Trypanosoma vivax is major cause of animal trypanosomiasis and responsible for enormous economic burden in Africa and South America animal industry. T. vivax infections mostly run low parasitaemia with no apparent clinical symptoms, making diagnosis a challenge. This work reports the design and evaluation of a loop-mediated isothermal amplification (LAMP) test for detecting T. vivax DNA based on the nuclear satellite repeat sequence. The assay is rapid with results obtained within 35 min. The analytical sensitivity is ~ 1 trypanosome/ml while that of the classical PCR tests ranged from 10 to 10(3)trypanosomes/ml. The T. vivax LAMP test reported here is simple, robust and has future potential in diagnosis of animal trypanosomiasis in the field.  相似文献

One goal of mammalian neurobiology is to understand the generation of neuronal activity in large networks. Conceptual schemes have been based on either the properties of single cells or of individual synapses. For instance, the intrinsic oscillatory properties of individual thalamic neurons are thought to underlie thalamic spindle rhythms. This issue has been pursued with a computer model of the CA3 region of the hippocampus that is based on known cellular and synaptic properties. Over a wide range of parameters, this model generates a rhythmic activity at a frequency faster than the firing of individual cells. During each rhythmic event, a few cells fire while most other cells receive synchronous synaptic inputs. This activity resembles the hippocampal theta rhythm as well as synchronized synaptic events observed in vitro. The amplitude and frequency of this emergent rhythmic activity depend on intrinsic cellular properties and the connectivity and strength of both excitatory and inhibitory synapses.  相似文献

Reconstitution of 30S ribosomal particles was performed with 16S ribosomal RNA, "core" proteins, and "split" proteins from 30S particles derived from streptomycin-sensitive and streptomycin-resistant Escherichia coli cells in various combinations. Analysis of streptomycin sensitivity of the reconstituted particles has shown that the alteration induced by the resistance mutation resides in the core proteins, and not in the RNA or in the split proteins of the 30S particles.  相似文献

A new type of electrometer is described that uses a single-electron transistor (SET) and that allows large operating speeds and extremely high charge sensitivity. The SET readout was accomplished by measuring the damping of a 1.7-gigahertz resonant circuit in which the device is embedded, and in some ways is the electrostatic "dual" of the well-known radio-frequency superconducting quantum interference device. The device is more than two orders of magnitude faster than previous single-electron devices, with a constant gain from dc to greater than 100 megahertz. For a still-unoptimized device, a charge sensitivity of 1.2 x 10(-5) e/hertz was obtained at a frequency of 1.1 megahertz, which is about an order of magnitude better than a typical, 1/f-noise-limited SET, and corresponds to an energy sensitivity (in joules per hertz) of about 41 Planck's over 2pi.  相似文献

Data on rocks from Spitsbergen and the equatorial sections of Italy and Slovenia indicate that the world's oceans became anoxic at both low and high paleolatitudes in the Late Permian. Such conditions may have been responsible for the mass extinction at this time. Since many of the problems that beset readers, authors, and publishers of scientific journals are caused by the growth of science or by the frailties of human nature, we cannot hope for complete solutions. In an effort to make progress, within the framework of the possible, we propose that journals stop binding papers into issues and, instead, distribute to each subscriber a personalized stream of papers, abstracts, and titles. This type of distribution, which has been made possible by the advent of high-speed computers, would not affect the traditional roles of editors, referees, and libraries. We also propose that journals recognize the need for very rapid communication in certain fields, and meet the threat of public preprint-exchange systems in these fields by themselves publishing preprints in an appropriately limited manner.  相似文献
