OBJECTIVE: To determine serum amyloid A (SAA) concentrations in serum and synovial fluid from healthy horses and horses with joint disease and assess the effect of repeated arthrocentesis on SAA concentrations in synovial fluid. Animals-10 healthy horses and 21 horses with various types of joint disease. PROCEDURES: Serum and synovial fluid samples were obtained from each horse. In 5 of the 10 healthy horses, arthrocentesis was repeated 9 times. Concentrations of SAA were determined via immunoturbidometry. RESULTS: Serum and synovial fluid SAA concentrations were less than the assay detection limit in healthy horses and did not change in response to repeated arthrocentesis. Synovial fluid SAA concentrations were significantly higher in horses with suspected bacterial joint contamination or infectious arthritis, or tenovaginitis than in healthy controls, and serum concentrations were significantly higher in horses with infectious conditions than in the other groups. Neither serum nor synovial fluid SAA concentrations in horses with low-inflammation joint conditions differed significantly from those in healthy controls. Concentrations of SAA and total protein in synovial fluid were significantly correlated. CONCLUSIONS AND CLINICAL RELEVANCE: Synovial fluid SAA concentration was a good marker of infectious arthritis and tenovaginitis and appeared to reflect changes in inflammatory activity. The advantages of use of SAA as a marker include the ease and speed of measurement and the fact that concentrations in synovial fluid were not influenced by repeated arthrocentesis in healthy horses. Further study of the SAA response in osteoarthritic joints to assess its usefulness in diagnosis and monitoring of osteoarthritis is warranted. 相似文献

Vaccination of cattle against Mycobacterium avium subsp. paratuberculosis (MAP) provides partial protection by delayed shedding of MAP and reduced numbers of clinically affected animals. The duration of vaccine induced immune response is not known. The primary objective of this study was therefore to characterize the long-term effect of whole-cell based vaccination against MAP on the immune response. A secondary objective was to evaluate whether immunodiagnosis of MAP and Mycobacterium bovis infections is affected by MAP vaccination. Two studies were performed: (1) A retrospective longitudinal study including 895 vaccinated and 2526 non-vaccinated dairy cows in 9 Danish dairy herds aiming at characterizing the long-term antibody-response to vaccination; and (2) a cross-sectional study of responses in the IFN-γ assay carried out in 140 vaccinated animals in two herds to evaluate the effect of vaccination on the cell-mediated immune response and to evaluate a possible interference with the diagnosis of M. bovis infections. The results showed that 37% of samples from vaccinated animals and 5% of samples from non-vaccinated animals, respectively, were test positive in the milk antibody ELISA. The prevalence of antibody responses of the vaccinated animals was relatively constant from 2 to 6 years of age, but decreased in older animals. Among the 140 vaccinated animals 88% tested positive with the IFN-γ test to johnin PPD and 50% responded to PPDb with IFN-γ production above a similar cut-off. Although Denmark is free of M. bovis, two of the vaccinated animals responded with higher IFN-γ levels when cultured with PPDb compared to PPDa. In conclusion, immunization with whole-cell MAP vaccines elicits both humoral and cell-mediated immune reactions, which may interfere with surveillance and diagnosis of both MAP and M. bovis infections using currently available tests. 相似文献

Background
A high use-efficiency of fertilizer N remains essential to sustain high crop productivity with low environmental impact. However, little is known on the long-term lability of mineral fertilizer N.Aims
To quantify crop uptake and leaching of 15N-labelled mineral fertilizer that has been retained in an agricultural soil for 25–30 years in crops with variable growing season.Methods
A field plot received 15N-labelled mineral fertilizers over a period of 5 years and was then kept under arable cropping for 12 years. After relocation to 16 lysimeters, the topsoil grew set-aside grassland for the next 13 years. Then crop uptakes and leaching losses of 15N remaining in soil was tested over a 2-year period by either converting set-aside grass to production grassland, or by replacing it with spring barley (+/− autumn cover crop) or vegetation-free fallow. All treatments received unlabelled mineral N fertilizers.Results
Crop uptake and leaching of 15N were generally highest in the first test year after termination of the set-aside. The leaching of residual 15N in soil declined in the order: vegetation-free soil (4.7%), spring barley (1.9%), spring barley + cover crop (0.7%) and production grassland (0.2%). Corresponding losses for the second leaching period were 2.7%, 0.9%, 0.4% and 0.06%. There was a fixed relationship between leaching losses of 15N and total N.Conclusions
After residing in soil for 25–30 years, the lability of labelled mineral N fertilizer residues appeared slightly higher than the lability of bulk soil N. Autumn vegetation was crucial for reducing leaching losses. 相似文献24.
The epidemic outbreak in northern Europe of Neonectria neomacrospora, the causal agent of dieback in Abies spp., led the European and Mediterranean Plant Protection Organization (EPPO) to include the pathogen on its alert list in 2017. Effective monitoring of this pathogen calls for a rapid and sensitive method of identification and quantification. A probe‐based real‐time PCR (qPCR) assay based on the β‐tubulin gene was developed for the detection and quantification of N. neomacrospora in infected wood samples, and directly for ascospores. This study presents the first published species–specific molecular detection assay for N. neomacrospora. The analytical specificity was validated on taxonomically closely related fungal species as well as on 18 fungal species associated with the host (Abies sp.). The analytical sensitivity was tested on naturally infected wood, on purified pathogen DNA in a matrix of host DNA and on N. neomacrospora ascospores for detection of airborne inoculum. The latter was tested both with a DNA extraction step prior to qPCR and without DNA extraction by direct qPCR on collected ascospores. The assay was specific to N. neomacrospora, with a sensitivity of 130 fg purified DNA, or 10 ascospores by direct qPCR. Life history theory predicts a trade‐off between migration and residency where migration is favoured when it infers elevated fitness. Although migration to more favourable environments may offer higher growth rates, migrants often experience increased mortality due to predation. Here, we investigated mortality and migration behaviour of the North Sea houting (Coregonus oxyrinchus), an anadromous salmonid endemic to the Wadden Sea. We used acoustic telemetry to map the migration of the only remaining indigenous population by applying stationary hydrophones combined with manual tracking. Data suggested a total mortality of 26%, with 30% of the total mortality attributed to predation by great cormorants (Phalacrocorax carbo sinensis), highlighting that North Sea houting conservation could be jeopardised by increased cormorant predation. Risk of cormorant predation was size‐dependent, with smaller fish suffering higher risk of predation. The study found North Sea houting to exhibit disparate migration strategies and identified a lentic area in the estuary as an important habitat. Two newly established artificial lakes within the river system significantly reduced the migration speeds, possibly indicating constrained navigation through the lakes. The migration into the Wadden Sea correlated with temperature perhaps indicating osmoregulatory constraints of sea entry. Unlike many salmonid species, migration occurred both day and night. Moreover, fish exhibited repeatable individual differences in diel activity patterns, suggesting that individuals differ consistently in their migratory activity throughout the 24‐hr period. Global maps of thermal and fast neutron fluxes from the moon suggest three end-member compositional units. A high thermal and low fast neutron flux unit correlates with the lunar highlands and is consistent with feldspathic rocks. The South Pole-Aitken basin and a strip that surrounds the nearside maria have intermediate thermal and fast neutron flux levels, consistent with more mafic rocks. There appears to be a smooth transition between the most mafic and feldspathic compositions, which correspond to low and high surface altitudes, respectively. Co‐digestion of crop biomass improves the traditional manure‐based biogas yield due to an increased content of easily degradable carbon compounds. In this study, the methane potential of three perennials (grass, legumes, and grass+legume) was determined using various amounts together with animal manure. The nitrogen (N) mineralization dynamics in soil and the N‐fertilizer value of the derived digestates were subsequently tested in both a soil incubation study and a pot experiment with spring barley. Digestion of all tested perennials together with a manure‐based inoculum increased the cumulative methane yield four to five times compared to digestion of the inoculum alone, with the highest increases observed with pure grass. However, the methane potential decreased along with increasing grass biomass concentration. In the plant pot experiment, all tested digestates increased barley shoot biomass by 40–170%, to an extent statistically comparable to mineral N fertilizer. However, the application of the digestate originating from fermentation with pure grass resulted in lower plant growth and a more fluctuating soil mineral N content throughout the incubation study compared to the other digestates. Considering the high dry matter and methane yield ha?1, the possibility to substitute mineral N fertilizer inputs by leguminous biological N2 fixation capacity, and the digestate fertilizer value, the integration of grass–legume mixtures or sole legumes into anaerobic digestion systems as co‐substrate for manure seems to be promising. This could furthermore contribute to the diversification of cropping systems for bioenergy production. 相似文献

The combination of temperature and precipitation during the growth season could be correlated with the occurrence of Gremmeniella abietina epidemics in Denmark. The Gremmeniella index value (GIV) is the result of the monthly rain in mm divided by average monthly temperature in centigrade (mm/°C) and it was created to facilitate easy prediction of epidemics in Denmark. The two most severe outbreaks of G. abietina in pine and other conifers in Denmark took place in 1963–64 and 1984–85, following years where low average temperatures and high precipitation in May combined with the same weather patterns in either August or September. These meteorological conditions were not fulfilled from 1874 to 1900, in which period Pinus nigra was pronounced unsuitable for Danish forestry because of recurring problems with fungal disease. However, the earlier epidemics seemed to coincide with cold and rainy weather (high GIV) in August. In addition, severity of epidemics may also depend on the presence of dense stands between 30 and 40 years of age, as high GIV in August has occurred without disease outbreak in periods with low frequency of susceptible stands. Gremmeniella abietina attacks in Denmark are initiated by the climatic conditions expressed by the Gremmeniella index value, although it does not explain the biological process behind this phenomenon. Damage by Neonectria neomacrospora and Adelges piceae in provenance trials of subalpine fir (Abies lasiocarpa) in Denmark

Field trials of Abies lasiocarpa were undertaken with the aim of assessing the potential for Christmas tree production in Denmark. Twenty‐six provenances originating from Alaska to New Mexico were tested. Damage by the insect Adelges piceae and the fungus Neonectria neomacrospora was recorded for the first time 8 and 12 years after the initial planting. Damage from N. neomacrospora increased rapidly in the period 12–15 years after planting. Trees from the northern provenances and humid climates exhibited less damage than those from southern ones. Previous attack by A. piceae had a minor effect on N. neomacrospora infection. Greenhouse tests showed that detached shoots from healthy Abies lasiocarpa can be used to rank provenances for resistance to N. neomacrospora, but results varied according to host subspecies. 相似文献
