从潜伏期感染马立克氏病病毒(MDV)鸡淋巴组织中提取基因组DNA,采用梯度PCR的方法获得MDV的L-meq、meq基因,将其插入pMD18-T克隆载体,经测序并进行了序列分析。结果表明,L-meq、meq基因同源性很高,L-meq中只有180bp与meq不同。但是采用同样的方法在发病期病鸡淋巴组织中却只能获得MDV的meq基因。为了进一步研究发病期L-meq基因消失的原因,试验对L-meq、meq基因的氨基酸序列的结构域进行了分析。结果表明,L-meq基因中含有9个PRR(proline-rich-reapts)区域,meq中含有6个PRR区域,从而推断PRR区域可能与基因的转录激活及调控病毒DNA复制的功能有关,该180bp插入序列也许能解释初期感染MDV不致瘤的原因,同时也意味着L-meq基因对于潜伏期的维持是必要的。  相似文献

In order to explore the antibacterial activity and mechanism of the Radix isatidis powder water extract,the effects of the Radix isatidis powder water extract on the morphology and structure of Escherichia coli (E.coli) were tested by scanning electron microscopy and transmission electron microscopy.The effects of the Radix isatidis powder water extract on the conductivity and total leakage rate of E.coli and the content of alkaline phosphatase in the culture medium of the content of protein,DNA and RNA were stained by DAPI to detect the effect of the Radix isatidis powder water extract on the nucleic acid of E.coli,and the effect of the Radix isatidis powder water extract on the metabolism of E.coli in vivo,in vitro,ALT,AST,pyruvic acid and ATP.The results showed that after the Radix isatidis powder water extract acted on E.coli for 10 h,it was observed by SEM that the bacteria appeared to overflow and shrink,the length of the bacteria became shorter obviously,many residues were formed due to breakage,some of them were sunken in the middle and deformed.Under TEM,it was observed that the boundary of the cell wall of E.coli was unclear,the wall membrane was zigzag,deformed and some of the bacteria were broken.The protein content outside the cell was significantly different from that in the blank control group from 8 h (P<0.01), and that in the cell from 4 h (P<0.01). DNA content had no significant difference with blank control group before 12 h (P>0.05), but had significant difference with blank control group from 16 h (P<0.01); RNA content began to decrease at 8 h, and was significantly different from blank control group at 12 h (P<0.05), and was extremely significant from 16 h (P<0.01). There was no significant difference between ALT and AST (P>0.05). The pyruvate content in culture medium and bacteria was higher than that in blank control group, and the difference was very significant from 4 h (P<0.01). The ATP content in the culture medium was significantly different from that in the blank control group (P<0.01), and the ATP content in the cell was significantly different from that in the blank control group from 4 h (P<0.01).In conclusion,the Radix isatidis powder water extract could inhibit the synthesis and metabolism of bacterial genetic material and the content of pyruvate and ATP by destroying the integrity of cell wall and cell membrane.  相似文献

Separated lymphocytes from untreated and selenium-supplemented lambs were tested in culture against phytolectins (phytohaemagglutinin and pokeweed mitogen). Levels of response varied considerably among individuals, and this tended to mask the effects of selenium. However, cells from both groups were more active in the presence of serum from supplemented animals, and a similar stimulatory effect was achieved by adding sodium selenite in vitro. Thus selenium could exert a direct influence on the performance of immunocompetent cells.  相似文献

目的观察澳洲茄胺盐酸盐对HeLa细胞c-myc,H—ras和hTRT基因表达的影响.方法用鼠抗人c-mycMcAb,H-ras McAb和hTRTPcAb作为一抗,生物素化羊抗鼠IgG作为二抗,用免疫组化方法测定澳洲茄胺盐酸盐对HeLa细胞c-myc,H—ms和hTRT基因的表达.结果澳洲茄胺盐酸盐处理的HeLa细胞与对照组相比,c-myc,H-ras和hTRT基因的表达均明显降低(P〈0.05).结论澳洲茄胺盐酸盐下调HeLa细胞c-myc和H-ras基因的表达,抑制细胞增殖,诱导分化;澳洲茄胺盐酸盐下调HeLa细胞hTRT基因的表达,抑制端粒酶活性,抑制细胞生长.  相似文献

We tested the effect of extended drying of half the root system on fruit yield and fruit Ca concentration, an indirect measure of fruit quality, in avocado (Persea americana Mill. cv Hass). In a field experiment on a sandy soil, withholding irrigation and plastic sheeting was used to dry the root-zone beneath the whole canopy (DD) or half the canopy (WD), compared with well-watered trees (WW). The irrigation water contained added nutrients and was slightly saline. Yield, shoot growth, leaf conductance, leaf and fruit water status and mineral concentrations of leaves and fruit were studied. The responses of treated trees were assessed in the following season during which normal irrigation practices were restored. With respect to yield, the WD treatment behaved the same as the DD treatment. It reduced yield by more than half and proportionately more than the reduction in water supply thus reducing irrigation efficiency. Re-watering did not restore yield of WD or DD-trees in the next season. The WD and DD treatments had no effect on the concentration of Ca in the fruit mesocarp and so are unlikely to affect fruit quality. The main impact of reduced water supply on the trees was fruit abscission and this was linked to dry soil around the roots rather than the water status of the leaves or fruits. We conclude that extended drying of half of the root-zone in one season reduced irrigation efficiency for two seasons by promoting the abscission of developing fruit to the same extent as occurred when the whole root system was exposed to extended drying.  相似文献

通过文献检索刺五加生境、栽培、光合等对药用成分含量影响研究的相关论文并进行分析总结,讨论生境与刺五加药用成分含量的关系及最佳栽培模式.得知刺五加在野生生境下,黄酮含量与光照呈明显相关,含量呈季节性变化,春秋高,夏季低;皂苷类成分含量则随光强的增大而降低,无季节性变化;其它成分含量无明显的季节变化.较适宜的生境为60%~70%光照,黄酮类与皂苷类均可获得较好的含量与产量.利用不同的栽培密度或遮荫调节光照强度,进行仿生栽培或野生抚育,创造适宜刺五加生活生长的环境条件,并根据其各有效成分在不同部位及不同时期的含量差异,来确定刺五加适宜生产模式及采收期,将有助于刺五加人工栽培产量和药效成分含量的提高.  相似文献
