Bovine ephemeral fever (BEF) virus vaccines, prepared from the brains of suckling mice infected with strain 525 BEF virus, were evaluated in housed cattle and in the field. The virus in lyophilised preparations was stable for 6 months at -50 degrees C. Thirty-four calves, 5 to 18 months old, were used in laboratory vaccination trials. An increase in serum neutralising antibody was detected in 13 of 14 calves initially free of serum antibody, and all 13 failed to develop clinical illness following challenge with virulent BEF virus. Vaccination resulted in no detectable serum antibody increase in 4 calves, 5 months old, with pre-existing antibody of presumed maternal origin. Seven animals, 18 months of age with serum antibody presumed due to previous BEF infection, developed increased antibody titres following vaccination. In 3 animals vaccinated but not challenged, vaccine-induced antibodies decreased to low levels over 5 months. In contrast, the antibody titres following infection with virulent virus in 2 calves were maintained over 5 months. Field trials, involving 236 animals initially free of serum antibody, were conducted on 5 properties near Mackay and 4 properties near Brisbane. Most of 164 animals were vaccinated with a single dose of lyophilised vaccine containing aluminium hydroxide adjuvant. Only 4 animals failed to develop serum antibody and no adverse reactions to vaccination were reported. Natural infection with BEF occurred in 4 herds at Mackay and clinically mild BEF occurred in 3 of 109 vaccinated and 3 of 46 control animals. On the basis of measured serum antibody titres it was assumed that 8 of 53 animals receiving full vaccine volume, 20 of 40 animals receiving half vaccine volume and 18 of 40 control animals became infected with BEF virus. Two dairy herds in Brisbane became naturally infected with virulent BEF virus 7 months after vaccination. Clinical BEF was observed in 8 of 11 control animals and in 3 of 26 animals which received 2 doses of vaccine. Two strains of BEF virus were isolated from unvaccinated animals that developed clinically mild BEF in the field. These strains either failed to infect, or produced subclinical or very mild BEF, when inoculated intravenously into susceptible calves. The anitbody response to natural infection with apparently mild viruses was short-lived, similar to that produced by vaccination.  相似文献

The isolation and characterization of 71 strains of slowly cytopathic herpesviruses (EHV2) from horses in Queensland is described. Specimens from aged horses at slaughter yielded 7 isolates from leucocytes and 4 from tonsils, but EHV2 strains could not be detected in salivary glands, spleen, kidney or intestinal wall. A further EHV2 strain (EK550) was isolated from cell cultures prepared from the kidney of an adult horse. Repeated sampling yielded 39 nasal, ocular and genital EHV2 strains from a group of 16 mares and their young foals, and 20 strains from a group of 6 yearlings.
Laboratory examination of these EHV2 strains demonstrated variations in plaque size and growth in non-host cell cultures, and at least two serological groups were detected. EHV2 infection of foals transplacentally was not detected, but neonatal infection of foals from chronically infected mares appeared probable.  相似文献

Summary Uncooked white rice and cooked white rice were tested as vehicles for the V4 strain of oral Newcastle disease vaccine. The results of feeding experiments were evaluated by the measurement of haemagglutination inhibition antibodies against Newcastle disease virus. Little of the virus applied to uncooked white rice could be recovered, even immediately after mixing, whereas when the virus was applied to cooked white rice most of it could be recovered. In 4 separate experiments, chickens failed to respond serologically to vaccine supplied on uncooked white rice. In all of 4 experiments with cooked white rice, there were serological responses in vaccinated chickens, from 45% to 100% of the chickens developing titres sufficiently high to indicate protection against challenge with virulent virus. Development of haemagglutination inhibition antibodies in some control chickens indicated the ability of the vaccine virus for lateral spread or persistence in the environment.
Vacunacion Oral De Pollos Con La Cepa V4 Del Virus De La Enfermedad De Newcastle Utilizando Como Vehiculo Arroz Blanco Cocinado Y No Cocinado
Resumen Se utilizo arroz blanco sin cocinar y cocinado como vehículo para la vacuna oral con la cepa V4 de la enfermedad de Newcastle. Los resultados de los experimentos fueron evaluados mediante la determinación de los anticuerpos de inhibición-hemaglutinación contra el virus de la enfermedad de Newcastle. Poca cantidad del virus de la enfermedad de Newcastle pudo recuperarse en el arroz crudo, incluso inmediatamente despues de revolverlo, mientras que la mayoría del virus pudo recuperarse del arroz cocinado. En 4 experimentos separados, los pollos no respondieron serológicamente a la vacuna aplicada en arroz blanco crudo. En los 4 experimentos con arroz cocinado, se detectaron respuestas serológicas en los pollos vacunados; 45% a 100% de los pollos desarrollaron títulos suficientes como para resistir la descarga de virus virulento. El desarrollo de anticuerpos de inhibición-hemaglutinación en algunos de los pollos controles indicó la capacidad del virus vacunal para dispersarse lateralmente y persistir en el ambiente.

Vaccination Par Voie Orale De Poulets Avec La Souche V4 Du Virus De La Maladie De Newcastle Avec Comme Support Du Riz Blanc Cuit Et Du Riz Blanc Cru
Résumé Du riz blanc crû et du riz blanc cuit ont été testés en tant que supports de la souche de virus V4 du vaccin contre la maladie de Newcastle. Les résultats de ces expériences d'alimentation ont été évalués par la mesure des anticorps d'inhibition de l'hémagglutination du virus de la maladie de Newcastle.Seule une petite quantité du virus mélangé dans le riz blanc crû a pu être récupérée même immédiatement après le mélange. Par contre la majeur partie du virus a pu être récupérée lorsqu'il s'agissait du riz blanc cuit.Dans 4 expériences séparées, ils n'y avait pas de réponses sérologiques chez les poulets recevant du vaccin ajouté au riz blanc crû. Dans toutes les 4 expériences avec du riz blanc cuit, les réponses sérologiques ont été positives chez les poulets vaccines, 45p. 100 à 100p. 100 des poulets développant des titres suffisamment élevés pour démontrer une protection contre l'épreuve avec du virus virulent. Le développement des anticorps d'inhibition de l'hémagglutination chez quelques poulets témoins a montré la capacité de diffusion latérale ou la persistance dans l'environnement du virus.

One-day-old chickens were transported from Australia to Malaysia and vaccinated orotracheally with an uninactivated vaccine prepared from avirulent Australian V4 strain of Newcastle disease virus (NDV). The vaccination regimes were as follows: group A, once, at 2 weeks old; group B, once, at 3 weeks old; group C, twice, at 2 and at 3 weeks old; group D, direct contact with groups A, B, and C; and group E, indirect contact with groups A, B, C, and D. Group F was unvaccinated controls. Challenge was with NDV virulent Ipoh AF 2240-226 strain, administered at 4 weeks old intramuscularly to 10 chickens in each group and orotracheally to 10 chickens in each group. The remaining chickens were challenged by contact with the inoculated chickens. Group mortalities following challenge were: A, 1/77; B, 1/34; C, 0/39; D, 0/45; E, 6/43; and F, 60/60.  相似文献
