分析在植物花分子生组织及花器密形成过程中起重要作用的APETALA1(AP1)基因的保守区序列,设计1对长度为23pb的PCR引物,以毛叶枣基因组DNA为模板,采用PCR方法扩增出长为648bp的DNA片段,克隆人pUCm-T载体,测序和序列分析结果 表明获得了毛叶枣AP1同源基因中部的1个片段,该片段有2个内含子,长度分别为152bp和386pb,编码区共编码36个氨基酸,其序列已在GenBank中登记(登记号为AF356541),在GenBank中进行同源性检索,结果表明其氨基酸序列与其他植物AP1同源基因的氨基酸序列同源性高达66%-88%,推测它们在功能上也是相似的。  相似文献

集约经营雷竹林土壤磷素的时空变化   总被引:2,自引:1,他引:2  
2006年,在雷竹Phyllostachys praecox主产区浙江省临安市三口镇,选取集约经营下不同栽植年限(1,5,10,15 a)的雷竹林以及与未栽雷竹的邻近水田(0 a)采集土样,旨在分析集约经营雷竹林土壤磷素随雷竹栽植年限和剖面深度的时空变化规律,以便为雷竹林土壤的可持续利用提供依据。研究表明:雷竹林土壤磷素随栽植年限增长而增加。0,1,5,10和15 a的10~20 cm土层全磷质量分数分别是0.39,0.58,0.58,0.85和1.57 g.kg-1;有机磷质量分数为130.16,179.83,183.61,209.46和262.79 mg.kg-1;有效磷为6.78,17.41,33.49,100.64和326.36 mg.kg-1。土壤中的全磷和有机磷随着栽植年限的增长而稳步上升;速效磷则随着栽植年限的增加而急剧上升。但有机磷占全磷的比例较小并随着栽植年限的增加而更趋下降,表明累积的磷素主要来自大多数还没有被利用而残留的无机磷肥,有机覆盖物对土壤磷素累积的贡献不大。雷竹林土壤全磷、有机磷和有效磷在剖面深度上的分布总体上是随深度的加深而逐渐减少:即表层(0~10 cm)>亚表层(10~20 cm)>底层(20~40 cm)。然而在10 a以前,土壤全磷呈现两头高而中间低的分布态势,1~15 a的土壤有效磷和有机磷均呈现从表层到底层逐渐减少的趋势,其幅度随着栽植年限的增加而扩大。集约经营模式虽然有较高的经济效益,但土壤中大量磷素的累积既是资源的浪费又对环境具有潜在的威胁,应调整施肥措施和配方。图3参20  相似文献

Special diets are used to mitigate many human diseases. When these diets require changes in carbohydrate content, then sweetness becomes an important characteristic. The range of low-calorie sweeteners available to the food industry is expanding. It is essential to have an exact knowledge of the relative sweetness of various sweeteners in relation to different sucrose concentrations. The objective of this study was to determine the variation on the relative sweetness of aspartame (APM), stevia [Stevia rebaudiana (Bert.) Bertoni] leaf extract (SrB) and the mixture cyclamate/saccharin – two parts of cyclamate and one part of saccharin – (C/S) with the increase in their concentrations, and in neutral and acid pH in equi-sweet concentration to 10% sucrose, using magnitude estimation. Sweetness equivalence of SrB in relation to sucrose concentrations of 20% or higher and of APM and C/S to sucrose concentrations of 40% or higher could not be determined, because a bitter taste predominated. The potency of all sweeteners decreased as the level of sweetner increased. In equi-sweet concentration of sucrose at 10%, with pH 7.0 and pH 3.0, the potency was practically the same for all sweeteners evaluated.  相似文献

Chagas’ disease, a vector-transmitted infectious disease, is caused by the protozoa parasite Trypanosoma cruzi. Drugs that are currently available for the treatment of this disease are unsatisfactory, making the search for new chemotherapeutic agents a priority. We recently described the trypanocidal action of (−)-elatol, extracted from the macroalga Laurencia dendroidea. However, nothing has been described about the mechanism of action of this compound on amastigotes that are involved in the chronic phase of Chagas’ disease. The goal of the present study was to evaluate the effect of (−)-elatol on the formation of superoxide anions (O2•−), DNA fragmentation, and autophagy in amastigotes of T. cruzi to elucidate the possible mechanism of the trypanocidal action of (−)-elatol. Treatment of the amastigotes with (−)-elatol increased the formation of O2•− at all concentrations of (−)-elatol assayed compared with untreated parasites. Increased fluorescence was observed in parasites treated with (−)-elatol, indicating DNA fragmentation and the formation of autophagic compartments. The results suggest that the trypanocidal action of (−)-elatol might involve the induction of the autophagic and apoptotic death pathways triggered by an imbalance of the parasite’s redox metabolism.  相似文献

对煤笼红萝卜雄笥不育系及其保持系不同个体发育过程中的碳水化合物含量、淀粉酶活性、可溶性蛋白质含量及脯氨酸含量进行研究。结果表明:在各营养生长期,不育株中的碳水化合物含量高于可育株;而在生殖生长期,不育株花蕾中碳水化合物含量低于可育株。无论在营养生长期带是在生殖生长期,不育株个体中淀粉酶活性均低于可育株。另外,不育株中可溶性蛋白及游离脯氨酸含量低于可育株。上述研究结果表明:不育株中营养物质的缺乏是导致小孢子败育的一个重要原因,而营养器官中的营养物质向生殖器官运输受阻又是引起不育花蕾中物质含量下降的原因之一。  相似文献

试验采用埃希氏大肠杆菌(E.coli)和鼠伤寒沙门氏菌(S.typhimurium),接种于含0.5%单糖和低聚糖的糖发酵培养基内,置于36℃培养箱内发酵6h,转入25℃室温下继续发酵,分别用0.0455 mol/L NaOH溶液滴定产酸量,用倒置产气管测量产气量作为参比试验,计算产酸量和产气量的相对比值,估算非发酵糖的相对含量。试验结果表明:几种低聚糖用埃希氏大肠杆菌和鼠寒沙门氏菌测得的非发酵糖含量分别为:低聚果糖(FOS)52%-54%;低聚异麦芽糖(IMO-500)86%-89%;低聚异麦芽糖(IMO-900)92%-94%;甘露糖蛋白(MOS-Pr)92%-93%。  相似文献

Emergence of multiple pollen tubes from single pollen grains occurred both in vitro and in vivo in sponge gourd (Luffa cylindrica (L.) Roem). The frequency with which pollen grains produced multiple pollen tubes in vivo (7.2%) was lower than that under in vitro conditions (14.9%). In pollen grains germinated in vitro, the total length of the multiple pollen tubes was greater than that of single pollen tubes, but individual tubes among the multiple tubes did not reach the same length as single tubes. Moreover, the growth of the single pollen tubes continued for a longer period in vitro than that of the multiple tubes. Fluorescence microscopy showed that callose was present throughout the pollen tube wall except in the apical part of growing pollen tubes, and nuclei moved into the longest of the multiple tubes. Results of Fourier transform infrared microspectroscopy indicated that abnormal cell wall components (peaks at 800–1000 cm−1) were more frequent in multiple pollen tubes lacking nuclei, and the pectin content (1733 cm−1) in multiple pollen tubes was much lower than that in single pollen tubes. These findings suggested that there were significant differences in pollen tube growth and wall composition between single and multiple pollen tubes, and that multiple pollen tubes had much less opportunity than single pollen tubes to reach the embryo sac and achieve double fertilization.  相似文献

【目的】为了探明盐胁迫下枣蛋白表达的差异,【方法】以较耐盐碱的‘七月鲜’枣品种的扦插苗为材料,经过临界浓度(0.60%)的钠盐(NaCl)处理,取其叶片进行蛋白质双向电泳,经ImageMaster 2D分析,【结果】结果表明,处理与对照之间总蛋白的整体分布模式非常相似,在pH 3~10和MW50~90 kD内的蛋白点分布最多,处理与对照之间量变倍数大于1.5倍的差异点有26个,其中6个蛋白受胁迫上调,20个蛋白受胁迫下调。在上调的蛋白质点中,3个点受盐胁迫诱导增加了近4倍,2个蛋白质点相对丰度增加了2倍以上,表现出强烈的盐诱导特性。说明他们可能在枣的抗盐机制中发挥重要作用。在下调的蛋白质点中,2个点相对丰度降低至对照的5倍左右,表现出强烈的盐抑制特性,说明其可能是对盐胁迫比较敏感的2个蛋白质或多肽,并且在枣的抗盐性机制中也发挥比较重要的作用。【结论】在临界浓度钠盐胁迫下,枣叶片蛋白质表达有显著差异。  相似文献

The unsustainable use of phosphate-based fertilizers can lead to the depletion of accessible phosphorous reserves and cause environmental problems. As well as finding new phosphate supplies, we ought to be prioritizing sustainable phosphate-based fertilizers. By adopting microbial, physical and chemical modifications, low-solubility phosphate minerals could be used in an efficient way for nutrient supply and heavy metal immobilization.
