This study evaluated the performance of an immunochromatographic test (ICT) for the diagnosis of canine brucellosis caused by Brucella canis, comparing its results with that of the rapid slide agglutination test with and without the use of 2‐mercaptoethanol and the agar gel immunodiffusion test (AGID). The microbiological culture, PCR and clinical examination were used as reference. According to the results obtained in clinical examination, blood culture, culture of semen and vaginal swab and PCR in blood, semen and vaginal swab, a total of 102 dogs were divided into three groups: B. canis‐infected dogs (Group 1), B. canis‐non‐infected dogs (Group 2) and dogs with suspected brucellosis (Group 3). The diagnostic sensitivity of RSAT, 2ME‐RSAT, AGID and ICT in Group 1 was, respectively, 75%, 37.5%, 27.8% and 89.58%. The diagnostic specificity of RSAT, 2ME‐RSAT, AGID and ICT in Group 2 was, respectively, 91%, 100%, 100%, and 100%. In dogs with suspected brucellosis, 9.67% were RSAT positive, none was positive by 2ME‐RSAT, 3.22% were AGID positive and 6.45% were ICT positive. The main drawback concerning canine brucellosis diagnosis is the lack of a highly sensitive serological assay to be used as a screening test to the rapid identification of infected animals. The ICT showed a high diagnostic specificity and a diagnostic sensitivity value greater than that observed in the RSAT, 2ME‐RSAT and AGID. However, 10.41% of infected dogs had negative results by ICT. These dogs were positive by microbiological culture and/or PCR, indicating active infection and consequently a higher potential of spreading Brucella. Although rapid and simple to perform, the ICT lacked sensitivity to be used as a screening test.  相似文献

A 10‐year‐old, castrated male, German Shepherd mixed‐breed dog was presented to Kansas State University Veterinary Health Center for evaluation of a subconjunctival swelling in the ventral fornix of the left orbit. The owner elected to pursue excision of the mass 2 years after initial consultation following a sudden change in the size and color of the lesion. An excisional biopsy was performed, and the mass along with its associated capsule were submitted to the Comparative Ocular Pathology Laboratory of Wisconsin for histopathologic evaluation, which confirmed the diagnosis of a hibernoma. Fourteen months following excision, the patient showed no evidence of tumor regrowth.  相似文献

Global Oscillation Network Group data reveal that the internal structure of the sun can be well represented by a calibrated standard model. However, immediately beneath the convection zone and at the edge of the energy-generating core, the sound-speed variation is somewhat smoother in the sun than it is in the model. This could be a consequence of chemical inhomogeneity that is too severe in the model, perhaps owing to inaccurate modeling of gravitational settling or to neglected macroscopic motion that may be present in the sun. Accurate knowledge of the sun's structure enables inferences to be made about the physics that controls the sun; for example, through the opacity, the equation of state, or wave motion. Those inferences can then be used elsewhere in astrophysics.  相似文献

Atmospheric general circulation models used for climate simulation and weather forecasting require the fluxes of radiation, heat, water vapor, and momentum across the land-atmosphere interface to be specified. These fluxes are calculated by submodels called land surface parameterizations. Over the last 20 years, these parameterizations have evolved from simple, unrealistic schemes into credible representations of the global soil-vegetation-atmosphere transfer system as advances in plant physiological and hydrological research, advances in satellite data interpretation, and the results of large-scale field experiments have been exploited. Some modern schemes incorporate biogeochemical and ecological knowledge and, when coupled with advanced climate and ocean models, will be capable of modeling the biological and physical responses of the Earth system to global change, for example, increasing atmospheric carbon dioxide.  相似文献   

Detection of potato viruses S and X in dormant potato tubers by the latex agglutination test

A simple procedure for detection of potato viruses S and X from dormant tubers by a latex agglutination test was developed. Eyes were pricked with a toothpick after having been covered with 50μ1 of buffer. Approximately 20μ1 of buffer then were drawn into a capillary pipette containing 10μ1 of antibody-sensitized latex and a latex agglutination test was performed. Both viruses were detected in all infected tubers tested. For potato virus S, the mean agglutination value was shown to vary significantly among cultivars. Tests on one eye were sufficient for detection with some cultivars, but tests on three eyes were necessary for detection with other cultivars. Eye location on tubers did not appear to influence test results, but better results were obtained when eyes were tested than when other tuber locations were tested.  相似文献

Eva is a new potato variety being released by the New York and Pennsylvania Experiment Stations. Eva is notable for its bright clear skin and attractive round to oval shape. It produces a chip color comparable to Monona from 9 C storage and has specific gravity 014 less than that of Atlantic. It has exceptionally long tuber dormancy and has extreme resistance to PVX, PVY, and race RO1 of the golden nematode. Scab resistance is like that of Monona. Quantitative differences among strains ofCorynebacterium sepedonicum (Spieck. and Kotth.) Skapt. and Burkh. (=Clavibacter michiganense subsp.sepedonicum) were distinguished in eggplant (Solanum melongena L. ‘Black Beauty’) bioassays with a simple leaf axil inoculation technique. Latent period of infection (time from inoculation to first appearance of symptoms) varied inversely with inoculum dose. Latent period of infection varied significantly among strains ofC. sepedonicum. Symptoms developed more rapidly in plants grown under controlled conditions in a growth chamber than those grown in a greenhouse. Plants treated with inoculum prepared from broth cultures exhibited symptoms more rapidly than those treated with inoculum grown on agar.  相似文献
