To evaluate the dielectric anisotropy caused by wood structure at a millimeter wave frequency of 100 GHz, the dielectric parameters for flat-sawn specimens of nine wood species at 0 and 11 % moisture content (MC) were measured using a free space method devised for reducing the multiple reflections under an electric field of millimeter waves parallel to longitudinal and tangential directions of wood, and those in radial direction were estimated using a conventional approximation theory. The dielectric parameters in the tangential and radial directions were almost identical and constantly smaller than those in the longitudinal direction. All the dielectric parameters increased with wood density and were larger at 11 than 0 % MC. The dielectric parameters in the longitudinal and transverse directions and the dielectric anisotropy between them were well fitted to the regression lines based on a dielectric mixture model composed of pores and dielectric isotropic wood substance, and a parallel capacitor and Lichtenecker’s exponential formulas were employed to represent the dielectric parameters of the mixture in the longitudinal and transverse directions, respectively. It was concluded that the dielectric anisotropy at 100 GHz is caused by the pore alignment and that the dielectric parameters are almost unaffected by anatomical structures, such as the rays. It was also confirmed that the free space method was effective for the measurement of the dielectric parameters for the flat-sawn specimens.  相似文献

For the application of millimeter wave (MMW) technique to nondestructive evaluation of wood, the effect of annual rings on the behavior of a 100 GHz MMW transmitted through wood was examined. The complex amplitude was measured for 2 mm thick flat- (LT) and quarter-sawn (LR) specimens of several species with different annual ring structures at 11 % moisture content, of which the density distribution was measured using X-ray radiography. For the LT specimens of all species and the LR specimens with small density fluctuation or with earlywood width smaller than the wavelength of the MMW (=3 mm), the amplitude and phase of the transmitted wave were similar to those of the wave without a specimen. For the LR specimen with large density fluctuation and with earlywood width close to or larger than the wavelength, the amplitude and phase were different from those of the wave without a specimen. All the measured complex amplitudes were well expressed using a diffraction model. It was concluded that the MMW is deformed by the density distribution, and then its components with periods shorter than the wavelength by diffraction are attenuated.  相似文献

Members of the Toll-like receptor (TLR) and interleukin-1 receptor (IL-1R) superfamily share an intracytoplasmic Toll-IL-1 receptor (TIR) domain, which mediates recruitment of the interleukin-1 receptor-associated kinase (IRAK) complex via TIR-containing adapter molecules. We describe three unrelated children with inherited IRAK-4 deficiency. Their blood and fibroblast cells did not activate nuclear factor kappaB and mitogen-activated protein kinase (MAPK) and failed to induce downstream cytokines in response to any of the known ligands of TIR-bearing receptors. The otherwise healthy children developed infections caused by pyogenic bacteria. These findings suggest that, in humans, the TIR-IRAK signaling pathway is crucial for protective immunity against specific bacteria but is redundant against most other microorganisms.  相似文献

To clarify phylogenetic relationships and genetic diversity among Bartonella washoensis strains obtained from squirrels, multi-locus sequence analysis (MLSA) with the 16S rRNA, ftsZ, gltA, groEL, ribC, and rpoB genes was applied for 20 strains of B. washoensis isolated from five genera of squirrels (Tamias, Tamiasciurus, Glaucomys, Sciurus, and Spermophilus) within the family Sciuridae. Sequence similarities in the concatenated sequences of B. washoensis strains from squirrels of different genera ranged from 94.7% (Sciurus vs. Spermophilus) to 98.4% (Tamiasciurus vs. Glaucomys). Phylogenetic trees based on the concatenated sequences revealed that B. washoensis strains formed five distinct clades and each clade correlated with the genus of squirrel from which the strains were originally obtained. The discrimination was supported by 100% bootstrap values and posterior probabilities, respectively. These results suggest that B. washoensis strains may have co-speciated with their squirrel hosts and provide new insights into the application of the MLSA to identify sources of B. washoensis infection with accuracy.  相似文献

Differences in the distribution of single nucleotide polymorphisms (SNPs) and haplotypes in the estrogen receptor α gene (ESR1) were examined in Miniature Dachshunds (n = 48), Chihuahuas (n = 20) and Toy Poodles (n = 18). Five DNA fragments located in the 40‐kb region at the 3′ end of ESR1 were amplified by polymerase chain reaction and were directly sequenced. We compared allele, genotype and estimated haplotype frequencies at each SNP in the 3′ end of ESR1 for these three breeds of small dog. The frequency of the major allele and the genotype frequency of the major allele homozygotes, were significantly higher in Toy Poodles for five SNPs (SNP #5, #14–17) than in Miniature Dachshunds, and significantly higher in Toy Poodles than Chihuahuas for three SNPs (SNP #15–17). A common haplotype block was identified in an approximately 20‐kb region encompassing four SNPs (SNPs # 14–17). The frequencies of the most abundant estimated haplotype (GTTG) and GTTG homozygotes were significantly higher in Toy Poodles than in the other two breeds. These results imply that homozygosity for the allele, genotype and haplotype distribution within the block at the 3′ end of ESR1 is greater in Toy Poodles than in Miniature Dachshunds and Chihuahuas.  相似文献

Bifidobacterial plasmids reported so far are derived from a limited number of strains and plasmids of bifidobacterial type strains isolated from humans are unknown. We found that Bifidobacterium kashiwanohense JCM 15439 (type strain) isolated from a healthy infant contained two cryptic plasmids, designated pBBKW‐1 and pBBKW‐2. We determined and analyzed the complete sequences of both plasmids. pBBKW‐1 (7716 bp) was predicted to replicate by a rolling‐circle mechanism and encode six protein‐coding genes, two of which are putative replication proteins. pBBKW‐1 seems to be a cointegrate plasmid containing two copies of the plasmid pMG1 from Bifidobacterium longum. pBBKW‐2 (2920 bp) was predicted to encode six protein‐coding genes and be a theta‐type replicating plasmid, which has been reported to be more stable than a rolling circle‐type replicating plasmid frequently found in bifidobacteria. Our finding will provide new insights into safe recombinant plasmid constructions for humans.  相似文献

We investigated the prevalence of Bartonella species in 10 rodent and one shrew species in Thailand. From February 2008 to May 2010, a total of 375 small animals were captured in 9 provinces in Thailand. Bartonella strains were isolated from 57 rodents (54 from Rattus species and 3 from Bandicota indica) and one shrew (Suncus murinus) in 7 of the 9 provinces, and identified to the species level. Sequence analysis of the citrate synthase and RNA polymerase β subunit genes identified the 58 isolates from each Bartonella-positive animal as B. tribocorum in 27 (46.6%) animals, B. rattimassiliensis in 17 (29.3%) animals, B. elizabethae in 10 (17.2%) animals and B. queenslandensis in 4 (6.9%) animals. R. norvegicus, R. rattus, and Suncus murinus carried B. elizabethae, which causes endocarditis in humans. The prevalence of Bartonella bacteremic animals by province was 42.9% of the animals collected in Phang Nga, 26.8% in Chiang Rai, 20.4% in Sa Kaeo, 16.7% in Nakhon Si Thammarat, 12.0% in Surat Thani, 9.1% in Mae Hong Son and Loei Provinces.  相似文献

Preimplantation genomic selection combined with an in vitro embryo production system is expected as a means of accelerating genetic improvement in cattle. While micromanipulation-based biopsy approaches are often used to collect embryonic cells for genetic testing, they require expensive equipment and sophisticated skills, hindering the adoption of this system. In the present study, to develop a simple method for preimplantation genomic selection using the blastomere separation (BS) technique in bovine in vitro fertilized embryos, we examined the accuracy of single nucleotide polymorphism (SNP) genotyping and optimal cryopreservation method in demi-blastocysts produced by the BS technique. We demonstrated reliable SNP genotyping using DNA derived from demi-blastocysts. We indicated a suitable equilibrium time in vitrification solution for demi-blastocysts and succeeded obtaining pregnancies by the transfer of vitrified demi-blastocysts. In conclusion, our findings suggest that the BS technique provides a simple method for preimplantation genomic selection in bovine in vitro fertilized embryos.  相似文献

A method for designing microirrigation laterals on nonuniform slopes was developed using the finite element method. Six representative nonuniform slope patterns were discussed in detail. The design principle was implemented based on the results of computer simulations. It was found that a single lateral is suitable for Pattern I while paired laterals are better for Patterns II ∼ VI in most cases. The diameter of a single lateral or paired laterals may have two solutions for a required uniformity of water application and the length may have multiple solutions. When the required average emitter discharge, required uniformity of water application, and one parameter (either length or diameter) of a single lateral or paired laterals are given, the unknown parameter, best submain position (paired laterals) and operating pressure head can be accurately designed using personal computers. The surface antigen P50 of Babesia gibsoni is an important candidate for the development of a diagnostic reagent for canine piroplasmosis. In order to establish an effective diagnostic method for practical use, the gene encoding truncated P50 (P50t) lacking a signal peptide and C-terminal hydrophobic regions were cloned and expressed in Escherichia coli as a fusion protein with glutathione S-transferase (GST). More than 90% portion of the GST-P50t was expressed as a soluble form, in contrast with GST-P50f (full-length), which was completely expressed as an insoluble form. This result indicates that removal of the hydrophobic signal peptide and C-terminus had dramatically improved its hydrophilicity. The purified GST-P50t was tested in an enzyme-linked immunosorbent assay (ELISA) for detection of antibodies to B. gibsoni in dogs. The ELISA with GST-P50t clearly differentiated between B. gibsoni-infected dog sera and uninfected dog sera. In addition, the ELISA detected no cross-reactivity with sera from dogs experimentally infected with the closely related parasites, B. canis canis, B. canis vogeli, and B. canis rossi. Field serum samples collected from dogs in Japan and China were examined for the diagnosis of B. gibsoni infection by using the ELISA. 14.5% (9/62), 5.8% (7/120), and 5.4% (2/37) of tested samples were positive for dogs from Okinawa, Yamaguchi, and Osaka prefectures, Japan, respectively. On the other hand, 4.8% (2/41) of tested samples were positive for dogs from Nanjing, China. These results suggest that the GST-P50t could be a reliable reagent for practical use in ELISA for the serodiagnosis of canine piroplasmosis caused by B. gibsoni.  相似文献
