Members of heat shock protein 70 (HSP70) are highly conserved proteins of about 70 kDa and play important roles in protein folding. Levels of these proteins increase when cells are under stress. Environmental temperature influences both the basal and induced levels of HSPs. However, studies on HSPs in fishes from a tropical country such as India are lacking. In the present study, Indian major carp (IMC) Cirrhinus mrigala (Ham.) acclimatized at 25±2°C had high levels of HSP70, viz., 1.2–1.3 ng μg?1 total protein in kidney and gill and 4.2–5.3 ng μg?1 total protein in liver and brain tissues, indicating the presence of biochemically significant levels of stress. However, maintenance of acclimatized fish at 17°C for up to 48 h did not lead to a significant decrease in stress protein levels. A heat shock at 37°C for up to 48 h resulted in only two to threefold increase in HSP70 levels in these organs. Although the increase in HSP70 levels was apparent from the first hour of heat stress in all these tissues, the increase was significant from the second hour in the brain, the sixth hour in liver and kidney and the 20th hour in the gills. 相似文献
Impact of aeration on growth of silver barb, Puntius gonionotus during fingerling rearing was studied through a 100‐d rearing experiment conducted in 18 concrete tanks of 50 m2 (10 × 5 × 1.2 m) size. Fry (0.74 ± 0.27 g, 35 ± 6 mm) were stocked in the experimental tanks at three stocking densities (25, 50, and 75 fry/m2) and were evaluated with and without provision of 6 h (2400–0600 h) of night time aeration. Aeration resulted in higher pH and dissolved oxygen regime and increased fingerling length and weight. The results suggest a rearing density of 75/m2 to be ideal for rearing fry to fingerling of this species when aeration is provided, whereas, under non‐aerated condition, rearing the fry to fingerling stage at 50/m2 was found advantageous over those at 25 and 75/m2. 相似文献
Neonatal calf mortality is a major concern to livestock sector worldwide. Neonatal calf diarrhoea (NCD), an acute severe condition causes morbidity and mortality in calves. Amongst various pathogens involved in NCD, E. coli is considered as one of the major causes. The study was targeted to characterize E. coli isolates from neonatal calves for diarrhoeagenic Escherichia coli (DEC) types (pathotyping), antimicrobial resistance (AMR) profiling and to correlate with epidemiological parameters. From neonates, a total of 113 faecal samples were collected, out of that 308, lactose fermenting colonies were confirmed as E. coli. Pathotypable isolates (12.3%) were represented by STEC (6.1%), EPEC (2.9%), ETEC (1.9%), EAEC (0.9%) and EHEC (0.3%). Occurrence of STEC was more in non-diarrhoeic calves, whereas ETEC was observed more in diarrhoeic calves. EPEC occurrence was observed in both diarrhoeic and non-diarrhoeic calves. Fishers extract test showed no significant association for occurrence of DEC types to type of dairies, health status, species, breed, age and sex of neonatal calves. Two hundred and eighty isolates were tested for antimicrobial susceptibility. The isolates showed maximum resistance towards ampicillin (55.4%) followed by tetracycline (54.3%), while minimum resistance was observed towards meropenem (2.5%). Multidrug resistant E. coli isolates were found to be 139 (49.6%), and Extended-spectrum beta-lactamase (ESBL) producers were 120 (42.9%). DEC pathotypes like STEC, ETEC, EHEC and EAEC that are also multidrug resistant present in neonatal calves have zoonotic potential and hence are of public health significance.
We studied leaf litter fall, decomposition and nutrient release patterns of Shorea robusta and Tectona grandis by using a litter bag technique to better understand the release pattern of nutrients to soil from leaf litter. Annual litterfall varied from 13.40 ± 2.56 t ha?1 a?1 for S. robusta to 11.03 ± 3.72 t ha?1 a?1 for T. grandis and the decay constant (k) of decomposed leaf litter was distinctly higher for T. grandis (2.70 ± 0.50 a?1) compared to S. robusta (2.41 ± 0.30 a?1). Biomass loss was positively correlated with the initial litter C, WSC, C/N and ash content in S. robusta and N, P and K concentration for T. grandis. Biomass was negatively correlated with lignin and L/N ratio for S. robusta and L, WSC, L/N and C/N ratio for T. grandis (P < 0.01). Nutrient use efficiency (NUE) and nutrient accumulation index (NAI) of S. robusta was higher than for T. grandis. The retranslocation of bioelements from senescent leaves ranked as P > N > K. Annual N, P and K input to soil through litterfall differed significantly between the two species in the following order: N>K>P. S. robusta was superior in terms of K and P return and T. grandis was superior in terms of N return. The two tree species showed a similar patterns of nutrient release (K > P > N) during decomposition of their leaf litter. In India, mangroves occupy 6740 km2 of which the Mahanadi delta andBhitarkanika mangrove forests of Orissa cover 191.44 km2, most of whichhas been severely overexploited. World wide concern to conserve mangrovesnecessitated propagation of mangroves to re-establish them on barren andswampy land along tidal creeks around Paradeep port of Orissa. Avicenniaofficinalis, Aegiceras corniculatum, Bruguiera parviflora, B. sexangula,Ceriops decandra, Excoecaria agallocha, Heritiera fomes, Kandelia candel,Rhizophora mucronata, Sonneratia apetala were vegetatively propagated andplanted over 10 ha of degraded salt-marshy wetlands of Mahanadi delta inpure and mixed stands depending on the intensity and the frequency of tidalinundation at the experimental site. Survival was significantly higher(80%) in S. apetala followed by A. officinalis, R. mucronata, K.candel and H. fomes (70–75%). S. apetala recorded the maximumgrowth in height (3.0 m after 2 years) whereas C. decandra showed theminimum growth (0.5 m). Growth performance was better with plants in mixedstands than the plantation with single species. This gives us hope topropagate and re-establish mangroves for conservation in scientificallymanaged plantations in a physiologically arid environment. 相似文献
Fifteen Bhadawari buffalo heifers of 207?±?9.78 kg mean body weight were randomly distributed into three dietary groups to evaluate the effect of protein level on nutrient utilization, nitrogen (N) balance, growth rate, blood metabolites, and puberty. All animals were offered wheat straw-berseem diets supplemented with concentrate mixtures of similar energy (2.7 Mcal/kg) and different protein levels (14.3–22 %). Animals of standard-protein group (SPG) were offered protein and energy as per requirement, while animals of low-protein group (LPG) and high-protein group (HPG) were fed 20 % less and 20 % more protein, respectively, than SPG. Feed dry matter (DM) and metabolizable energy (ME) intake (% body wt. and g/kg w0.75) were similar for all three diets; however, the crude protein (CP) and digestible crude protein (DCP) intake on percent body weight and per kilogram metabolic weight was higher (P?<?0.05) in HPG than in SPG or LPG. Digestibility of CP, cellulose, and hemicellulose was higher (P?<?0.05) in HPG versus LPG. Fecal N excretion was similar, while urinary N excretion was highest (P?<?0.05) in HPG (74.83 g/day) compared with SPG (50.03 g/day) and LPG (47.88 g/day), which resulted in lower N retention in HPG than in the other dietary groups. Level of dietary N had no effect on blood metabolites viz. glucose, urea, and N. Digestible energy (DE) and ME contents of diets were identical, while DCP contents were higher (P?<?0.05) in HPG than in LPG. Feed and nutrient (CP and ME) conversion efficiency to produce a unit kilogram weight gain was identical among the dietary groups. Dietary protein level had no effect on the heifer’s weight and age at puberty. The mean growth rate of heifers at 240 days was higher (P?>?0.05) in SPG (330.8 g/day) than in LPG (296.7 g/day), while the animals gained more weight in January to March months and the lowest weight in May to July months. Protein level had no effect on conception rate of heifers. Results revealed that 20 % higher or less protein than the ICAR requirement had no significant (P?>?0.05) on feed intake, nutrient conversion efficiency for weight gain, heifer growth, and puberty; however, 20 % more protein increased urinary N loss.
This experiment was conducted to study the effect of different levels of wheat roti (WR) on nutrient utilization and blood metabolites in Asian elephants fed roughages ad libitum. Nine (3 M, 6 F) Asian elephants (14–52 years of age, 1909–3968 kg BW) were used in an experiment based on replicated Latin square design. Animals in each group (n = 3) were assigned to one of the three dietary treatments in a manner that animals in all the three groups were exposed to all the three treatments in three different periods. Each feeding trial comprised 30 days (25 days of adaptation and 5 days collection period). The amount of WR fed to the elephants was 0.18, 0.12 and 0.06% of BW in groups I, II and III, respectively. They were allowed to forage in the nearby forests for 6 h/day and to bathe for 2 h/day. The animals had ad libitum access to cut Rohini (Mallotus philippensis) trees in their night shelter. Intake and apparent digestibility of dry matter (DM), crude protein (CP), gross energy (GE), Ca, P, Fe, Cu and Zn were measured. Feed consumption was not significantly different among the groups. Significant (p < 0.01) decrease in digestibility of DM and GE and blood glucose concentration was observed with decreased level of WR in the diet. Feeding of WR at 0.06% of BW supplied adequate amount of DE, CP, Ca, P, Fe, Cu and Zn to meet requirement for adult maintenance. Feeding of WR in excess of 0.06% of BW supplied DE in excess of requirement, increased blood glucose concentration which may cause obesity and other associated health problems. It was concluded that the amount of WR should be restricted to 0.06% of BW in the diet of captive Asian elephants. 相似文献
The growth and survival of kalbasu, Labeo calbasu, was evaluated at stocking densities of 5, 10 and 15 million spawn ha−1 in nursery rearing in concrete tanks of 50 m2, each for a period of 25 days. Survival of fry was density dependent and significantly higher (54.5%) at 5 million ha−1 than those at 10 million ha−1 (50.1%) and 15 million ha−1 (46.9%). Similarly, growth and specific growth rate were inversely related to the stocking density and varied significantly among the three densities. 相似文献
A new mannose-binding lectin was purified from plasma of freshwater fish, rohu (Labeo rohita) by ammonium sulphate precipitation followed by DEAE-cellulose ion-exchange chromatography. The hemagglutinating activity
is strong for neuraminidase-treated rabbit erythrocytes. Mannose, glucose and their derivatives inhibit hemagglutination.
The apparent molecular weight was determined to be 210 kDa in native PAGE. Analysed on SDS-PAGE under reducing and non-reducing
conditions, the protein migrated as a single band of mol.wt. 40,000. The lectin is acidic in nature showing a pI of 4.5. It
is a glycoprotein containing complex glycan part as indicated by carbohydrate analysis using high-pressure anion exchange
chromatography with a pulsed amperometric detector. The N-terminal sequence (WLNGIGTQIPMKITT) shows no significant homology
with known proteins. It appears to be a C-type lectin as Ca2+ is essential for carbohydrate-binding activity. Methyl -α- D-mannopyranoside was found to be the most potent inhibitor among
the monosaccharides and disaccharides tested. Purified lectin was found to induce intracellular superoxide production following
opsonization of E. coli by its own macrophages.
