The availability of cow ovaries from the slaughterhouse has been very limited in Taiwan. To maximize the use of cow ovaries for research purposes, whole ovary dissection was performed and the developmental competence of the oocytes derived from different sizes of follicles was assessed by the rates of in vitro maturation (IVM) and parthenogenetic activation of the oocytes in Experiment 1 (Exp 1). Cumulus-oocyte complexes (COCs) derived from small (1-2 mm) and large (3-8 mm) follicles were subjected to standard IVM culture for 24 h. Mature oocytes were selected and then parthenogenetically activated using A23187 (5 microm, 5 min) or thimerosal (200 microm, 10 min) alone or combined with 6-dimethylaminopurine (2.5 mm and 3.5 h, respectively). Activation rates of the oocytes, neither from the large nor small follicles, were affected by different activation treatments (single or combined stimuli). Whereas maturation rates for the oocytes from large follicles were superior to those from small follicles in both the single (59% vs 45%) and combined treatments (76% vs 40%; p < 0.05). To understand how prolonged heat shock (HS) influences cytoskeletal configurations of mature bovine oocytes, in Experiment 2 (Exp 2), matured oocytes derived from large follicles were randomly allocated to different durations of HS treatments at 41.5 degrees C for 0 (C0h, control, n = 12), 1 (HS1h, n = 28), 2 (HS2h, n = 31), and 4 h (HS4h, n = 30). An additional control group was cultured for 4 h without HS (38.5 degrees C, 4 h, n = 35). Alterations in nuclear structures, microtubules (MTs), and microfilaments (MFs) of the oocytes were examined. Abnormalities in the chromosomes, spindle MTs and the percentages of oocytes with cytoplasmic MTs increased with time of HS treatment. The intensity of the MF distribution in the HS oocytes was also altered. Significant changes in the cytoskeleton after HS may be associated with the reduced development under hyperthermia and, perhaps, with the low pregnancy rates of the animals during hot seasons.  相似文献

The fertility of ram spermatozoa that had undergone flow cytometric sorting (MoFlo SX) and cryopreservation was assessed after low-dose insemination of synchronized Merino ewes. Oestrus was synchronized with progestagen-impregnated pessaries, PMSG and GnRH treatment. Ewes (n = 360) were inseminated with 1 x 10(6), 5 x 10(6) or 15 x 10(6) motile sorted frozen-thawed (S(1), S(5), or S(15) respectively) or non-sorted frozen-thawed (C(1), C(5) or C(15) respectively) spermatozoa from three rams. An additional group of ewes were inseminated with 50 x 10(6) motile non-sorted frozen-thawed spermatozoa (C(50)) to provide a commercial dose control. The percentage of ewes lambing after insemination was similar for C(50) (24/38, 63.2%), C(15) (37/54, 68.5%), S(15) (38/57, 66.7%), S(5) (37/56, 66.1%) and S(1) (32/52, 61.5%) groups (p > 0.05), but lower for C(5) (19/48, 39.6%) and C(1) (19/55, 34.5%) treatments (p < 0.05). This study demonstrates sorted ram spermatozoa are equally fertile to non-sorted spermatozoa even when inseminated at 2% of the dose. Furthermore, at very low artificial insemination doses (1 or 5 million motile) the fertility of sorted ram spermatozoa is superior to non-sorted spermatozoa inseminated in equal numbers. These results have significance for the future commercialization of sex-preselection technology in sheep as a reduction in the minimum effective sperm number will allow a corresponding decrease in the associated cost per dose.  相似文献

Time-resolved absorption spectroscopy on the femtosecond time scale has been used to monitor the earliest events associated with excited-state relaxation in tris-(2,2'-bipyridine)ruthenium(II). The data reveal dynamics associated with the temporal evolution of the Franck-Condon state to the lowest energy excited state of this molecule. The process is essentially complete in approximately 300 femtoseconds after the initial excitation. This result is discussed with regard to reformulating long-held notions about excited-state relaxation, as well as its implication for the importance of non-equilibrium excited-state processes in understanding and designing molecular-based electron transfer, artificial photosynthetic, and photovoltaic assemblies in which compounds of this class are currently playing a key role.  相似文献

One hundred veterinarians and 100 breeders of traditionally docked dogs from Queensland were surveyed by telephone to determine their attitudes towards tail docking. Eighty-four percent of the breeders surveyed were in favour of docking, whereas 83% of veterinarians were opposed to the practice. Most pups were docked between 1 and 3 days of age. All veterinarians surgically amputated the tail, whereas 16% of breeders applied rubber bands to the tail. Seventy-six percent of the veterinarians, but only 18% of the breeders believed that docking caused significant to severe pain. No veterinarians, but 25% of the dog breeders believed that docking was painless. Although recent changes to the Queensland Canine Control Council's rulings allow dogs with intact tails to be shown in traditionally docked classes, the requirement of breed standard was cited as the major reason for tail docking by both breeders and veterinarians.  相似文献

Objective To measure acute pain in sheep, based on a human pain model, and examined changes in both electroencephalogram frequency spectrum and behavioural responses to increased electrical stimulation in sheep. Design Analysis of variance (treatment and animal effects) for stimulus intensity where each animal received each electric shock treatment given in the order 0, 5,10 and 20 mA. Procedure Eight sheep with electrodes implanted over the surface of the brain were examined for escape-avoidance and electroencephalogram responses to four levels of electrical stimulation from 0–20 mA. Results With increasing stimulus intensity at the time of feeding, the sheep were more hesitant to return to the feeder or remain near the feeder following stimulation. There was little difference between the 0 and 5 mA stimuli for any of the behaviour variables (P > 0.05). However, there were marked increases in the time taken to re-approach the feeder after receiving an electric shock of 5 mA and of 20 mA (P < 0.05; mean values 3 and 119 s, respectively) and remaining near the feeder for 5 s (P < 0.001; mean values 10 and 167 s, respectively). Following the stimulus, there was an overall increase in the electroencephalogram power spectrum in the first four seconds, which then rapidly returned to normal. In particular, the 20 mA stimulus resulted in higher absolute power values than in the control (0 mA) treatment for delta 2 (P < 0.001), theta 1 (P < 0.05), theta 2 (P < 0.05), alpha 1 (P < 0.001), alpha 2 (P < 0.001) and beta 1 (P < 0.01) band-widths. Similarly, for the 10 mA stimulus, the absolute power values were greater than the control treatment for delta 2 (P < 0.05), alpha 1 (P < 0.01), alpha 2 (P < 0.001) and beta 1 (P < 0.01) bandwidths. Conclusion The experiment suggests that a human acute pain model is applicable to sheep and that these electroencephalogram changes may provide a good measure of acute pain in sheep.  相似文献

The pathogenesis of chronic rhinitis in cats is poorly understood and consistently effective therapies are not currently available. Therefore, randomized clinical trials were conducted to evaluate a novel immunotherapy for treatment of chronic rhinitis in adult (n=12) and young cats (n=28). In addition, cytokine profiles in cats with chronic rhinitis were compared to those of healthy cats. Cats were treated with a potent stimulator of innate immunity (liposome-IL-2 DNA complexes) and the effects of treatment on clinical signs and immune function were assessed. In adult cats with chronic rhinitis, immunotherapy led to significant improvement in frequency of sneezing but not in other clinical signs when compared to the placebo group, whereas immunotherapy failed to improve clinical signs in younger cats. Analysis of cytokine expression in cats with rhinitis did not reveal evidence of a Th2 cytokine bias in cats with rhinitis. We conclude that chronic rhinitis in cats is not a Th2-biased disease and that immunotherapy may lead to clinical improvement in adult cats with the disease.  相似文献

Two groups of feline panleukopenia virus (FPV), feline calicivirus (FCV), and feline herpesvirus-1 (FHV-1) seronegative cats (five cats per group) were administered one of two modified live feline viral rhinotracheitis, calicivirus, and panleukopenia virus (FVRCP) vaccines and the serological responses to each agent were followed over 28 days. While all cats developed detectable FPV and FCV antibody titers; only two cats developed detectable FHV-1 antibody titers using the criteria described by the testing laboratory. For FPV and FHV-1, there were no differences in seroconversion rates between the cats that were administered the intranasal (IN) FVRCP vaccine and the cats that were administered the parenteral FVRCP vaccine on any day post-inoculation. For FCV, the cats that were administered the IN FVRCP vaccine were more likely to seroconvert on days 10 and 14 when compared to cats that were administered the parenteral FVRCP vaccine.  相似文献

There are few reports which were designed to compare the survival rate of human primary follicles with primordial follicles after cryopreservation. This study was designed to evaluate whether such a difference occurs. Human ovarian biopsies were cryopreserved using dimethylsulphoxide/sucrose as the cryoprotectants. Fresh and cryopreserved ovarian samples were evaluated for viability differences between the two types of follicles using the endpoints of histology, ultrastructure and DNA fragmentation. In comparison with fresh ovarian tissue (83.9% ± 10.0%), the percentage of morphologically normal primordial follicles was not significantly different in cryopreserved tissue (73.9% ± 17.2%). However, a lower percentage of primary follicles with normal morphology was seen in the cryopreserved group (43.3% ± 25.7% vs 74.8% ± 19.4% for the fresh group). Transmission electron microscopy revealed that the cryopreservation did not appear to affect the structural integrity of primordial follicles; however, varying ultrastructural damage to the cytoplasm was observed in the majority of the cryopreserved primary follicles. Using a DNA fragmentation assay, the percentage of apoptotic primordial and primary follicles in the unfrozen (26.3% and 20%) and frozen (23.3% and 25%) ovarian tissue was similar. A higher proportion of primary follicles, compared to primordial follicles, suffer histological damage after slow freezing.  相似文献
