Age-related macular degeneration (AMD) is the most common cause of irreversible vision loss in the developed world and has a strong genetic predisposition. A locus at human chromosome 10q26 affects the risk of AMD, but the precise gene(s) have not been identified. We genotyped 581 AMD cases and 309 normal controls in a Caucasian cohort in Utah. We demonstrate that a single-nucleotide polymorphism, rs11200638, in the promoter region of HTRA1 is the most likely causal variant for AMD at 10q26 and is estimated to confer a population attributable risk of 49.3%. The HTRA1 gene encodes a secreted serine protease. Preliminary analysis of lymphocytes and retinal pigment epithelium from four AMD patients revealed that the risk allele was associated with elevated expression levels of HTRA1 mRNA and protein. We also found that drusen in the eyes of AMD patients were strongly immunolabeled with HTRA1 antibody. Together, these findings support a key role for HTRA1 in AMD susceptibility and identify a potential new pathway for AMD pathogenesis.  相似文献

The objective of this study was to determine the effect of feeding a fish oil (FO)‐containing diet on lipid and protein metabolism, postprandial glycaemia and body weight (BW) of mature, overweight dogs. Seven female dogs were randomly assigned to one of two isonitrogenous and isocaloric diets, control (CO) or FO (FO), in a crossover design. Experimental periods were 69 day, separated by a washout period of 30 day. At the beginning of the experiment, and at 30 and 60 day of feeding the experimental diets, the dogs were infused with D‐glucose (2 g/kg BW) through an intravenous catheter. Blood samples were collected for 3 hr to perform a glucose tolerance test. Nitrogen balance measurements began at 06:30 on d 63 of each experimental period and ended at 06:30 on d 69. On d 66 of each period, a single dose (7.5 mg/kg) of 15N‐glycine was administered orally for determination of protein turnover. Incremental area under the curve and glucose concentration at peak did not differ between treatments or among sampling days within treatment. Glucose half‐life tended to decrease (< .10) in the FO treatment on day 30 when compared to baseline (day 0). β‐hydroxybutyrate, non‐esterified fatty acid (NEFA) and triglycerides did not differ within or between treatments. Cholesterol decreased (p < .05) on the FO treatment on day 30, 60 and 69 when compared to day 0. High‐density lipoprotein (HDL) decreased (p < .05) in the FO treatment on day 69 when compared to day 0. Body weight, food intake, faecal excretion, DM and N digestibilities, N balance and protein turnover were not different between diets. Overall, FO‐containing diet decreases cholesterol in mature overweight dogs; however, further research is warranted to verify the effects of FO on glucose metabolism.  相似文献

Abstract –  We examined prey utilisation patterns of brown trout ( Salmo trutta Linnaeus) as related to available benthos in a cold tailwater (Little Red River, Arkansas) having low biodiversity of benthic macroinvertebrates and low availability of fish as prey. Benthic macroinvertebrates were sampled monthly in upstream and downstream sites over a 1-year period. Stomach content analysis was also performed on 30 brown trout (10 each of three size classes) collected monthly from each site. Benthic and diet diversities were low at both sites, as isopods ( Lirceus ) accounted for 67% and 51% of the numerical abundance upstream and downstream, respectively, and 80% and 70% of all prey consumed by upstream and downstream brown trout. Physid snails (upstream) and chironomid larvae (downstream) accounted for most of the remaining prey taxa consumed. Piscivory and consumption of terrestrial invertebrates were rare. There was no clear relation between diet diversity and trout size. Densities of benthic macroinvertebrates and prey consumption were significantly greater upstream than downstream. Benthic macroinvertebrate assemblages and prey consumption were seasonally constant at both sites. Generally, prey consumption was consistent with availability of each prey in the benthos; the lack of benthic diversity and overwhelming abundance of one taxon ( Lirceus) most likely contributed to food selection patterns. Although isopods are abundant within this tailwater to serve as a forage base, the displacement of native fish fauna because of the thermal regime of hypolimnetic release from Greers Ferry Reservoir probably serves as a major limitation to brown trout growth.  相似文献

A degenerative skeletal muscle disease with vascular, neurologic, and renal lesions and a probable familial distribution was identified in 4-20-month-old purebred Gelbvieh cattle. Thirteen affected animals were confirmed from 6 separate beef herds, with a mortality rate of 100%. Clinical signs in affected animals consisted of ataxia, weakness, and terminal recumbency. Gross and histologic muscle lesions were indicative of nutritional myopathy of ruminants, with a lack of myocardial lesions in most cases and only rare myocardial changes in a few animals. Acute to chronic lesions in most large skeletal muscle groups consisted of degeneration, necrosis, regeneration, fibrosis, and atrophy. Fibrinoid necrosis of arterioles was a common feature in multiple tissues. Lesions in the spinal cord white matter and peripheral nerves consisted of degeneration of the dorsal columns and axons, respectively. Changes in the kidneys consisted of chronic interstitial nephritis with fibrosis, hyaline droplet change and tubular epithelial vacuolar change and were most severe in the older calves. Intracytoplasmic myoglobin and iron were demonstrated within the hyaline droplets in degenerate renal cortical tubular epithelial cells. Vitamin E levels were deficient in most (6/7) of the animals tested. Investigation of the pedigree of affected animals revealed a common ancestry for all but 1 of the animals whose parentage could be traced. This investigation suggests that a hereditary metabolic defect, possibly involving antioxidant metabolism, could be responsible for this condition. Renal disease, possibly secondary to myoglobinuria, may be unique to this bovine condition.  相似文献

Six Angus steers (260+/-4 kg initial BW) fitted with ruminal, duodenal, and ileal cannulas were used in a 6 x 6 Latin square design to evaluate the effect of feeding poultry by-product meal (PBM) on small intestinal flow and disappearance of amino acids. The diets were provided at 2% of BW on a DM basis, formulated to contain 11.5% CP, and consisted of 49% corn silage, 36% cottonseed hulls, and 15% supplement on a DM basis. Supplements were formulated to contain 37% CP with sources of supplemental N being soybean meal (100% SBM) and 0, 25, 50, 75, and 100% PBM, with urea used to balance for N. Duodenal flow of all amino acids increased linearly (P < .07) as PBM increased in the diet and, except for His, increased (P < .09) for 100% PBM compared with 100% SBM. Similar results were observed for duodenal flow of nonbacterial amino acids, which linearly increased (P < .05) with PBM and were greater (P < .05) for 100% PBM than for 100% SBM. Soybean meal increased (P < .09) the duodenal flow of nonbacterial Lys compared with 0% PBM, and 0% PBM increased (P < .04) flow of Val, Ala, and Pro compared with 100% SBM. Duodenal bacterial essential, nonessential, and total amino acid flows were not affected (P > .80) by PBM; however, they were greater (P < .02) for 100% SBM than for 100% PBM. In addition, nonessential and total bacterial amino acid flows were increased (P < .06) for 100% SBM compared with 0% PBM. Small intestinal disappearance of Lys and Pro increased linearly (P < .09) as PBM increased, and 100% PBM increased (P < .07) disappearance of Arg and Ala compared with 100% SBM. Supplemental N source had no effect (P > .31) on apparent small intestinal disappearance of essential, nonessential, and total amino acids. These data suggest that when PBM, SBM, and urea were used as sources of supplemental N, the daily disappearance of amino acids from the small intestine of steer calves consuming a corn silage- and cottonseed hull-based diet was similar.  相似文献

Responses to prepartum injection of sows with Se and vitamin E (E) were evaluated by determining immunoglobulin (IgA, IgM, IgG) levels in the colostrum and serum of the sows and the serum of their offspring. Fifty-four sows (40 multiparous, 14 primiparous) receiving diets adequate in E and Se according to current NRC (1988) standards were randomly allotted to four treatment groups in which a single i.m. injection of saline (controls), 5 mg of Se, 1,000 IU of E, or both Se and E were given on d 100 of gestation. Sows were bled prior to and 7 d after injection, at farrowing and on d 14 and 28 of lactation. Colostral samples were collected at the initiation of farrowing. Pigs were bled 20 h postpartum and at 14 and 28 d of age. Major immunoglobulin changes in the serum of the sows due to treatment were not seen prior to parturition. Injections of Se and(or) E resulted in higher colostral IgM levels (8.4, 10.7, 9.8 and 9.6 mg/ml, respectively), but only the response from Se was significant (P less than .05). Concentrations of colostral IgA or IgG were not affected by treatment (P greater than .30). Compared with controls, all three treatments increased (P less than .10) IgM concentrations in serum from pigs at birth (28.3, 33.3, 36.0 and 33.5 mg/ml, respectively), whereas IgA and IgG concentrations were not affected (P greater than .30).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Because of the unique features of the ruminant digestive system, variations in diet composition and intake produce dramatic changes in ruminal fermentation. Optimizing nutritional management requires an understanding of how these variations and changes influence digestion and metabolism. Although the pancreas plays a central role in digestion and subsequent nutrient metabolism, relatively little is known about pancreatic adaptation to nutritional changes in the ruminant. Increasing starch intake has been suggested to increase pancreatic alpha-amylase; however, recent work suggests that dietary energy per se may drive these changes, and interactions with other nutrients, such as protein, may exist. Studies describing the influence of altered protein and lipid intakes on pancreatic adaptation in ruminants are lacking. Pancreatic secretion of both insulin and glucagon respond to the intravenous infusion of VFA in a dramatic fashion; however, feeding studies suggest that the influence of VFA on insulin and glucagon may be more subtle. Interactions exist between stimulatory signals and physiological state, such as lactation. Assessment of pancreatic endocrine secretion is further complicated by a variable removal of insulin and glucagon by hepatic tissues. These studies point out that pancreatic hormone secretion is controlled by integrated and complex mechanisms. Studies of these controlling mechanisms should consider the entire array to more fully understand hormone secretion.  相似文献
