A pseudorabies virus (PRV) mutant with deletions in genes for glycoprotein X (gX) and thymidine kinase, designated delta GX delta TK, was constructed and evaluated as a vaccine for protecting swine against PRV-induced mortality. Doses greater than or equal to 10(3) plaque-forming units (PFU) of this strain given to mice provided protection from challenge exposure with virulent PRV. Sera tested from mice inoculated with delta GX delta TK had high titers of neutralizing antibody to PRV, but reactivity in the same sera was not significantly different from that in sera from noninoculated mice (controls) when sera from both groups were evaluated by use of an ELISA with gX antigen produced in Escherichia coli. Compared with noninoculated pigs (controls), those given delta GX delta TK (greater than or equal to 10(2) PFU) were protected completely from lethal challenge exposure, without experiencing adverse effects on weight gain and with reduction of shedding of virulent challenge virus. Serotest results indicated that, although inoculated pigs responded with strong neutralizing antibody titers, the response of delta GX delta TK-inoculated pigs to gX, as determined by ELISA before challenge exposure, was not significantly greater than the ELISA values obtained from control pigs. The ELISA values from a group of pigs inoculated with a commercially available vaccine were significantly (P less than 0.05) higher than those of control pigs. The experimental vaccine, delta GX delta TK, was avirulent for mice, swine, and sheep, but was mildly virulent for calves (mortality, 1 of 12) and more virulent for dogs (mortality, 3 of 6) and cats (mortality, 2 of 6).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

A method of simultaneous amplification at three different loci (Y-specific DNA, kappa-casein gene, growth hormone gene) in cattle is presented. The use of this procedure for genetic typing on bovine crude DNA samples from less than ten diploid cells is shown. Regulators need a reliable, precise and easy to use tool for predicting the occurrence of pesticides in drain water and catchments in agricultural areas. Occurrence depends on a wide range of substance and site specific factors and this study presents a simple model built on the results from simulations of a detailed model system that does not neglect or omit any of these factors. A drainage dominated sub-catchment (0.03 km2) of the Lillebaek catchment (4.4 km2) on Funen, Denmark, represented by the catchment model MIKE SHE is considered. Detailed analyses have been made with respect to geological and hydrodynamic conditions as well as measurements of pesticide concentrations in ground and surface waters. Maximum concentrations in drain water, the time for reaching this concentration and the time interval for exceeding the limit value have been derived empirically from MIKE SHE simulations using degradation rates and sorption coefficients values for 37 pesticides included in the Danish PATE database. The relatively hydrophilic bentazon and hydrophobic pendimethalin are used as model pesticides for illustration. A simple tool applicable for a wide range of pesticides has thus been designed based on detailed analyses of a limited number of pesticides. The user requirements are degradation rates, sorption coefficients, application rates and regulatory limit values for the pesticides of interest.  相似文献

Several methods were developed for the redistribution of nitrogen (N) fertilizer within fields with winter wheat (Triticum aestivum L.) based on plant and soil sensors, and topographical information. The methods were based on data from nine field experiments in nine different fields for a 3-year period. Each field was divided into 80 or more subplots fertilized with 60, 120, 180 or 240 kg N ha−1. The relationships between plot yield, N application rate, sensor measurements and the interaction between N application and sensor measurements were investigated. Based on the established relations, several sensor-based methods for within-field redistribution of N were developed. It was shown that plant sensors predicted yield at harvest better than soil sensors and topographical indices. The methods based on plant sensors showed that N fertilizer should be moved from areas with low and high sensor measurements to areas with medium values. The theoretical increase in yield and N uptake, and the reduced variation in grain protein content resulting from the application of the above methods were estimated. However, the estimated increases in crop yield, N-uptake and reduced variation in grain protein content were small.  相似文献

Dairy cow mortality (unassisted death and euthanasia) has increased, worldwide and in Sweden. On-farm mortality indicates suboptimal herd health or welfare and causes financial loss for the dairy producer. The objective of this study was to identify cow-level risk factors associated with on-farm cow mortality. Cows with at least one calving between 1 July 2008 and 30 June 2009 from herds enrolled in the Swedish official milk recording scheme with >40 cow-years were included. Each cow was followed from the day of calving until she calved again or left the herd (died, slaughtered or sold). The effects of potential risk factors on on-farm cow mortality were analysed using a Weibull proportional hazard model with a gamma distributed frailty effect common to cows within herd. The event of interest (failure) was euthanasia or unassisted death. An observation was right censored if the cow was slaughtered, sold, calved again or had an on-going lactation at 500 days after calving. The lactations were split into seasons (January to April, May to August and September to December) and at 30 and 100 days in milk in order to evaluate seasonal effects and the effect of disease in different lactation stages. Primiparous and multiparous cows were analysed separately. The highest hazards for both primiparous and multiparous cows were found for traumatic events and diseases, both in the lactation stage in which the cow died and in the preceding stage. The hazard was higher in early lactation and lower in 2nd parity compared to higher parities. Increased age at first calving (for primiparous cows), calving between January and April, dystocia and stillbirth also increased the mortality hazard. Differences were also found between breeds, between milk production parameters at first test milking and between management types. The results from this study show the importance of good management and preventive health actions, especially around calving, to avoid mortality in dairy cows.  相似文献

Pathology of the colon in pigs infected with Trichuris suis and fed an inulin and a non-inulin containing diet was studied to investigate the inflammatory response induced and the influence of inulin on this. Twenty-nine pigs were allocated into four groups (N-7, I-7, N-9 and N/I-9). Groups N-7, N-9 and N/I-9 were given a diet with non-fermentable carbohydrates, and group I-7 was fed a diet including fermentable carbohydrates (inulin). After 2 weeks, all pigs were inoculated with 2000 T. suis eggs. Seven weeks post-infection (p.i.), groups N-7 and I-7 were killed and group N/I-9 changed to inulin diet to study the influence of inulin on already established T. suis. Nine weeks p.i., groups N-9 and N/I-9 were killed. Trichuris suis were collected from the colon for enumeration. Tissues from the colon of each pig were taken for histological and immunohistochemical quantitative and semi-quantitative evaluations of heterophils, eosinophils, mast cells, IgA(+), IgG(+), IgM(+) and CD3(+) cells. The findings were compared with tissues from seven uninfected pigs of the same age. Pigs fed inulin had significantly fewer worms compared with pigs on the non-fermentable carbohydrate diet. The number of inflammatory cells in the colonic mucosa was increased in all T. suis-infected pigs compared with uninfected controls. The numbers of eosinophils, mast cells, IgA(+), IgG(+) and CD3(+) cells were significantly lower in pigs fed inulin than pigs fed non-fermentable carbohydrate. Whether this is a direct effect of inulin or a secondary effect due to the lower parasitic burden in inulin fed pigs is not known.  相似文献

In 2004,five veterinarians examined 283 dairy cows from four Danish dairy herds and independently assigned scores for the clinical signs lameness, hock lesions and other cutaneous lesions to each cow using a clinical protocol. We evaluated the inter-observer and the intra-observer agreement using prevalence-adjusted, bias-adjusted kappa (PABAK). We chose two different cut-offs between the ordinal scores for classifying cows as healthy or diseased for each of the clinical signs, and we compared the ability of the observers to discriminate between healthy and diseased cows for the two cut-offs. Without any formal training of the observers we found PABAK in the range 0.36-0.88 and we concluded that the choice of cut-off had an effect on the observed agreement among the observers.  相似文献

Four types of cattle manure were prepared by mixing faeces with different ratios of urine + straw, one manure being faeces without straw and urine. Following 3 to 6 months of storage, the manures were applied in spring or autumn to a sandy loam and a coarse sand in amounts corresponding to 120 kg N ha−1. Spring barley (Hordeum vulgare L.) undersown with perennial ryegrass (Lolium perenne L.) were used as test crops. Barley was harvested at maturity and the wintering ryegrass was cut three times in the second year. Additional mineral N in the form of 15N-labelled mineral fertilizer (50 kg N ha−1) was added to all barley plots at planting in spring. Non-manured plots receiving only the 15N-labelled mineral fertilizer were included. Faeces without urine + straw contained more NH4-N than the other manures and spring incorporation of this manure gave the highest dry matter yield and N uptake of the barley. When faeces without urine + straw was applied in autumn, the yield and N uptake were increased only on the sandy loam. On the coarse sand, the NH4-N was probably leached as NO3-N during the winter. The three manures with urine + straw did not affect barley grain yield or total N uptake. Thus these manures did not mineralize nor immobilize measurable quantities of N in the first growth period. On the sandy loam, the patterns of N uptake in ryegrass indicated an increased availability of manure N during the second growth period. The availability of N from the 15NH415NO3 was similar in all plots when manures were applied in spring. Less 15N was taken up by the spring barley when manure was added in autumn compared with crops given 15NH415NO3 only. Thus short-term effects of N in cattle faeces stored with or without straw + urine are low. For soils low in mineral N such manures may potentially reduce crop N uptake.  相似文献

This study investigated the effects of long‐term annual inputs of animal manure and straw on the rate of gross nitrogen (N) mineralization–immobilization turnover (MIT), net N mineralization and potential nitrification, and examined how these N transformation rates affect plant N availability. The experiment was conducted during May–June 2001 in long‐term field experiments in Askov, Denmark, where organic manure and barley straw had been applied annually for 11 and 20 years prior to the year 2000, respectively. Thus, any differences could be attributed to residual effects from the previous years of application. Inputs of straw and organic manure to soil increased soil organic matter (SOM)‐N content in soil in the order: without straw, without manure < without straw, with manure < with straw, without manure < with straw, with manure. The inputs did not change net N mineralization in the soil. There was a distinct but non‐significant trend towards higher gross N mineralization with increasing SOM‐N. Gross N immobilization was enhanced by straw inputs and to a lesser extent by organic manure inputs, while potential nitrification was enhanced by both amendments. The results show that long‐term annual inputs of straw and organic manure can increase MIT rate and potential nitrification rate without influencing net N mineralization rate. MIT and potential nitrification explained 23–31% of the variation in plant N uptake, while net N mineralization rate only explained 1%. Plant N uptake therefore seems to be more influenced by MIT rate and potential nitrification rate than by net mineralization rate, presumably because mineral N in the transition between gross N mineralization and gross N immobilization is available for assimilation by plants.  相似文献
