PrP(c) (cellular prion protein, CD230) expression by subpopulations of lymphoid cells has been widely investigated in a variety of species, possibly because of the possible link between transmissible spongiform encephalopathies (TSE) transmission and blood transfusion. However, the role of the immune cells in the transmission of the disease is still unclear. Here we describe the optimisation and standardisation of a three-colour staining procedure to detect PrP in association with phenotypic and activation markers in ovine immune cells. We demonstrate a reproducible, flexible and sensitive method and that the combination of isotype-specific antibodies and Fab fragments is feasible. To our knowledge, this is the first report of such labelling of ovine cells. Using this method, we were able to detect differences in levels of PrP expression between blood and lymph node cells of the same animal, and considerable variability between animals. Moreover, we were able to explore possible associations between PrP expression and cellular activation and to identify cell subsets with different labelling patterns. We are currently employing this approach to evaluate variations in immunological parameters during experimental infection in sheep.  相似文献

A short-term anaerobic incubation technique using the C2H2 inhibition of N2O-reductase for comparing denitrification potentials of soils is described. Twenty grams of soil with added NO?13 are incubated in the presence of He and 0.1 atm C2H2 at 25°C and 0 soil matric potential for 8 h. N2O evolution is linear within 60 to 120 min. The denitrification potential of soils stored at 4°C decreased markedly over 21 days of storage in accordance with changes in the available C. Denitrification under an anaerobic atmosphere was observed at 4 C. Denitrification potentials were independent of NO?33 concentrations above 25 μg NO?3-N g?1 soil. Biphasic linear rates of N2O evolution were observed in one soil. Incubation of this soil with chloramphenicol suggested the first linear phase is attributable to the in situ enzyme activity at the time of sampling. The second linear phase is indicative of the dentrification potential and is attributed to the full induction of denitrifying enzymes. The denitrification potential of a soil was maintained at or close to the maximum for 8 months of the year. During midsummer months the denitrification potential decreased markedly and the soil demonstrated a biphasic rate of denitrification suggesting an in situ denitrification activity less than the maximum potential. Results indicate that the maximum denitrification potential of this soil may often be limited not by NO?3 but by available C.  相似文献

The localization of gammadelta T cells within epithelia suggests that these cells may contribute to the down-regulation of epithelial malignancies. We report that mice lacking gammadelta cells are highly susceptible to multiple regimens of cutaneous carcinogenesis. After exposure to carcinogens, skin cells expressed Rae-1 and H60, major histocompatibility complex-related molecules structurally resembling human MICA. Each of these is a ligand for NKG2d, a receptor expressed by cytolytic T cells and natural killer (NK) cells. In vitro, skin-associated NKG2d+ gammadelta cells killed skin carcinoma cells by a mechanism that was sensitive to blocking NKG2d engagement. Thus, local T cells may use evolutionarily conserved proteins to negatively regulate malignancy.  相似文献

A marker-assisted back-cross (MABC) programme was used to introgress four root quantitative trait loci (QTLs) from the tropical japonica rice variety Azucena into the Indian upland rice variety, Kalinga III. Previously we tested the products for root traits and reported that the introgressed QTL9 (on chromosome 9) significantly increased root length in the new genetic background. Here we describe field testing for agronomic traits in near-isogenic lines (NILs) that differ for introgressed QTLs. Four NILs were selected and characterised in replicated field experiments in eastern and western India over 3 years. They were tested by upland farmers in a target population of environments (TPE) in three states of eastern India, over 2 years. NILs out-performed Kalinga III for grain and straw yield and there was interaction between the genotypes and the environment (G × E). No effect was found for the root QTL9 on grain or straw yield, however, the presence of several introgressions significantly improved both traits. Some of this effect was due to introgression of Azucena alleles at non-target regions. Overall, the Azucena introgressions increased straw yield more than grain yield. While it has yet to be demonstrated whether this effect is due to improved root systems, this finding fits with the assumption that introgressed genes are involved in partitioning of biomass to the roots and stems, rather than to the grain. The NILs could replace Kalinga III for cultivation in medium upland environments in eastern India.  相似文献

Degradation of added folic acid and native folates in micronutrient‐fortified corn masa and tortillas was evaluated using masa prepared from either nixtamalized corn flour or fresh nixtamal. Variations in masa pH, masa holding time at an elevated temperature, and iron source failed to show significant differences in folate loss in corn flour masa prepared in the laboratory. Masa was subsequently prepared from fresh nixtamal in a commercial mill in Mexico, and fortified with one of two different micronutrient premixes containing iron, zinc, B‐vitamins, and either unencapsulated or lipid‐encapsulated folic acid. Folate loss in commercial masa increased significantly with prebake masa holding time for both premixes. Unencapsulated folic acid showed a 73% loss after 4 hr of holding, compared to 60% loss for encapsulated. The difference was statistically significant, indicating a protective effect from the lipid coating. No significant differences in folate levels were found between prebake masa and baked tortillas. Holding baked tortillas for up to 12 hr also had no effect on folate levels. Native folate showed no significant losses throughout the process. Results from the commercial tortilla mill indicate that most of the loss in added folic acid occurs during prebake holding of masa, possibly from microbial degradation.  相似文献

In flour milling, a granulation sensor for ground wheat is needed for automatic control of a roller mill's roll gap. A near‐infrared (NIR) reflectance spectrometer was evaluated as a potential granulation sensor of first‐break ground wheat using offline methods. Sixty wheat samples, ground independently, representing six classes and five roller mill gaps, were each used for calibration and validation sets. Partial least squares regression was used to develop the models with cumulative mass of size fraction as the reference value. Combinations of four data pretreatments (log (1/R), baseline correction, unit area normalization, and derivatives) and three wavelength regions (700–1,500, 800–1,600, and 600–1,700 nm) were evaluated. Unit area normalization combined with baseline correction or second derivative yielded models that predicted well each size fraction of first‐break ground wheat. Standard errors of performance of 4.07, 1.75, 1.03, and 1.40 and r2 of 0.93, 0.90, 0.88, and 0.38 for the >1,041‐, >375‐, >240‐, and >136‐μm size ranges, respectively, were obtained for the best model. Results indicate that the granulation sensing technique based on NIR reflectance is ready for online evaluation.  相似文献
