Best Linear Unbiased Prediction (BLUP) has become the most widely used method for genetic assessment of animal and perennial species, and it is potentially relevant for annual crops. The objective of this study was to assess this method for selection within non-inbred families in recurrent breeding programs. The ‘animal model’ was fitted. The data were expansion volume (EV) and grain yield of plants in recombination plots of two to three selection cycles in the popcorn population Viçosa, with half- and full-sib progenies. The ASReml program was used to perform the analyses. For both EV and yield the breeding values predicted from the additive and additive-dominant models were highly correlated. Multi-generation BLUP was, in general, more accurate than single-generation analysis. These two methods resulted in highly correlated predicted breeding values. The dominance genetic values predicted from the single- and multi-generation analysis were also highly correlated. The pedigree information reduced the percentage of coincidences among the selected individuals relative to phenotypic selection mainly in the population structured in half-sib families. Based on breeding values predicted by BLUP analysis, the most efficient selection procedure was mass selection.  相似文献

This study evaluates the treatment efficacy and biogas yield of an integrated system composed of a plug-flow biodigester (with sludge recirculation) followed by polishing in a stabilization pond. The system was operated in real scale for 12 months at ambient temperature and under continuous flow. The volumetric yields of biogas varied according to the organic loads applied, between 114 and 294 Kg COD day?1, reaching levels of 0.026 to 0.173 m3 m?3 day?1, with concentrations of CH4 between 56 and 70%. The monthly biogas productions were between 378.5 and 2186.1 m3 month?1 equal to an energy potential of approximately 2070 to 19,168 KWh month?1.The average yearly removals of BOD5,20 and COD by the integrated treatment system were 70 and 86%, respectively. The average annual removals of NH4 and TKN were 88.5 and 85.5%, respectively. The pH values were always near neutral, and the alkalinity was in ranges propitious for anaerobic digestion. The results of this study indicate good efficacy in terms of removal of organic matter and nitrogen compounds, with the added benefits of generation of energy and use of the treated effluent as biofertilizer, enabling significant cost reductions to cattle farmers.  相似文献

ABSTRACT Lettuce mosaic virus (LMV) causes an economically important seedborne and aphid-transmitted disease of lettuce and ornamental crops worldwide. The genetic diversity among 73 LMV isolates was examined based on a 216-nucleotide sequence at the variable region encoding the NIb-coat protein junction. Three clusters of LMV isolates were distinguished: LMV-Yar, LMV-Greek, and LMV-RoW. In the latter cluster, two subgroups of isolates, LMV-Common and LMV-Most, accounted for a large proportion of the LMV isolates analyzed. These two subgroups included the seedborne isolates, consistent with this property contributing a selective advantage and resulting in widespread distribution. In addition to being seedborne, LMV-Most isolates overcome the two resistance genes commonly used in lettuce, mo1(1) and mo1(2), and thus represent a potential threat to lettuce cultivation. The complete sequence of an LMV-Most isolate (LMV-AF199) was determined, allowing a better definition of the genetic relationships among LMV-Most, LMV-Common, and an additional isolate of the LMV-RoW cluster.  相似文献

The tick genus Haemaphysalis is represented by four species in the New World, of which only the species Haemaphysalis leporispalustris has been associated with Rickettsiae. The present study reports for the first time the presence of a Rickettsia strain in the tick Haemaphysalis juxtakochi. A free-living male of H. juxtakochi, collected in the state of Rondonia, Western Amazon, Brazil, was subjected to DNA extraction and tested by PCR targeting the four rickettsial genes: gltA, 17-kDa, ompA and ompB. The nucleotide sequences obtained from the PCR products were, by BLAST analyses, closest to Rickettsia rhipicephali sharing 99.7% (1147/1150), 98.8% (429/434), 99.0% (486/491) and 99.0% (809/817) identities with the partial sequences of the gltA, 17-kDa, ompA and ompB genes, respectively. Phylogenetic analyses inferred from the four rickettsial genes showed a high-degree of similarity of this H. juxtakochi-Rickettsia with R. rhipicephali. These two agents grouped together in all trees, always with high bootstrap support (75-96%). This study gives molecular evidence for the presence of a Rickettsia species, designated as strain R300, in the tick H. juxtakochi from the Western Amazon area of Brazil. Genetic analyses showed R300 to be closely related to R. rhipicephali.  相似文献

Streptococcus suis serotype 2 is a swine pathogen responsible for diverse diseases and may be present in the tonsils of pigs which show no sign of illness. Because adhesion to host cells may be important in the carrier state, this study was undertaken to characterize a 39 kDa surface protein identified as a glyceraldehyde-3-phosphate dehydrogenase (GAPDH), possibly implicated in the adhesion of the bacteria. The gene encoding for the GAPDH of S. suis was cloned and sequenced. The DNA sequence contained an open reading frame encoding for a 336 amino acid polypeptide exhibiting 95% sequence identity with the GAPDH from Streptococcus pyogenes and from other streptococci. Using the Qiaexpress expression plasmids, the gapdh gene was inducibly overexpressed in E. coli to produce GAPDH with a hexahistidyl N-terminus to permit its purification. The (His)6GAPDH protein was found to possess functional GAPDH enzymatic activity after the purification. An adherence assay with S. suis and porcine tracheal rings pre-incubated with (His)6GAPDH and non-incubated rings was showed a significant reduction in the adhesion of S. suis in the (His)6GAPDH pre-incubated rings compared to the non-incubated rings. The GAPDH protein of S. suis seems to be involved in the first steps of the bacterial adhesion to host cells.  相似文献

Three experiments were conducted to investigate the ischiorectal fossa (IRF) as a route for the administration of prostaglandin F2 alpha (dinoprost) in cattle. In Experiment 1, 21 nonlactating Holstein cows were given 100 micrograms of gonadotropin releasing hormone (GnRH), intramuscularly (i.m.), and, 7 d later, 25 mg of dinoprost into the IRF. Sixteen cows had serum progesterone concentrations > or = 1.0 ng/mL at the time of dinoprost treatment, and all of these had rapid and complete luteolysis; the other 5 cows were not considered to have a functional corpus luteum (CL) at the time of treatment. There were minimal adverse behavioral reactions to the IRF injections and no visible or palpable tissue reactions at the injection site. In Experiment 2, 74 Holstein heifers were given 25 mg of dinoprost by IRF injection. Luteolysis occurred in 84.3% of the heifers with a functional CL (as determined by the serum progesterone concentration at the time of treatment). Of the heifers bred by either natural service or artificial insemination, 61.8% became pregnant. In Experiment 3, 48 beef heifers received dinoprost 7 d after ovulation, as follows: 25 mg, i.m. (n = 9); 25 mg, IRF (n = 10); 10 mg, IRF (n = 10); 10 mg, subcutaneously (s.c.) (n = 10); or 10 mg, intravulvosubmucosally (IVSM) (n = 9). Fewer heifers (P < 0.05) were found to be in estrus or ovulating in the 10 mg IVSM group (0% and 11%, respectively) than in the 25 mg i.m. group (100% and 100%), the 25 mg IRF group (90% and 100%, respectively), or the 10 mg IRF group (80% and 80%). The rates of estrus (50%) and ovulation (50%) were intermediate in the 10 mg s.c. group. In summary, 25 mg of dinoprost injected into the IRF caused minimal behavioral or tissue response and induced luteolysis and fertile estrus. In addition, 10 mg of dinoprost injected into the IRF was as efficacious as 25 mg given either i.m. or into the IRF in inducing estrus and ovulation.  相似文献
