The effects of direct electrical current (DC) and alternating current (AC), were compared with bromoethane (BE), gibberellic acid (GA) and kinetin (Ki) for breaking tuber dormancy of potato (Solanum tuberosum L.). BE was the most effective treatment for high sprout numbers and for short emergence times of sprouts, but the growth regulators (GA and Ki) were most effective in promoting sprout elongation. Electrical currents were more effective for sprout numbers than were growth regulators, except for BE. Amylase and polyphenol oxidase (PPO) enzyme activities were different in the tuber parenchyma and the sprouts of treated tubers. Amylase activity was higher in the parenchyma than in sprouts, in contrast to PPO activity.  相似文献

In this study certain biological and prey consumption features of the predator of mealybug, namely Nephus kreissli Fürsch & Uygun (Coleoptera: Coccinellidae), fed on vine mealybug Planococcus ficus (Signoret) (Hemiptera: Pseudococcidae), were determined under the laboratory conditions. Experiments were performed in the acclimatized room under 28?±?1°C, 60?±?10% r.h. and a photoperiod of 16:8 h(L:D) conditions. To determine the biological features of the coccinellid, predator individuals were continuously observed from egg stage to end of adult stage. The development, survival and fecundity of N. kreissli were determined and all the measured data were recorded daily. The total development time from egg to the eclosion of adult stage was determined as 31.6?±?0.4 days. The lifespan of males and females was 66.0?±?3.8 and 65.8?±?3.5 days, respectively; and a single female laid 122.6 eggs. The raw data related to life history were analyzed by using the age-stage, two-sex life table. The intrinsic rate of increase, the net reproductive rate, and the mean generation time were 0.0709?±?0.0036 d?1, 53.1?±?9.5 offspring and 55.8?±?1.3 days, respectively. The first and second instars of N. kreissli preferred egg stages of P. ficus to other stages of mealybug. On the other hand, third and fourth instars and adult predators preferred the second and third instar nymphs and adult females of the prey. It was determined that the egg consumption of N. kreissli increased with the development of larval stages, and that the highest consumption was recorded in its fourth instar.  相似文献

Melon (Cucumis melo L.) landraces of the Madrid provenance, Spain, have received national distinction for their high fruit quality and sensorial attributes. More specifically, a unique array of Group Inodorus landraces have been continuously cultivated and conserved by farmers in the municipality of Villaconejos since the 19th century. Their genetic relationships to other Group Inodorus and Flexuous melon market classes is not known, and, thus, a study was designed to determine their genetic relationships using 52 simple sequence repeat (SSR) markers, and then make genetic comparisons between these accessions and a previously published “Standard Reference Germplasm Array” (RA) containing Group Inodorus (14 Spanish and one USA), Flexuosus (1 Spanish), and Cantalupensis ( 2 USA) melon accessions. This subset consisted of 15 Spanish Group Inodorus landraces that circumscribed the genetic variation of major Spanish melon market classes (Groups Inodorus and Flexuosus), and USA commercial varieties (Groups Cantalupensis and Inodorus). Based on genetic distances, Villaconejos (Madrid) genotypes differed substantially from RA subset accessions, thus defining their genetic uniqueness. Principal component analysis (PCA) partitioned the accessions examined into four distinct groups revealing that Villaconejos black epidermis melons (landraces ‘Largo’, ‘Largo Negro Escrito’ and ‘Puchero’) were distinctly different from all other accessions examined, as cluster analysis separated Rochet market type Villaconejos’ accessions (landraces ‘Mochuelo’, ‘Mochuelo Tradicional’ and ‘Melón de Villaconejos’) from RA of the same market type. Genetic assessment of principal Spanish market classes revealed comparatively low intra-market heterogeneity in Piel de Sapo type accessions and high heterogeneity in Black and Yellow market type accessions. While a relatively high level of genetic introgression was detected between Yellow and Green market types, black epidermis market types were genetically unique. Given the uniqueness and high genetic diversity resident in Villaconejos landraces, this germplasm pool should be considered as a genetic source for broadening the comparatively narrow genetic base of Group Cantalupensis and Inodorus melon market types, especially standard commercial Spanish Group Inodorus market types (e.g., Piel de Sapo, Rochet, and Canari).  相似文献

This study compared the metabolic fate of [(14)C]-DCP, [(14)C]-residues from radish plants, and purified [(14)C]-DCP-(acetyl)glucose following oral administration in rats. A rapid excretion of radioactivity in urine occurred for [(14)C]-DCP, [(14)C]-DCP-(acetyl)glucose, and soluble residues, 69, 85, and 69% within 48 h, respectively. Radio-HPLC profiles of 0-24 h urine from rats fed [(14)C]-DCP and [(14)C]-DCP-(acetyl)glucose were close and qualitatively similar to those obtained from plant residues. No trace of native plant residues was detected under the study conditions. The structures of the two major peaks were identified by MS as the glucuronide and the sulfate conjugates of DCP. The characterization of a dehydrated glucuronide conjugate by MS and NMR of DCP was unusual. In contrast to soluble residues, bound residues were mainly excreted in feces, 90% within 48 h, whereas total residues were eliminated in both urine and feces. For total residues, the radioactivity in feces was higher than expected from the percentage of soluble and bound residues in radish plants. This result highlighted that less absorption took place when residues were present in the plant matrix as compared to plant-free residues and DCP.  相似文献

The optical sensor for "tea catechins" was built by immobilizing 2,2'-(1,4-phenylenedivinylene)bis-8-hydroxyquinoline (PBHQ) on TiO? nanoparticles (NPs). The sensor worked by "indophenol blue" dye formation on PBHQ-immobilized TiO? NPs as a result of p-aminophenol (PAP) autoxidation with dissolved O? at pH 10. Among quercetin, rutin, naringenin, naringin, gallic acid, caffeic acid, ferulic acid, p-coumaric acid, catechin, epicatechin, epicatechin gallate, epigallocatechin, epigallocatechin gallate, and trolox, only catechin group antioxidants delayed the color formation on NPs, as measured by the reflectance signal at 710 nm. For quantitative analysis, reflectance signal versus time was recorded, and the difference between the areas under curve (ΔAUC) in the presence and absence of catechin was correlated (r = 0.98) to catechin concentration. The selectivity of the sensor for catechins was shown in tea infusions compared to other plant extracts and was ascribed to the nonplanar structure of catechin interfering with the formation of perfectly conjugated indophenol blue on TiO? surface.  相似文献

Protein haze development in white wines is an unacceptable visual defect attributed to slow protein unfolding and aggregation. It is favored by wine exposure to excessive temperatures but can also develop in properly stored wines. In this study, the combined impact of pH (2.5-4.0), ionic strength (0.02-0.15 M), and temperature (25, 40, and 70 °C) on wine protein stability was investigated. The results showed three classes of proteins with low conformational stability involved in aggregation at room temperature: β-glucanases, chitinases, and some thaumatin-like protein isoforms (22-24 kDa). Unexpectedly, at 25 °C, maximum instability was observed at the lower pH, far from the protein isoelectric point. Increasing temperatures led to a shift of the maximum haze at higher pH. These different behaviors could be explained by the opposite impact of pH on intramolecular (conformational stability) and intermolecular (colloidal stability) electrostatic interactions. The present results highlight that wine pH and ionic strength play a determinant part in aggregation mechanisms, aggregate characteristics, and final haze.  相似文献

Marennine, the blue pigment produced by the diatom Haslea ostrearia , exists in two different forms, the intra- and extracellular forms. We investigated the antibacterial, antiviral, and antiproliferative properties of both of these forms. Both forms of marennine inhibited the development of marine bacteria, in particular the pathogenic organism Vibrio aesturianus , at concentrations as low as 1 μg/mL, but they did not display any effect on a wide range of pathogenic bacteria that are relevant for food safety. Both forms of the pigment produced by H. ostrearia also exhibited antiviral activity against the HSV1 herpes virus, with intra- and extracellular marennine having EC(50) values of 24.0 and 27.0 μg/mL, respectively. These values are 2 orders of magnitude higher than the value for the reference drug, Zovirax. Moreover, both forms of marennine were effective in slowing or inhibiting the proliferation of cancer cells. This study confirms the potential of marennine as a biologically active organic molecule, which could have a protective effect on bivalves, which filter seawater and fix the pigment on their gills. Moreover, marennine could be used in food engineering and chemistry as a natural blue pigment. However, despite that it is eaten and possibly assimilated by green oyster consumers, it also deserves in depth evaluation before being considered for use as a nutraceutical.  相似文献

Native agars from Gracilaria vermiculophylla produced in sustainable aquaculture systems (IMTA) were extracted under conventional (TWE) and microwave (MAE) heating. The optimal extracts from both processes were compared in terms of their properties. The agars' structure was further investigated through Fourier transform infrared and NMR spectroscopy. Both samples showed a regular structure with an identical backbone, β-d-galactose (G) and 3,6-anhydro-α-l-galactose (LA) units; a considerable degree of methylation was found at C6 of the G units and, to a lesser extent, at C2 of the LA residues. The methylation degree in the G units was lower for MAE(opt) agar; the sulfate content was also reduced. MAE led to higher agar recoveries with drastic extraction time and solvent volume reductions. Two times lower values of [η] and M(v) obtained for the MAE(opt) sample indicate substantial depolymerization of the polysaccharide backbone; this was reflected in its gelling properties; yet it was clearly appropriate for commercial application in soft-texture food products.  相似文献

Colocasia esculenta (L.) Shott, commonly called taro, is an ancient species selected for its edible tuber. Its huge "elephant ear" like leaves are also consumed in sauces and stews or as soups. Forty-one phenolic metabolites (11 hydroxycinnamic acid derivatives and 30 glycosylated flavonoids) were identified by high-performance liquid chromatography-diode array detection-electrospray ionization/mass spectrometry (HPLC-DAD-ESI/MS(n)) in the leaves of two C. esculenta varieties cultivated in Azores Islands. To our knowledge, 34 of the 41 phenolic compounds are being reported for the first time in this species. Phenolics quantification was achieved by an HPLC-DAD accurate and sensitive validated method. Although the qualitative profile of the two varieties is quite similar, quantitative differences were observed between them. "Giant white" and "red" varieties (local denomination) contain, respectively, ca. 14 and 21% of phenolic acids, 37 and 28% of flavones mono-C-glycosides, 42 and 43% of flavones di-C-glycosides, 3 and 4% of flavones mono-C-(O-glycosyl)glycosides, and both of them ca. 2% of flavones di-C-(O-glycosyl)glycosides and 2% of flavones-O-glycosides. Luteolin-6-C-hexoside was the compound present in higher amounts in both varieties. The established phenolic profile is an added value for the authenticity and quality control of C. esculenta and may be useful in the discrimination of its varieties.  相似文献

The metabolites of three species of Apiaceae, also known as Pegaga, were analyzed utilizing (1)H NMR spectroscopy and multivariate data analysis. Principal component analysis (PCA) and hierarchical cluster analysis (HCA) resolved the species, Centella asiatica, Hydrocotyle bonariensis, and Hydrocotyle sibthorpioides, into three clusters. The saponins, asiaticoside and madecassoside, along with chlorogenic acids were the metabolites that contributed most to the separation. Furthermore, the effects of growth-lighting condition to metabolite contents were also investigated. The extracts of C. asiatica grown in full-day light exposure exhibited a stronger radical scavenging activity and contained more triterpenes (asiaticoside and madecassoside), flavonoids, and chlorogenic acids as compared to plants grown in 50% shade. This study established the potential of using a combination of (1)H NMR spectroscopy and multivariate data analyses in differentiating three closely related species and the effects of growth lighting, based on their metabolite contents and identification of the markers contributing to their differences.  相似文献
