The aim of the study was to determine the changes in composition and physicochemical features (pH, density, thermostability and acidity) of mare colostrum and milk, and of protein fraction contribution (serum albumin, β-casein, γ-casein, α-lactalbumin, G class immunoglobulins) depending on lactation stage. The research material was colostrum and milk samples from 12 Arabian mares. Colostrum samples were collected within 2 h after parturition and milk samples were collected twice, in the 3rd and 6th weeks of lactation. The level of basic milk components decreased significantly (only lactose content increased) as compared to colostrum. Total bacteria count and somatic cell count decreased significantly with an increase in resistance and urea level. The changes observed were connected to differentiated contribution of particular protein fractions and their relative proportions. Lower levels of γ-casein (P ≤ 0.05), β-casein, serum albumin as well as α-lactalbumin were observed in colostrum as compared to those in milk. Any relationship between lactation stage and β-casein content was observed. Serum albumin and α-lactalbumin content increased in subsequent milkings. The level of G class immunoglobulins decreased significantly and its highest level was noted in colostrum. Any significant differences between the 3rd and 6th lactation weeks were obtained.  相似文献

Excessive nutrient loading is a major threat to aquatic ecosystems worldwide that leads to profound changes in aquatic biodiversity and biogeochemical processes. Systematic quantitative assessment of functional ecosystem measures for river networks is, however, lacking, especially at continental scales. Here, we narrow this gap by means of a pan-European field experiment on a fundamental ecosystem process--leaf-litter breakdown--in 100 streams across a greater than 1000-fold nutrient gradient. Dramatically slowed breakdown at both extremes of the gradient indicated strong nutrient limitation in unaffected systems, potential for strong stimulation in moderately altered systems, and inhibition in highly polluted streams. This large-scale response pattern emphasizes the need to complement established structural approaches (such as water chemistry, hydrogeomorphology, and biological diversity metrics) with functional measures (such as litter-breakdown rate, whole-system metabolism, and nutrient spiraling) for assessing ecosystem health.  相似文献

Using fenylephrine, isoprenaline, noradrenaline, adrenaline and receptor blockers, adrenergic regulation of metabolism in working heart as well as in skeletal resting muscle of rats was investigated. Changes were determined in: phosphocreatine level and32P incorporation into phosphocreatine; phosphorolytic and hydrolytic activities of glycogenolysis; glycogen synthetase activity and14C-glucose incorporation into glycogen; and the level of cAMP. It was found that, contrary to the β-receptor, stimulation of the α-adrenergic receptor inhibits catabolism of macroenergetic phosphates and diminishes glycogen decomposition on the phosphorolytic pathway while increasing the hydrolysis of glycogen and glycogen synthetase activity as well as glucose incorporation into glycogen. On the basis of these data a hypothesis may be put forward that the influence of the α-adrenergic receptor results in the diminishing of energy utilization and in the activation of glycogen anabolism in heart and skeletal muscle. An adrenergic intrasystemic receptor antagonism in the regulation of energy consumption by heart and skeletal muscle is indicated. The α-receptor seems to be an adrenergic intrasystemic moderator of the metabolism rate which is accelerated by β-receptor influence. This suggests that besides sympathetico-parasympathetic intersystemic antagonism regulating the heart work and thereby cardiac energy consumption, there is a separate adrenergic mechanism directly controlling the balance between catabolism and anabolism in heart muscle. This mechanism, based on α- and β-adrenergic receptor antagonism, also controls the metabolic balance in skeletal muscles.  相似文献

Context

Black poplar (Populus nigra L.) is an alluvial forest tree species whose genetic pool is decreasing in Europe. Poplar trees produce short-lived seeds that do not store well.

Aim

The feasibility of seed storage in conventional and cryogenic conditions after their desiccation from water content (WC) of 0.15 to 0.07 g H2O g?1 dry mass (g g?1) was investigated.

Methods

Seed germinability was evaluated (seeds with a radicle and green cotyledons were counted) after storage of seeds for a period of 3 to 24 months at different temperatures: 20°, 10°, 3°, ?3°, ?10°, ?20° or ?196°C.

Results

Seeds desiccated to a 0.07 g g?1 WC can be stored successfully at ?10 °C and ?20 °C for at least 2 years. A significant decrease in germination was observed only after 12 months of seed storage (WC 0.15 g g?1) at temperatures above 0 °C. We demonstrated that both fresh (0.15 g g?1 WC) and desiccated (0.07 g g?1 WC) seeds can be preserved at ?196 °C for at least 2 years.

Conclusions

Seed storage temperature and time of storage were statistically significant factors affecting seed storability. The presented data provide a foundation for the successful gene banking of P. nigra seeds.  相似文献

Auto- and alloplasmic doubled haploid (DH) lines of barley were examined for susceptibility to Fusarium culmorum (W.G.Sm.) Sacc. (isolate KF350) seedling blight. Inoculated kernels were incubated at 20 °C in a chamber saturated to 100% RH a with 12/12 h dark/light cycle. Germination capacity, disease score on a 5-degree scale and root length were evaluated. The data were analysed statistically using three-factor analysis of variance. It was observed that the infection score of roots of DH lines with H. bulbosum cytoplasm was higher than that of their autoplasmic analogues. Length of infected seedling roots expressed as per cent of the control root length ranged from 81 to 56% in autoplasmic DH lines and from 60 to 48% in alloplasmic lines. These differences were statistically significant (at P = 0.01). It was evident, that H. bulbosum cytoplasm increase susceptibility of barley genotypes to Fusarium seedling blight.  相似文献

Summary The doubled haploid (DH) system and the single seed descent (SSD) technique are frequently applied in breeding of self-pollinated crops to rapidly obtain homozygous lines from heterozygous hybrids. This study presents a comparison of populations of barley DH and SSD lines in terms of traits of stem structure. The SSD and DH lines derived from F1 and F2 hybrids Roland x Apex were examined in a field experiment. On the basis of a comparison of means, variances and correlations between traits in the F1DH, F2DH and SSD populations the occurrence/absence of linkage between genes responsible for the analysed traits was inferred. Independent inheritance was found for 1000-grain weight and the length of particular internodes, spike length and stem wall thickness. Moreover, no linkage was found for stem wall thickness and spike length, length of internodes I, II and thickness of stem walls, stem diameter and thickness of stem walls. The results obtained for the other pairs of traits indicate the presence of linkage.  相似文献

In the present study 20 staphylococci isolated from lymph node abscesses of 19 goats of two herds in Western Poland could be identified as Staphylococcus aureus subsp. anaerobius. All 20 strains grew under microaerobic conditions, were negative in the catalase test, showed the typical phenotypic properties of 5. aureus and could genotypically be identified by a positive sa442, 235 rDNA, nuc, coa and spa PCR reaction. The variable regions of the coa and spa gene of the 20 strains appeared with uniform amplicon sizes, respectively. All 20 strains were negative for 12 additionally investigated enterotoxin encoding genes, tst and ssl7 and positive for the gene cap8. Identical properties could be observed for S. aureus subsp. anaerobius DSM 20714. Amplification and sequencing of kat gene of a single Staphylococcus aureus subsp. anaerobius strain of the present study and S. aureus subsp. anaerobius DSM 20714 revealed a complete identity of the kat sequences of both strains and a katB sequence obtained from GenBank (AJ000471). The bacteria were additionally investigated for relatedness by macrorestriction analysis of chromosomal DNA with subsequent pulsed-field gel electrophoresis (PFGE), yielding, corresponding to the above mentioned PCR results, identical PFGE patterns for all 20 Staphylococcus aureus subsp. anaerobius strains isolated in Western Poland and the S. aureus subsp. anaerobius reference strain DSM 20714.This indicates the clonal identity of the strains isolated in Western Poland and the S. aureus subsp. anaerobius reference strain. The route of infection of the two herds in Western Poland with a bacterial clone originally isolated in Spain remains unclear.  相似文献

Betanidin is a basic betacyanin with a 5,6-dihydroxyl moiety which causes its high antioxidant activity. For the purpose of structural study, the enzymatic oxidation of betanidin and betanin (5-O-glucosylated betanidin), followed by chromatographic separation of the oxidation products with spectrophotometric and mass spectrometric detection (LC-DAD-MS/MS) was performed. Within the pH 4-8 range, two main oxidation peaks of betanidin were observed, betanidin quinonoid (possibly betanidin o-quinone) and 2-decarboxy-2,3-dehydrobetanidin, whereas at pH 3 only dehydrogenated and decarboxylated derivatives were detected, suggesting different stabilities of the products at different pH values. The presence of two prominent oxidation products, 2-decarboxy-2,3-dehydrobetanidin and 2,17-bidecarboxy-2,3-dehydrobetanidin, at pH 3 indicates their generation via two possible reaction routes with two different quinonoid intermediates: dopachrome derivative and quinone methide. Both reaction paths lead to the decarboxylative dehydrogenation of betanidin. Subsequent oxidation and rearrangement of the conjugated chromophoric system results in the formation of 14,15-dehydrogenated derivatives. Betanin is oxidized with generation of a quinone methide intermediate, which rearranges to 2,3-dehydro- or neoderivatives. The products of enzymatic oxidation of betacyanins thus formed are derivatives of 5,6-dihydroxyindole and related structures known as the key intermediates in melanogenesis.  相似文献
