Several disease indices (DI) have been proposed to assess the severity of phoma stem canker at late growth stages. The DI commonly used in France (called G2 DI) consists of integrating the distribution of the proportions of plants observed within six canker severity classes into a single index. This index uses arbitrarily chosen coefficients associated to each class of canker severity. A given value of this DI can therefore be obtained with different distributions among the canker severity classes. Consequently, it is important to assess to what extent the G2 DI is representative of the distribution of plants in canker severity classes. We show in this study that the G2 DI is a good indicator of the distribution of the observed canker severities, using a dataset from different years, sites and cultivars; this illustrates the robustness of the G2 DI.  相似文献

The use of High Volume Instrument (HVI) to measure cotton lint characteristics produces high dimensional data. A model which utilized Kohonen Self Organizing Maps (SOM) to visualize cotton lint HVI data, k-means clustering technique to cluster the data and Probabilistic Neural Network (PNN) for data classification was designed and tested using Kenyan cotton lint. According to the model the Kenyan cotton lint can be grouped into four clusters, which were successfully classified by using PNN with a correlation coefficient (R-value) of 1.  相似文献

A new methodology based on pulsed lasers has been developed in order to estimate wheat outer layer mechanical properties without sample preparation. Laser experiments were carried out with an Argon Fluoride (λ = 193 nm) excimer laser source delivering pulses of 15 ns duration. Wheat grains from two cultivars were irradiated by single laser pulses with a quasi-uniform irradiation and two fluences (2.5 and 5 J cm−2). The ablation flux was characterized by environmental scanning electron microscopy before measuring the removed material on cross-sections observed by confocal scanning laser microscopy. Specific image treatment was carried out to obtain the ablation flux (amount of removed matter per pulse). Pericarp, seed coat and aleurone layer were gradually ablated under the laser conditions used in this work. Their ablation thresholds were different and could be related to tissue cohesion. Specific behaviour of seed coat layer (8 μm) could be emphasised with this technique. Pulsed laser ablation could be a potential methodology to reveal indirectly wheat grain layer cohesion.  相似文献

Complementarity in acquisition of nitrogen (N) from soil and N2-fixation within pea and barley intercrops was studied in organic field experiments across Western Europe (Denmark, United Kingdom, France, Germany and Italy). Spring pea and barley were sown either as sole crops, at the recommended plant density (P100 and B100, respectively) or in replacement (P50B50) or additive (P100B50) intercropping designs, in each of three cropping seasons (2003–2005). Irrespective of site and intercrop design, Land Equivalent Ratios (LER) between 1.4 at flowering and 1.3 at maturity showed that total N recovery was greater in the pea–barley intercrops than in the sole crops suggesting a high degree of complementarity over a wide range of growing conditions. Complementarity was partly attributed to greater soil mineral N acquisition by barley, forcing pea to rely more on N2-fixation. At all sites the proportion of total aboveground pea N that was derived from N2-fixation was greater when intercropped with barley than when grown as a sole crop. No consistent differences were found between the two intercropping designs. Simultaneously, the accumulation of phosphorous (P), potassium (K) and sulphur (S) in Danish and German experiments was 20% higher in the intercrop (P50B50) than in the respective sole crops, possibly influencing general crop yields and thereby competitive ability for other resources. Comparing all sites and seasons, the benefits of organic pea–barley intercropping for N acquisition were highly resilient. It is concluded that pea–barley intercropping is a relevant cropping strategy to adopt when trying to optimize N2-fixation inputs to the cropping system.  相似文献

Significant differences were found in soil physicochemical and biological effects between various forest restoration approaches of a hilly red‐soil region, southern China. Soil quality was the highest in natural secondary forest (0·95), while in sites revegetated with tea‐oil camellia (Camellia oleifera), Chinese fir (Cunninghamia lanceolata) and slash pine (Pinus elliottii) and control sites, integrated soil quality indices were 0·68, 0·55, 0·36 and 0·04 respectively. The lower soil quality of plantations and controls resulted from increased disruption of soil physical structure, lower quality litter fall, lower litter fall production, reduction in microbial biomass, decline of microbial function and loss of soil nutrients due to periodically artificial tending and accelerated soil erosion. Improvement in soil physicochemical properties and enhanced soil microbial function at a natural secondary forest site demonstrated the inherent restoration of these soils. Hence, natural restoration, as well as reducing human disturbance, is a better approach to improving soil properties than returning farmland to planted woodlands. Low temperature is one of the major abiotic stresses limiting the productivity and geographical distribution of many important crops. To gain a better understanding of chilling stress responses in cotton ( Gossypium hirsutum L . ), we carried out a comparative proteomic analysis. Cotyledon proteins of 1-week-old cotton seedlings treated with or without chilling treatment at 4 °C were extracted, separated by a two-dimensional gel electrophoresis and compared. Among 1500 protein spots reproducibly detected on each gel, 25 protein spots were down-regulated and 29 were up-regulated. Seven cotton proteins were identified by mass spectrometry analysis as beta-globulin A precursor fragments. One was identified as a Rubisco large subunit fragment, providing evidence of both seed storage protein and photosynthetic protein destruction by chilling stress. The related physiological and biochemical analysis showed that there was a significant increase in endopeptidase activity and activated oxygen generation rate, and an obvious decrease in carboxylation efficiency and maximum net photosynthetic rate of cotton seedlings under chilling stress. Based on the above results, the degradation mechanisms of beta-globulin and Rubisco under chilling stress were discussed. In conclusion, our study provides new insights into chilling-stress responses in cotton and demonstrates the advantages of proteomic analysis.  相似文献

In this research we established a particular vector-free and marker-free plant transformation system of maize to overcome the obstacles of biosafety limits. The BADH gene was introduced into maize by pollen-tube pathway, using the principle of minimum linear length of the transformation element, which was composed of only the BADH gene, expression regulatory sequence (35S CAMV promoter, NOS terminator), and T-DNA border sequence at both sides. Twenty-seven of 2076 transformed samples were positive in PCR amplification and the PCR positive rate of T1 generation was 1.3%. Further Southern blotting results indicated that the BADH gene was integrated into maize genome. Transgenic lines of progeny were examined for tolerance to NaCl by induced salt stress with 250 mM NaCl Hoagland solution. After 15 days of treatment, 73.9–100% of the transgenic seedlings survived and grew well, whereas most wild-type seedlings wilted and showed loss of chlorophyll. Only 8.9% of the wild-type plants survived but gradually died after salt stress. The electrical conductivity of the transgenic line of progeny after salt stress was lower than wild type. The transgenic progeny had higher glycinebetaine and Chlorophyll content than wild type after salt stress.  相似文献

Wheat quality properties are genetically determined by the compositions of high and low molecular weight glutenin subunits, grain hardness, polyphenol oxidase (PPO) activity and starch viscosity. Two multiplex PCR assays were developed and validated using 70 cultivars and advanced lines from Chinese autumn‐sown wheat regions. Multiplex PCR I includes molecular markers for genes/loci ω‐secalin, Glu‐B1‐2a (By8), Glu‐D1‐1d (Dx5), Glu‐A3d, Glu‐B3 (for non‐1B·1R type) and Pinb‐D1b targeting improved gluten parameters and pan bread quality. Multiplex PCR II comprises markers for genes/loci Ppo‐A1, Ppo‐D1 and Wx‐B1b targeting improved noodle quality. The results were consistent with those achieved by SDS‐PAGE and RP‐HPLC, indicating that the two multiplex assays were highly effective, with good repeatability and low costs enabling their use in wheat breeding programmes. In total, nine alleles (subunits) at locus Glu‐B1, four at Glu‐D1 and five at Glu‐A3 locus were identified, and the alleles (subunits) Glu‐B1b (7 + 8), Glu‐B1c (7 + 9), Glu‐D1a (2 + 12), Glu‐D1d (5 + 10), Glu‐A3a, Glu‐A3c and Glu‐A3d were most frequently present in the cultivars and lines tested. The 1B·1R translocation was present in 28 (40.0%) lines, whereas the Wx‐B1 null allele for better noodle quality was present in only seven (10.0%) cultivars and advanced lines, and 37 (52.9%) lines had Pinb‐D1b associated with hard grains. The allele Ppo‐A1b on chromosome 2AL associated with lower PPO activity was present in 38 (54.3%) genotypes, whereas the less effective allele Ppo‐D1a on chromosome 2DL, also associated with low PPO activity was present in 45 (64.3%) of genotypes. These two multiplex PCR assays should be effective in marker assisted selection targeting improved pan bread‐making and noodle qualities.  相似文献

White rust (Albugo candida) is a highly destructive disease of oilseed Brassicas such as Brassica juncea and B. rapa, and has caused serious yield losses in Australia, China and India on both species. The first commercial B. juncea varieties are now being deployed in Australia, but their response to Australian strains of A. candida is yet to be defined under Australian field conditions. To identify useful sources of host resistance for Australia, China and India, in B. juncea, three field trials were undertaken in Western Australia. Forty-four B. juncea genotypes, viz. 22 from India, 12 from Australia and 10 from China, were tested. Varying levels of host resistance to Australian strains of A. candida (race 2) were identified among the genotypes from the three countries. Genotypes CBJ-001, CBJ-003 and CBJ-004 from China consistently showed high levels of resistance to A. candida on leaves across the three trials. Overall, the genotypes from China showed the best resistance, followed by the genotypes from Australia, with those from India being the most susceptible. The most susceptible genotypes were RL1359, RH30 and Seetha from India. It is noteworthy that both the incidence and severity of disease reflected varying levels of host resistance in the germplasm from the three countries, irrespective of whether screening was undertaken in the field using natural or artificial inoculation. Differentiation of resistance among these genotypes was similar to that we reported previously for artificially-inoculated seedlings or adult plants under glasshouse conditions, indicating that a choice of options is available to plant breeders to reliably differentiate host resistance among genotypes to white rust in B. juncea.  相似文献
