Centaurea solstitialis, commonly known as yellow starthistle, is an invasive plant listed as a noxious weed in the western areas of North America and is the target of classical biological control, which involves release of herbivores known to be specific to this plant. These insects often choose their host plant on the basis of the volatile organic compounds (VOCs) emitted. Accordingly, volatile analysis of host plants can provide insight into VOCs that may attract and/or repel the insect. To this end, solid-phase microextraction (SPME) and a customized collection bag were utilized to perform in situ volatile collection on intact and mechanically damaged leaves of Centaurea solstitialis, Centaurea cyanus, and Centaurea cineraria. Volatile identification was performed by GC-MS, and the VOC differences were determined. The plants C. solstitialis and C. cyanus have been reported to attract the weevil, Ceratapion basicorne, a candidate for biological control, whereas C. cineraria does not attract the weevil. Major VOCs unique to C. cineraria include the sesquiterpenes cyclosativene, alpha-ylangene, and trans-alpha-bergamotene. The compound trans-beta-farnesene was unique to C. solstitialis and C. cyanus.  相似文献

Samples (n = 620) of homogenized red grape berries were analyzed using a visible and near-infrared (NIR) spectrophotometer (400-2500 nm) in reflectance. The spectra and the analytical data were used to develop partial least-squares calibrations to predict dry matter (DM) content and condensed tannins (CT) concentrations. The coefficient of determination in cross-validation and the standard error of cross-validation were 0.92 and 0.83% w/w for DM and 0.86 and 0.46 mg/g epicatechin equivalents for CT, respectively. The standard error in prediction was 1.34% w/w for DM and 0.89 mg/g epicatechin equivalents for CT, respectively. By implementing a NIR spectroscopy method to measure DM and CT in red grape homogenates, we have developed an approach that is suited to large-scale compositional analysis in commercial wine production facilities, as it enables the analysis of large numbers of samples needed to stream batches of fruit. From an economical point of view, the calibration models could be achieved with relatively small data sets. Thus, NIR offers a suitable and efficient tool for the simultaneous measurement of DM and CT in addition to other important parameters in red grape homogenates such as total anthocyanins, total soluble solids, and pH, with minimal sample preparation and low cost.  相似文献

Carrot cell walls have been shown to contain significant quantities of esterified p-hydroxybenzoic acid, which is presumed to be esterified to cell wall polymers. The purpose of this study was to investigate the distribution of p-hydroxybenzoic acid and related phenolics among carrot cell wall polysaccharides. Cell wall material was prepared from fresh carrot root tissues and extracted sequentially with water, imidazole, cyclohexane- trans-1,2-diamine- N, N, N', N'-tetraacetate, Na 2CO 3, and KOH (0.5, 1, and 4 M) to leave a cellulose-rich residue. The fractions were analyzed for their carbohydrate and phenolic acid components. Selected soluble fractions were subfractionated further by graded precipitation in ethanol. The majority of the polymer fractions comprised pectic polysaccharides, with varying quantities of neutral sugars (arabinose and galactose). Hemicellulosic polymers were generally found only in the strong alkali extracts (4 M KOH). p-OH-benzoic acid was the predominant phenolic ester and was associated with most fractions analyzed; p-OH-benzaldehyde was also detected in the fractions at much lower levels. Principal components analysis of the chemical data indicated that the p-OH-benzoic acid was associated predominantly with the branched pectic polysaccharides, in contrast to the p-OH-benzaldehyde. The possible roles and functional properties of these phenolics are discussed.  相似文献

Analysis of wheat bran and spent grain shows that concentrations of ferulate and diferulates offer considerable scope to modify the cross-linking of feruloylated polysaccharides and hence the mechanical properties of these residues. In solution ferulic acid (FA) can be readily polymerized by horseradish peroxidase, but when esterified to a polysaccharide, the profile of diferulates becomes restricted. This situation also exists in muro and suggests structural constraints may limit the availability of FA for cross-linking. At relatively low polysaccharide concentration, (approximately 3%), steric restrictions were apparent in gels prepared using isolated polysaccharides. Mechanical properties such as swelling also appear to be fixed at these relatively low polysaccharide concentrations. This limits the potential to modify mechanical properties in muro through oxidoreductase activity. To modify mechanical properties such treatments will need to focus on increasing the "flexibility" of the cell wall matrix and the accessibility of enzymes to the cell wall matrix.  相似文献

The yield and flux of nitrous oxide (N2O) emitted from continuous cereals (with and without urea), legumes/cereal in rotation and cereal/legume in rotation all with or without organic manure was monitored from January 2004 to February 2005. All treatments except continuous cereals had phosphate added. The cereal grown July–October in 2003 and 2004 was pearl millet ( Pennisetum glaucum) and the legume was a bean ( Phaseolus vulgaris ). The 10 m × 10 m plots were established in a semi-arid climate in Mali. The addition of organic manure and both inorganic fertilizers increased yield and N2O emissions. Continuous cereals treated with both organic manure and urea emitted significantly less N2O (882 g N/ha per year) than plots receiving no organic manure(1535 g N/ha per year). Growing N-fixing crops in rotation did not significantly increase N2O emissions. This study supports the new practice of growing cereal and legumes in rotation as an environmentally sustainable system in semi-arid Mali.  相似文献

A gnamma or weathering pit is a small-sized cavity sculpted into solid rock which refills with rainwater. This study describes the main hydrological processes in gnammas and the underlying chemical interactions with the environment. Four gnammas developed in a granite tor from central Spain were chosen for monitoring and studying the dynamics of the gnamma–water system. Parameters monitored were air and water temperature, pH, ORP (redox potential), and conductivity. These measurements describe the system dynamics, without the need for special sampling or other potential interferences under low water volumes. Additionally, chemical analyses were carried out to contrast the chemical properties with the chemical composition. As a consequence of water volumes less than 100 L, daily fluctuations tended to cause the largest amplitude changes. Photosynthesis and respiration/putrefaction processes in the gnamma are responsible for daily pH fluctuations up to 4 units, with pH values often greater than 9. ORP generally alternates from positive to negative values as the solution oscillates diurnally between basic and acidic conditions. Conductivity values ∼ 10 μS/cm occur most of the year, with only minor daily oscillations. During dry periods, conductivity increases significantly with maximum values > 400 μS/cm. These high conductivity values reflect atmospheric inputs by summer rainstorms after prolonged dry periods. Therefore, conductivity does not relate directly with chemical species dissolved from the host rock. Silica was the main weathering product of granite dissolution during the year, with highest concentrations (> 200 ppm) in the rainy season. Low Si values measured during the dry season are in accordance with observed precipitation of amorphous silica within the gnamma. Seasonal variations in concentration are also present in Al and Fe as a response to ORP variations and evaporative dynamics, causing mineral dissolution and new mineral precipitation during the year. Relations among the measured parameters are influenced by several factors, including biological processes, temperature effects and water level variations. Factors such as large pH oscillations, basic pH values, acids from biological activity, and alkali cations all increase silicate solubility, thus favoring granite dissolution. Overflow of water during the winter, when the water is chemically enriched, is an effective mechanism to remove weathering products.  相似文献

Urbanization of watersheds previously managed for agricultural uses results in hydrologic changes associated with increased flooding and erosion. Few studies have been conducted to quantify these effects under controlled conditions and standard rainfall simulation methodologies have not been previously established. In this study, a laboratory rainfall simulation procedure was developed and utilized to evaluate hydrologic and sheet erosional responses to various configurations of impervious surface cover at the small scale. Runoff and sediment losses from a sloped (5%) cascade of soil boxes having 50% impervious cover located at the top of the slope or at the bottom of the slope, or having 0% impervious cover were measured. Results indicate that the 50% upslope impervious treatment generated sediment at 3–5 times the rate of the 50% downslope impervious treatment. Upslope impervious cover resulted in initially lower water runoff rates than channel development, but this effect narrowed or reversed with continued rainfall. These results suggest that upslope impervious surfaces may represent a larger total on-site erosion risk than equivalent impervious surfaces located at lower positions along the slope, especially under high antecedent soil moisture and/or high intensity rainfall.  相似文献

The soil fauna is often a neglected group in many large-scale studies of farmland biodiversity due to difficulties in extracting organisms efficiently from the soil. This study assesses the relative efficiency of the simple and cheap sampling method of handsorting against Berlese–Tullgren funnel and Winkler apparatus extraction. Soil cores were taken from grassy arable field margins and wheat fields in Cambridgeshire, UK, and the efficiencies of the three methods in assessing the abundances and species densities of soil macroinvertebrates were compared. Handsorting in most cases was as efficient at extracting the majority of the soil macrofauna as the Berlese–Tullgren funnel and Winkler bag methods, although it underestimated the species densities of the woodlice and adult beetles. There were no obvious biases among the three methods for the particular vegetation types sampled and no significant differences in the size distributions of the earthworms and beetles. Proportionally fewer damaged earthworms were recorded in larger (25 × 25 cm) soil cores when compared with smaller ones (15 × 15 cm). Handsorting has many benefits, including targeted extraction, minimum disturbance to the habitat and shorter sampling periods and may be the most appropriate method for studies of farmland biodiversity when a high number of soil cores need to be sampled.  相似文献

We developed a PCR assay for the rapid and sensitive detection of virulent Streptococcus suis type 2 and highly virulent S. suis type 1 in tonsillar specimens from pigs. The PCR primers were based on the sequence of the gene encoding the EF-protein of virulent S. suis type 2 strains (MRP+EF+) and highly virulent S. suis type 1 strains (MRP(s)EF+) and of the EF protein of weakly virulent S. suis type 2 strains (MRP+EF). The latter strains give rise to larger PCR products than the virulent strains of S. suis type 1 and 2. A positive control template was included in the assay to identify false negative results. The PCR was evaluated using tonsillar specimens from herds known (or suspected) to be infected and herds without an S. suis history. The results obtained with the PCR assay were compared with the results obtained with a newly developed bacteriological examination. In this bacteriological examination we were able to identify the EF-positive strains directly in the tonsillar specimens. From the 99 tonsils examined, 48 were positive in the PCR and 51 negative. All specimens from which EF-positive S. suis strains were isolated were also positive in the PCR assay. Three samples were positive in the PCR, but negative by bacteriological examination. The results demonstrated that the PCR is a highly specific and sensitive diagnostic tool for the detection of pigs carrying virulent strains of S. suis type 2 and highly virulent strains of type 1. Application of the assay may contribute to the control of S. suis infections.  相似文献

Hedgerows have the potential to influence ecosystem function in livestock‐grazed pasture. Despite this, they are often ignored when quantifying farmland ecosystem service delivery. In this study, we assess the contribution of hedgerows to the ecosystem function of carbon (C) storage, with a particular emphasis on soil organic carbon (SOC). We measured SOC stock (kg C m?2), on an equivalent soil mass basis, at 0–0.15 m depth in pasture adjacent to 38 hedgerows (biotic) and 16 stone walls or fences (abiotic controls) across ten farms in the county of Conwy, Wales, UK. Pasture SOC stock (~7 kg C m?2) was similar adjacent to biotic and abiotic field boundaries, positively associated with soil moisture and negatively with soil bulk density (BD). For biotic boundaries, two further variables were significantly associated with SOC stock, distance from hedgerow (decrease in SOC stock) and slope orientation (upslope SOC stock greater than downslope). For pasture adjacent to hedgerows, a model combining the aforementioned variables (BD, soil moisture, distance from hedgerow, slope orientation) explained 78% of variation in SOC stock. This study demonstrates that whilst hedgerows do have subtle positive effects on SOC stock in adjacent pasture, SOC storage adjacent to field boundaries is influenced more by soil moisture content and BD than field boundary type.  相似文献
