A quick method for separation of semivolatile organic compounds from fat in adipose tissue has been developed. This method uses commercially available solid phase cartridges for sample cleanup. The results indicate that the recoveries, from hexane-extracted fat, of 4 representative classes of organic compounds range from 86.2 to 116%. The solid phase cartridges provide excellent separations of the fat from the analytes; no extraneous interference peaks were detected in the gas chromatograms. The method requires only 0.1 g sample and is quick and simple to use. Although results are reported for samples containing 1-14 ppm, the final extract can be concentrated to a volume allowing detection between 10 and 100 ppb. 相似文献

Wild banana Musa balbisiana Colla is one of the progenitors of cultivated bananas and plantains. It is native to Southeast Asia and the western Pacific.
South China represents the northern limit of its distribution range. The genetic diversity of Musa balbisiana was assessed by the amplified fragment length polymorphism (AFLP) fingerprinting in 15 populations of China. Four primer

The objectives of this study were to isolate the antioxidative components in the broth filtrate of Aspergillus candidus (CCRC 31543), to characterize their antioxidative properties, and to evaluate their safety. Three major compounds were isolated and identified as 3,3' '-dihydroxyterphenyllin, 3-hydroxyterphenyllin, and candidusin B. In the linoleic acid peroxidation system, the inhibition of peroxidation in these three compounds was greater than 95% and was significantly higher than that of alpha-tocopherol but equal to that of BHA at 12.5-200 microg/mL. As measured using the Rancimat method in lard, 3,3' '-di-OH-terphenyllin exhibited a protection factor value of 7.82, which was substantially higher than those of BHA (5.58) and alpha-tocopherol (4.29) at 200 microg/mL. 3,3' '-di-OH-terphenyllin and 3-OH-terphenyllin also exhibited marked scavenging effects on the alpha,alpha-diphenyl-beta-picrylhydrazyl radicals (94.7 and 96.0%, respectively), which were similar to those of BHA and alpha-tocopherol. Safety studies showed that these three compounds were neither cyto- nor geno-toxic toward human intestine 407 (INT 407) cells, nor mutagenic toward Salmonella typhimurium TA98 and TA100. 相似文献

Effects of dehulled adlay on the culture count of some microbiota and their metabolism in the gastrointestinal tract of rats 总被引:3,自引:0,他引:3
Experiments were conducted to study the effect of a dietary supplement of dehulled adlay (Coix lachryma-jobi L. var. ma-yuen Stapf) on the culture counts of some important groups of intestinal bacteria and their metabolism in the gastrointestinal (GI) tract of Sprague-Dawley rats. Rats were divided into four groups, and each group was fed a diet containing different levels of dehulled adlay for 30 days as follows: 0% (control), 5%, 20%, and 40%. All animals fed with adlay had normal healthy intestinal walls and no pathogenic signs whatsoever. There were no significant differences in body weight gain or the cecal pH between different groups of rats. Both the 20% and 40% groups had lower culture counts of enterics in their feces than the 5% and control groups, whereas the culture counts of fecal lactic acid bacteria were higher in feces of rats fed with adlay than in the control group. Cecal total short-chain fatty acid (SCFA) content and fecal SCFA were significantly higher in the 20% and 40% groups than in the control and 5% groups. All the adlay-fed rats had a higher fecal butyric acid concentration than the control rats. It is concluded that adlay has a significant influence on the growth of intestinal bacteria, which may ultimately affect the physiology and other functions of GI tracts of rats. 相似文献

Nattokinase, a serine protease, and pronattokinase, when expressed in Escherichia coli, formed insoluble aggregates without enzymatic activity. For functional expression and purification, nattokinase or pronattokinase was first overexpressed in E. coli as an insoluble recombinant protein linked to the C terminus of oleosin, a structural protein of seed oil bodies, by an intein fragment. Artificial oil bodies were reconstituted with triacylglycerol, phospholipid, and the insoluble recombinant protein thus formed. Soluble nattokinase was subsequently released through self-splicing of intein induced by temperature alteration, with the remaining oleosin-intein residing in oil bodies and the leading propeptide of pronattokinase, when present, spontaneously cleaved in the process. Active nattokinase with fibrinolytic activity was harvested by concentrating the supernatant. Nattokinase released from oleosin-intein-pronattokinase exhibited 5 times higher activity than that released from oleosin-intein-nattokinase, although the production yields were similar in both cases. Furthermore, active nattokinase could be harvested in the same system by fusing pronattokinase to the N terminus of oleosin via a different intein linker, with self-splicing induced by 1,4-dithiothreitol. These results have shown a great potential of this system for bacterial expression and purification of functional recombinant proteins. 相似文献

There is a growing need to understand, and ultimately manage, carbon storage by forest ecosystems. Broad-leaved evergreen forests of Taiwan provide an outstanding opportunity to examine factors that regulate ecosystem carbon storage. We utilized data from three Taiwan Forest Dynamics Plots (FS, LHC, and PTY) in which every tree is identified, measured, tagged and mapped, to examine factors regulating carbon storage as estimated from aboveground biomass. Allometric equations were used to estimate the aboveground biomass of each tree, and a model building procedure was used to examine relationships between plot-level aboveground biomass (AGB; Mg/ha) and a suite of topographic and biotic factors. We found that our study sites have AGB values comparable to some of the most carbon dense forests in the world. Across all three sites, maximum biomass was contained in the taxonomic families Fagaceae, Lauraceae and Theaceae. In the FS site, we identified slope convexity (P = 0.03) and elevation (P < 0.001) as topographic predictors of AGB and found that maximum AGB was found in topographically flat areas. In FS, stem density (P < 0.001) was a significant biotic predictor of AGB and the maxima occurred at intermediate densities. In LHC, we found that convexity (P < 0.001) and slope (P < 0.001) were significantly related to AGB which was maximized along a topographic ridge in the plot. Species richness (P < 0.001) was a significant biotic predictor of AGB in LHC, and the relationship indicated slightly higher AGB at higher levels of species richness. The only significant factor related to AGB in PTY was species richness (P = 0.02). Further work is needed to seek a mechanistic understanding of topographic factors and species richness as drivers of carbon storage in forests. 相似文献

The human gut is colonized with a vast community of indigenous microorganisms that help shape our biology. Here, we present the complete genome sequence of the Gram-negative anaerobe Bacteroides thetaiotaomicron, a dominant member of our normal distal intestinal microbiota. Its 4779-member proteome includes an elaborate apparatus for acquiring and hydrolyzing otherwise indigestible dietary polysaccharides and an associated environment-sensing system consisting of a large repertoire of extracytoplasmic function sigma factors and one- and two-component signal transduction systems. A young and energetic pulsar powers the well-known Crab Nebula. Here, we describe two separate gamma-ray (photon energy greater than 100 mega-electron volts) flares from this source detected by the Large Area Telescope on board the Fermi Gamma-ray Space Telescope. The first flare occurred in February 2009 and lasted approximately 16 days. The second flare was detected in September 2010 and lasted approximately 4 days. During these outbursts, the gamma-ray flux from the nebula increased by factors of four and six, respectively. The brevity of the flares implies that the gamma rays were emitted via synchrotron radiation from peta-electron-volt (10(15) electron volts) electrons in a region smaller than 1.4 × 10(-2) parsecs. These are the highest-energy particles that can be associated with a discrete astronomical source, and they pose challenges to particle acceleration theory. 相似文献

This study compared the differences of two types of buckwheat sprouts, namely, common buckwheat ( Fagopyrum esculentum Moench) and tartary buckwheat ( Fagopyrum tataricum (L.) Gaertn.), in general composition, functional components, and antioxidant capacity. The ethanol extracts of tartary buckwheat sprouts (TBS) had higher reducing power, free radical scavenging activity, and superoxide anion scavenging activity than those of common buckwheat sprouts (CBS). As for chelating effects on ferrous ions, CBS had higher values than TBS. Rutin was the major flavonoid found in these two types of buckwheat sprouts, and TBS was 5 fold higher in rutin than CBS. The antioxidant effects of buckwheat sprouts on human hepatoma HepG2 cells revealed that both of TBS and CBS could decrease the production of intracellular peroxide and remove the intracellular superoxide anions in HepG2 cells, but TBS reduced the cellular oxidative stress more effectively than CBS, possibly because of its higher rutin (and quercetin) content. 相似文献

Four food grade additives-sodium ascorbate, sodium bicarbonate, sodium carbonate-10-hydrate, and ferrous sulfate-7-hydrate-were selected as the basic ingredients to formulate the controlled atmosphere agents which could effectively remove oxygen and release carbon dioxide. The mathematical models giving the relationships between the formulations and the responses (oxygen and carbon dioxide contents) were developed using response surface methodology (RSM). Within 8-24 h, the oxygen and carbon dioxide contents of all tested formulations could reach constant levels, in the ranges of 2-9% and 0-41%, respectively. These formulations were considered to be effective, safe, and easy to prepare and could be applied to wide varieties of food products. 相似文献
