The hemocytes of the American bollwormHelicoverpa armigera (Hübner) were studied in the last instar larvae by phase contrast microscopy to investigate the differences among 14 populations, collected from ten locations, spread over a distance of approximately 3000 km in six different states of India. The cluster analysis of differential hemocytes showed as many as 11 clusters of populations at 5% and six clusters at 10% homogeneity level. The cluster analysis of hemocytes of seven populations from cotton crops showed five and three clusters at 5% and 10% homogeneity levels, respectively. Diversity of hemocytes of four populations from chickpea was wider than that from cotton. The differences in hemocytes of various populations may be due to space, time, host plant, insecticide use, other agroecological conditions, and insect genetic variabilityper se. The field populations ofH. armigera heavily treated with insecticides appeared to contain more spherulocytes at the expense of plasmatocytes and granulocytes. The treatment of 6-day-old larvae ofH. armigera with cypermethrin decreased plasmatocytes and granulocytes, and increased prohemocytes and spherulocytes in the hemolymph of last instar larvae. Provenance variation and genetic variability in pod and seed traits were carried out with 232 candidate plus trees of Pongamia pinnata collected from selected agro-ecological zones of southern peninsular India during February–March 2008 to 2010. Significant variation (P < 0.01) for pod and seed traits across zones and provenances within zones was recorded. The southern dry and transition zone of Karnataka showed the highest mean value for all the pod and seed traits. A significant negative (P < 0.05) correlation and positive correlation was recorded for latitude with 100-seed weight and seed germination, respectively. Altitude positively correlated with 100-pod weight and negatively correlated with oil content. Phenotypic co-efficient of variation was higher than the genotypic co-efficient of variation indicating the influence of environment. Central dry zone and eastern dry zone of Karnataka showed high heritability and genetic advance for 100-pod weight, 100-seed weight, and seed germination. Positive significant (P < 0.05) genotypic correlation of co-efficient was recorded between seed thickness and oil content (0.211). Germination percentage showed significant positive correlation with seed weight and oil content. Path analysis revealed that seed thickness contributed directly to oil content (0.272). Based on D2 analysis, pongamia accessions could be grouped into seven clusters. Maximum diverse Clusters I and VII were identified based on inter-cluster distance (40.19). Thus on the basis of the present findings, it might be suggested that accessions from these clusters could be most suitably used for hybridization and in situ germplasm conservation.  相似文献

The reactive oxygen species (ROS) which are produced during storage of boar semen are causing oxidative stress and leads to poor fertility. Also, tropical and sub-tropical weather condition adversely impacts the physicomorphological quality and fertility of boar sperm. The aim of this study was to examine the effects of feeding linseed oil to boar on its seminal attributes, sperm kinetics, biomarkers of antioxidant, fatty acid profile of seminal plasma (SP) and sperm and in vivo fertility. Six Hampshire crossbreed boars were fed with 90 ml linseed oil (LIN) whereas six Hampshire crossbreed boars were fed 90 ml canola oil (CON) for 16 weeks. Sperm quality was evaluated (60 ejaculates for each group; a total of 120 ejaculates) for motility, livability, abnormal morphology, acrosomal membrane integrity, hypo-osmotic swelling test (HOST) and sperm kinetic parameters by computer assisted semen analysis (CASA) at 0 h and at 72 h of storage at 17°C. Biomarkers of antioxidant (glutathione peroxidase; GPx, catalase; CAT, total antioxidant capacity; TAC) and malondialdehyde (MDA) were measured in SP and serum. Gas chromatography–mass spectrometry (GC–MS) was used for the estimation of fatty acid composition of SP and sperm. Boars fed with linseed oil had higher semen volume (p < .01) and more total sperm numbers (p < .01). Feeding linseed oil to boar enhanced seminal attributes (p < .05) at 0 h as well as at 72 h of storage. Linseed oil feeding (p < .01) improved biomarkers of antioxidants and significantly (p < .01) lowered the lipid peroxidation in serum and SP. Linseed oil feeding (p < .05) increased the proportion of alpha linolenic (ALA), arachidonic and docosahexaenoic (DHA) fatty acids in SP. The ratio of n-6 to n-3 fatty acids in sperm increased significantly (p < .01) in treatment group. Farrowing rate was significantly (p < .05) higher in treatment group. In conclusion, feeding linseed oil to boar improved the in vivo fertility, enhanced antioxidant capacity and increased the DHA content of SP and sperm.  相似文献

Lymphomatoid granulomatosis (LYG) is a rare variant of an angioinvasive T-cell lymphoproliferative disorder that primarily affects the lungs, with common sites of metastasis including the skin and subcutis. In humans, it is a B-cell lymphoproliferative disorder associated with Epstein-Barr virus infection. Our case is a 7-y-old, spayed female, domestic longhair cat that decompensated and was euthanized following an initial diagnosis of angioinvasive lymphoma from a skin biopsy. Autopsy revealed nodules in the lungs and subcutis, and corneal thickening and cloudiness. Histologic examination of cutaneous nodules, lungs, and eye showed similar angioinvasive cellular infiltrates and pattern to that of the original skin biopsy, consistent with a diagnosis of LYG. The neoplastic cells displayed CD3-positive immunoreactivity in the skin, eye, and lung, and PCR for antigen receptor rearrangement (PARR) showed T-cell clonality in all tissues tested. This is the third case of LYG to be reported in cats and is the only case in which PARR analysis and immunophenotyping immunohistochemical staining was performed. LYG with ocular involvement has not been reported previously in cats, to our knowledge. Our case demonstrates the necessity for considering LYG when presented with a cat with respiratory signs in conjunction with subcutaneous nodules and ocular lesions.  相似文献

Intergeneric hybridization is one of the effective methods to broaden the genetic base of cultivated species. Raphanus sativus, a member of the subtribe Raphaninae, has very hard pods at threshing time. In an attempt to transfer shattering resistance to Brassica napus, Raphanobrassica was used as the male parent in crosses with Brassica napus. Plantlets were obtained by embryo rescue and were further multiplied in vitro by micropropagation of nodal segments. Morphology, cytology and DNA analysis confirmed the hybrid nature of these plants. They were backcrosscd with Brassica napus and the progeny was raised. Plants of BC1 and BC2 generations showed wide variation in morphology, chromosome number and pollen fertility. Some of the plants showed up to 95 % pollen fertility and resistance to shattering, indicating the potential for developing B. napus with resistance to shattering.  相似文献

Substituting the nuclear genome of Brassica rapa into the cytoplasmic background of Enarthrocarpus lyratus through backcross substitution helped in developing cytoplasmic male sterility (CMS). Alloplasmic male sterile plants had pale green leaves, small flowers with narrow petals and rudimentary anthers. Female fertility, low initially, improved considerably with advanced backcross generations. Male sterility expression was stable throughout the growing season. Except for EC 339014, all B. rapa accessions (38) evaluated were partial maintainers of the male sterility. Introgression of gene(s) for fertility restoration from the cytoplasm donor species was facilitated by homoeologous pairing between B. rapa and E. lyratus genomes, as was apparent from the very frequent occurrence of a trivalent in the monosomic addition plants (2n = 10 II + 1 I). Backcrossing of fertile monosomic addition plants with B. rapa led to the recovery of male fertile plants possessing the stable euploid chromosome number (2n = 20). These plants restored male fertility in crosses with different (lyr) CMS B. rapa genotypes, confirming the introgression of fertility restorer gene(s) from E. lyratus, the cytoplasm donor species.  相似文献
