathogen in dogs. However, the density of colonization and limited duration of infection should be considered when interpreting these findings. 相似文献

1. Climate change can affect the survival, colonization and establishment of non‐native species. Many non‐native species common in Europe are spreading northwards as seawater temperatures increase. The similarity of climatic conditions between source and recipient areas is assumed to influence the establishment of such species, however, in a changing climate those conditions are difficult to predict. 2. A risk assessment methodology has been applied to identify non‐native species with proven invasive qualities that have not yet arrived in north‐west Europe, but which could become problematic in the future. Those species with the highest potential to become established or be problematic have been taken forward, as well as some that may be economically beneficial, for species distribution modelling to determine future potential habitat distributions under projected climate change. 3. In the past, species distribution models have usually made use of low resolution global environmental datasets. Here, to increase the local resolution of the distribution models, downscaled shelf seas climate change model outputs for north‐west Europe were nested within global outputs. In this way the distribution model could be trained using the global species presence data including the species' native locations, and then projected using more comprehensive shelf seas data to understand habitat suitability in a potential recipient area. 4. Distribution modelling found that habitat suitability will generally increase further north for those species with the highest potential to become established or problematic. Most of these are known to be species with potentially serious consequences for conservation. With caution, a small number of species may present an opportunity for the fishing industry or aquaculture. The ability to provide potential future distributions could be valuable in prioritizing species for monitoring or eradication programmes, increasing the chances of identifying problem species early.
