通过形态特征、生理生化特征和16SrDNA序列分析,对分离于番茄茎部能较强抑制番茄青枯病菌生长的内生细菌B47菌株进行了鉴定。结果表明,该菌为枯草芽孢杆菌,其最适生长pH5~6,最适生长温度为35℃。室内防治试验结果表明,用淋根法先接种B47菌后接种病原菌和用注射法先接种B47菌后接种病原菌的处理可取得81.25%和92.10%的防效,而用淋根法、注射法同时接种B47菌与病原菌的处理防效较低。  相似文献

参照前人对杂草天敌筛选标准、植物与昆虫互作理论和近年来对柽柳原产地主要天敌生物学特性的研究结果,制定出柽柳天敌遴选量化标准,共计10项特征,满分为47分。并以此对取食柽柳的12种天敌进行了筛选。柽柳条叶甲得分最高(43分),其次是斯氏伞锥象(41分),推荐这2种天敌昆虫作为首选天敌引入美国做进一步评价。  相似文献

采用6种溶剂对番茄茎叶进行平行提取,以葡萄白腐病菌、葡萄黑痘病菌、苹果腐烂病菌、苹果轮纹病菌、苹果斑点落叶病菌、棉花枯萎病菌、瓜类枯萎病菌和番茄早疫病菌为供试菌,对番茄不同提取液进行抑菌活性测定。结果表明,各溶剂提取液对特定病菌都有极好的抑制作用,对葡萄白腐病菌和苹果腐烂病菌的抑制率均为100%,对其它病菌也都有很好的活性。番茄茎叶的水提取液对供试8种病原菌的综合抑制效果最好,对葡萄白腐病菌、葡萄黑痘病菌和苹果腐烂病菌的抑制作用高达100%,对瓜类枯萎病菌抑菌效果最差也为67.74%。,因此,水应作为番茄茎叶农用抑菌活性物质提取的首选溶剂,石油醚可作为有机溶剂提取番茄茎叶农用抑菌活性物质的溶剂。  相似文献

AIM: To investigate the influence of the cornus officinalis glycosides (COG) on immunological function of corneal transplantation model of rats, and to clarify the immunosuppressive mechanism of COG through observing the activation of lymphocytes in blood. METHODS: Wister rats were used as recipients and SD rats were used as corneal graft donors, then the corneal allografts transplantation model on the closed colony rats were set up. Splenocytes proliferation and mixed lymphocyte reaction of Wister rats activated by ConA were observed. The phenotype change of CD4, CD8, CD25 in blood in different time postoperatively were observed by the di-sign flow cytometry, and the rate of CD4/CD8 was calculated. RESULTS: 1. The COG suppressed the proliferation of T lymphocytes and one-way mixed lymphocyte reaction on the corneal allografting. 2. The phenotype change of lymphocytes in boold was as follows: there was no significant difference between the different time of the CD4, CD8 expression and the CD4/CD8 rate in blood of the control group. The CD4 positive cells expressed CD25 postoperatively increased obviously. The CD4/CD8 rate of medicine group had the tendency to decrease. The CD4 positive cells expressed CD25 postoperation in the medicine group were less than that in the control group obviously. CONCLUSION: The suppression of the T lymphocyte proliferation, mixed lymphocyte reaction, CD molecule expressed by the activated T lymphocytes and the IL-2 receptor expression may be the main immunosuppressive mechanisms of Cornus officinalis glycosides on the cell-mediated immunity.  相似文献

AIM: To establish a new human endometial carcinoma cell line for basic and clinical study. METHODS: The minced tumor tissues were inoculated subcutaneously into the back of nude mice. After forming solid tumor, the mice were killed and the transplant was taken out for primery culture. RESULTS: The established cell line was maintained by serial passages, named EAC. EAC cell line grew rapidly,steadily, and it had similar microscopic morphology to adenocarcinoma cell. Chromosome analysis reveal EAC chromosome counts ranging in hyperdiploid. The expression of ER, PR, P53, MDM2 were negative by SP immunohistochemistry. Tumorigenicity of EAC in nude mice was 100%. CONCLUSION: A new human endometrial carcinoma, EAC, is established successfully, which is helpful for the basic and clinical study of endometrial carcinoma.  相似文献

AIM: To investigate the expression of the urotensin Ⅱ (UⅡ) receptor GPR14 in the aorta of apoE knockout mouse. METHODS: The expression of GPR14 in the aorta of apoE knockout C57BL/6J mice at various ages (18 weeks, 28 weeks, and 38 weeks old, respectively) was determined with competitive RT-PCR. A binding assay of [125I]-UⅡ on the aortic tissue was also performed in 28 weeks group. RESULTS: We found significant upregulation of GPR14 mRNA at all three ages. Compared with wild type group at the same age, the GPR14 mRNA level in apoE knockout mice increased 54.2% in 18 week group (P<0.05), 50.0% in 28 weeks group (P<0.05) and 97.0% in 38 weeks group (P<0.01). In the knockout group or in the wild type group, expressions of GPR14 in the 28 weeks time point were significantly higher than that in other two age groups, and there was no difference between the 18 weeks and 38 weeks group. In the binding assay, the Bmax of [125I]-UⅡ to the aorta of apoE knockout mouse at 28 weeks increased 64% compared with the wild type (P<0.01), and no difference about the Kd between the two groups was observed. CONCLUSION: UⅡ and its receptor probably play an important role in the development of atherosclerosis.  相似文献

AIM: To investigate the relation between the time- spatiotemporal expression of caspase-9 and photoreceptor apoptosis as well as electroretinography (ERG) in Royal College of Surgeons (RCS) rats. METHODS: Twelve non-pigmented degenerative RCS rats at age of 15, 20, 25, 30 days were sacrificed after ERG examination. The caspase-9 expression was analyzed with immunohistochemical staining, fluoro-metric activity assay and Western blotting. Apoptotic retinal neurons were detected by terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling (TUNEL) technique. RESULTS: The latency and amplitude of b-wave were (58.60±3.42) ms and (109.07±14.50) mV, (56.45±1.08) ms and (109.00±7.35) mV, respectively, in 15 and 20 days old degenerative RCS rats. The ERG resembled flat form in the degenerative RCS rats at age of 25 and 30 days. TUNEL-positive cells were located only in the outer nuclear layer, mostly prevailed in 25-day-old group. Caspase-9 protein expressed in the rat retina started to be found immunohistochemically in 25-day-old and was most predominant in the inner segment. Fluorometric activity assay showed that caspase-9 activity reached its peak, (10.98±0.13) nmol·g-1·min-1 at 20th day. Western blot assay revealed the small cleavage fragmentation at 20th day. No cleavage fragmentation was revealed in the 15-day-old degenerative RCS rats and all other control rats. CONCLUSION: The activation and time-spatiotemporal expression of caspase-9 are coincidence with photoreceptor apoptosis as well as the loss of vision function in RCS degenerative rats, which implicates the important role of caspase-9 in photoreceptor apoptosis.  相似文献

AIM: To screen the cytotoxicity of degradable calcium metaphosphate (dCMP) compared with hydroxyapatite (HA). The proliferation and differentiation abilities of human marrow mesenchymal stem cells (MSC) were used to exhibit the cytotoxicity. METHODS: The cell morphology of MSC was analysed after direct contact with dCMP at different time points by scanning electron microscopy analysis. The degradation products of dCMP and HA were analysed with inductively coupled plasma torch and ion chromatography. The cytotoxic effect of degradation products of dCMP was evaluated by FACS, quantitative assay of ALP and ARS, respectively. RESULTS: dCMP enhanced the proliferation of MSC, but didn't interfere the osteogenic differentiation process of MSC and its mineralization. HA inhibited the proliferation of MSC and the mineralization of osteogenic differentiated MSC, while it did not interfere the osteogenic differentiation process of MSC. CONCLUSION: dCMP had a better cytocompatibility with MSC than HA, which might allow for its use as skeleton scaffolds.  相似文献

本试验旨在研究日粮色氨酸水平对生长猪的氮利用率以及对血浆类胰岛素生长因子-I(IGF-I)、生长激素(GH)、胰岛素浓度的影响。试验以4头大白去势公猪作为试验动物,以色氨酸含量分别为1.2、1.7、2.2和2.7g/kg的4种日粮为试验处理,按照4×4拉丁方进行试验设计。结果表明,色氨酸水平对干物质消化率、有机物消化率和氮消化率没有显著性影响(P>0.05)。氮沉积和氮的生物学利用率随着日粮色氨酸水平由1.2提高到1.7、2.2和2.7g/kg而显著提高(P<0.05),但是试验处理1.7、2.2和2.7g/kg之间的氮沉积和氮的生物学价值没有显著差异(P>0.05)。不同处理之间的血浆IGF-I、GH和胰岛素浓度没有显著差异(P>0.05)。日粮色氨酸水平和血浆IGF-I、GH或胰岛素浓度之间以及氮沉积和血浆IGF-I、GH或胰岛素浓度之间均无显著的相关关系(P>0.05)。综合以上结果,日粮色氨酸水平对生长猪的氮沉积具有显著影响,而对血浆IGF-I、GH和胰岛素没有显著影响。  相似文献

鱼精蛋白(protam ine)又称精蛋白,是一种主要存在于各类雄性动物成熟精巢组织中的多聚阳离子肽,它与DNA紧密结合在一起,以核精蛋白的形式存在。鱼精蛋白是一种小而简单的球形碱性蛋白,一般由30~50个氨基酸组成,其中以精氨酸为主。目前,已从多种鱼类(如鲑鱼、红鳟鱼、鲱鱼等)和哺乳动物(如老鼠、人、野猪等)的成熟精巢组织中提取到了鱼精蛋白。根据氨基酸的种类和数量,鱼精蛋白可分为3种:①单鱼精蛋白,它的碱性氨基酸仅为精氨酸;②双鱼精蛋白,碱性氨基酸除含有精氨酸外,还含有组氨酸或赖氨酸;③三精蛋白,碱性氨基酸有精氨酸、组氨酸和赖氨…  相似文献
