Biosynthesis, molecular structure, and domain architecture of potato suberin: a (13)C NMR study using isotopically labeled precursors

Although suberin in potato wound periderm is known to be a polyester containing long-chain fatty acids and phenolics embedded within the cell wall, many aspects of its molecular structure and polymer-polymer connectivities remain elusive. The present work combines biosynthetic incorporation of site-specifically (13)C-enriched acetates and phenylalanines with one- and two-dimensional solid-state (13)C NMR spectroscopic methods to monitor the developing suberin polymer. Exogenous acetate is found to be incorporated preferentially at the carboxyl end of the aliphatic carbon chains, suggesting addition during the later elongation steps of fatty acid synthesis. Carboxyl-labeled phenylalanine precursors provide evidence for the concurrent development of phenolic esters and of monolignols typical of lignin. Experiments with ring-labeled phenylalanine precursors demonstrate a predominance of sinapyl and guaiacyl structures among suberin's phenolic moieties. Finally, the analysis of spin-exchange (solid-state NOESY) NMR experiments in ring-labeled suberin indicates distances of no more than 0.5 nm between pairs of phenolic and oxymethine carbons, which are attributed to the aromatic-aliphatic polyester and the cell wall polysaccharide matrix, respectively. These results offer direct and detailed molecular information regarding the insoluble intermediates of suberin biosynthesis, indicate probable covalent linkages between moieties of its polyester and polysaccharide domains, and yield a clearer overall picture of this agriculturally important protective material.  相似文献   

Molecular architecture of the multiprotein splicing factor SF3b

The splicing factor SF3b is a multiprotein complex essential for the accurate excision of introns from pre-messenger RNA. As an integral component of the U2 small nuclear ribonucleoprotein (snRNP) and the U11/U12 di-snRNP, SF3b is involved in the recognition of the pre-messenger RNA's branch site within the major and minor spliceosomes. We have determined the three-dimensional structure of the human SF3b complex by single-particle electron cryomicroscopy at a resolution of less than 10 angstroms, allowing identification of protein domains with known structural folds. The best fit of a modeled RNA-recognition motif indicates that the protein p14 is located in the central cavity of the complex. The 22 tandem helical repeats of the protein SF3b155 are located in the outer shell of the complex enclosing p14.  相似文献

Data from the Global Oscillation Network Group (GONG) project and other helioseismic experiments provide a test for models of stellar interiors and for the thermodynamic and radiative properties, on which the models depend, of matter under the extreme conditions found in the sun. Current models are in agreement with the helioseismic inferences, which suggests, for example, that the disagreement between the predicted and observed fluxes of neutrinos from the sun is not caused by errors in the models. However, the GONG data reveal subtle errors in the models, such as an excess in sound speed just beneath the convection zone. These discrepancies indicate effects that have so far not been correctly accounted for; for example, it is plausible that the sound-speed differences reflect weak mixing in stellar interiors, of potential importance to the overall evolution of stars and ultimately to estimates of the age of the galaxy based on stellar evolution calculations.  相似文献

The consensual response of the pupil in the cat was driven by means of a light flux impinging on the contralateral retina. Spike trains recorded extracellularly from single units in the midbrain show correlation with the concurrently recorded pupil area. The temporal dynamics found confirm two earlier studies of sinigle-unit responses and quantitative nerve stimulation. Both of these indicate that most of the 200-millisecond transport delay resides in the neuromuscular apparatus. Amoxycillin as an alternative to dihydrostreptomycin sulphate for treating cattle infected with Leptospira borgpetersenii serovar hardjo

Objective To assess the effect of amoxycillin treatment on urinary excretion of leptospires from cattle infected with Leptospira borgpetersenii serovar hardjo .
Design A chemotherapy trial with controls.
Procedure Fourteen heifers serologically negative to L hardjo were inoculated with L hardjo via the conjunctival route and assessed for evidence of infection by serological, fluorescent antibody and microbiological tests. Two injections (48 h apart) of amoxycillin at a dose of 15 mg/kg were administered intramuscularly to seven heifers 6.5 weeks after infection; the remaining heifers acted as untreated controls. Later, these seven control group heifers were treated with a single dose of amoxycillin (15 mg/kg). Samples of urine were collected before and after amoxycillin treatments; kidneys were collected at slaughter, and examined by fluorescent antibody test and microbiological culture.
Results Leptospires were isolated from the urine of 11 of 14 heifers inoculated with L hardjo . After treatment of six of these with two injections of amoxycillin, leptospires were not isolated. Of the controls, four of the five initially leptospiruric heifers continued to shed leptospires; after a single injection of amoxycillin, no leptospires were detected in the kidneys of these four.
Conclusion Amoxycillin may be an acceptable alternative to dihydrostreptomycin sulphate for the treatment of cattle infected with L hardjo .  相似文献

The clinical features of 24 cases of disseminated canine histoplasmosis are presented. The enteric form predominated and the age at presentation was from five months to ten years. The principal clinical findings were chronic diarrhea, weight loss, pyrexia and anemia.

A premortem diagnosis was reached in 20 cases, by demonstrating Histoplasma capsulatum organisms in peripheral blood smears, rectal scrapings or surgical biopsies. Five of seven dogs treated with amphotericin B were released in asymptomatic condition. Four of these cases relapsed six to 15 months following therapy. The overall mortality rate was 80%.

Optimizing Phosphorus Fertilizer Management in Potato Production

Management of fertilizer phosphorus (P) is a critical component of potato production systems as potato has a relatively high P requirement and inefficiently uses soil P. Phosphorus promotes rapid canopy development, root cell division, tuber set, and starch synthesis. Adequate P is essential for optimizing tuber yield, solids content, nutritional quality, and resistance to some diseases. Although soil test P is the primary tool for assessing P fertilizer needs, in some areas petiole P analysis has been successfully utilized to guide in-season P applications. Potato has been shown in some studies to respond to fertilizer P at soil test levels considered very high for most other crops (100+ mg kg^?1 Bray P₁ or Mehlich I or III and 20+ mg kg^?1 sodium bicarbonate) especially on medium- to finer-textured soils. Even on high-testing soils, fertilizer P rates for top yields sometimes exceed 150 kg P₂O₅ ha^?1. In addition, many states/provinces continue to recommend half or more of the amount of P in the harvested portion of the crop irrespective of soil test P level. In most situations, few differences are expected among fertilizer P sources; however, high rates of diammonium phosphate (DAP) or urea-phosphate (UAP) should not be band-applied in contact or near the seed piece. Most research determined that fertilizer P was most efficiently used when band-applied at planting (e.g., 5 cm to each side of the seed piece); however, some western USA work on high-pH soils showed increased yields and petiole P levels with preplant broadcast applications. In-season applications with the irrigation water can be successful when the potato roots are sufficiently close to the soil surface; however, most research indicates that P applications are more effective when applied at planting or early in the season. Potato fertilizer phosphorus best management practices include: (1) apply the fertilizer P rate calibrated for local soils; (2) band-apply fertilizer P at least 5 cm from the seed piece, especially on very sandy soils or where DAP or UAP are used; (3) use petiole P tests to determine the need for in-season applications; (4) account for all P sources applied, including animal manures; and (5) utilize the best soil conservation practices to reduce P losses to surface waters.  相似文献

Huckleberry Gold is a purple-skin, yellow-flesh fresh market cultivar with similar culinary qualities to the market standard Yukon Gold. It has lower specific gravity, sucrose and vitamin C content, but a significantly higher level of tuber antioxidants than Yukon Gold. Notable disease resistant characteristics are Potato virus X resistance based on the presence of molecular markers for the PVX resistance genes, Nb and Rx1. In addition it also has the H1 gene present which confers resistance to the potato cyst nematode, Globodera rostochiensis, which has been confirmed by bioassay to pathotype Ro1. The size profile of Huckleberry Gold is smaller than Yukon Gold, allowing a better fit into specialty markets that are geared to smaller size for fresh use. Huckleberry Gold represents the first purple-skin, yellow-flesh cultivar to come from the Northwest (Tri-State) Potato Variety Development program.  相似文献
