A large collection of good genetic markers is needed to map the genes that cause human genetic diseases. Although nearly 400 polymorphic DNA markers for human chromosomes have been described, the majority have only two alleles and are thus uninformative for analysis of genetic linkage in many families. A few known marker systems, however, detect loci that respond to restriction enzyme cleavage by producing a fragment that can have many different lengths. This polymorphism is due to variation in the number of tandem repeats of a short DNA sequence. Because most individuals will be heterozygous at such loci, these markers will provide linkage information in almost all families. Ten oligomeric sequences derived from the tandem repeat regions of the myoglobin gene, the zeta-globin pseudogene, the insulin gene, and the X-gene region of hepatitis B virus, were used to develop a series of single-copy probes. These probes revealed new, highly polymorphic genetic loci whose allele sizes reflected variation in the number of tandem repeats. 相似文献

Fibroblasts were genetically modified to secrete nerve growth factor (NGF) by infection with a retroviral vector and then implanted into the brains of rats that had surgical lesions of the fimbria-fornix. The grafted cells survived and produced sufficient NGF to prevent the degeneration of cholinergic neurons that would die without treatment. In addition, the protected cholinergic cells sprouted axons that projected in the direction of the cellular source of NGF. These results indicate that a combination of gene transfer and intracerebral grafting may provide an effective treatment for some disorders of the central nervous system. 相似文献

Because of the small thermal capacity of ultrathin ( approximately 200 nanometers) metal single crystals, it is possible to explore the coupling of catalytic and thermal action at low pressures. We analyzed a chemothermomechanical instability in this regime, in which catalytic reaction kinetics interact with heat transfer and mechanical buckling to create oscillations. These interacting components are separated and explored through experimentation, mathematical modeling, and scientific computation, and an explanation of the phenomenon emerges from their synthesis. 相似文献

This study is the first attempt using Levins's Theory (loop analysis) in order to develop a sustainable management for the scallop, Argopecten purpuratus, fishery in Peru during El Niño‐Southern Oscillation events (ENSO) and upwelling conditions. Based on this theoretical framework, it was possible to estimate the local stability for each of these model systems and to follow the qualitative changes of the variables in response to external factors.
- 2. Based on our results, we suggest the following management policies to be implemented: (1) during ENSO events the size at the first capture of the scallops should be >70 mm and (2) the increase in the number of fishermen during ENSO events must be prevented. Both measures increase the sustainability of fishery under ENSO and upwelling conditions. The ecological models predict that during ENSO and upwelling events, any management strategy to increase the recruitment of the scallop would not have a positive impact on the adult stock.
- 3. Finally, we suggest that more efforts must be focused on the development of extended eco‐social models, which incorporate further social and economic variables, increasing realism of the abstractions for this fishery activity.

OBJECTIVES: To determine retrospectively the prognosis and outcome for dogs diagnosed with thoracolumbar intervertebral disc disease treated with partial percutaneous discectomy (PPD). METHODS: Three hundred and thirty-one dogs presenting with symptoms of thoracolumbar intervertebral disc disease from 1998 to 2003 were treated with PPD. Diagnosis and location of intervertebral disc disease was confirmed by clinical examination, radiography, myelography and magnetic resonance imaging. PPD was performed via fluoroscopy-guided removal of a 5 mm bore cylinder out of the central intervertebral space. RESULTS: Clinical success after surgery was achieved in 159 (88.8 per cent) grade II to IV patients and 58 (38.2 per cent) grade V patients. The mean (sd) time from percutaneous discectomy to first improvement was 8.3 (13.2) days. CLINICAL SIGNIFICANCE: The PPD approach to the thoracolumbar spine involves minor trauma (yielding rapid recovery) and less pain, and produces results comparable with open fenestration. Consequently, this simple minimal invasive technique can be recommended as an alternative to the technique of fenestration and can be easily performed in addition to open surgical decompression techniques or prophylactically. However, it is not a replacement for surgical treatment in dogs with thoracolumbar disc disease that require removal of disc fragments causing spinal cord or nerve root compression. 相似文献

We have modified surface catalytic activity in real time and space by focusing an addressable laser beam to differentially heat a platinum (110) single-crystal surface. Ellipsomicroscopy imaging of local conditions (such as reactant and product local coverages) enabled us to close the loop between sensing and actuation (both spatiotemporally resolved). Pulses and fronts, the basic building blocks of patterns, could be formed, accelerated, modified, guided, and destroyed at will.
