Biosolids compost is a good organic amendment but immature compost can exhibit phytotoxic behavior which can be attributed to different toxic substances. Our objective was to determine the phytotoxicity of: i) Biosolids; ii) Mix of biosolids and wood sawdust sampled a day after composting started; iii) The same material sampled at the end of the thermophilic stage; iv) cured compost; v) cow manure and vi) horse manure A germination bioassay was carried out using Lolium perenne (ryegrass) seeds: germination and root growth percentage were determined as well as electrical conductivity, pH, phenol content and volatile organic acids. In three treatments, Ni, Pb, Zn, Cu and Cd were also determined. Ammonia volatilization was determined during biosolids composting. The germination percentage varied from 67% to 95% but the inhibition of root growth appears to be a more sensitive phytotoxicity indicator (18% to 74%. Phytotoxic effects on germinating ryegrass were mainly related to extract pH and electrical conductivity. Potentially toxic elements, volatile organic acids, phenolic compounds and ammonia were not related to germination.  相似文献

ABSTRACT

Smoking is a technique used to improve the sensory attributes and increase the shelf life of fish. The objective of the present study was to evaluate the nutritional, physicochemical, microbiological, and sensory characteristics of sausages made with sea catfish (Sciades herzbergii) fillets subjected to the traditional and liquid smoking processes. The traditionally smoked sausages showed lower (p < .05) moisture content and higher protein, fat, and mineral levels, higher weight loss during processing, higher pH, higher a* (red intensity), and lower L* (luminosity) values. The percentage of shrinkage, water holding capacity, water activity, hardness, cohesiveness, and elasticity were not influenced (p > .05) by the smoking methods tested. The microbial counts of both treatments were in compliance with the current Brazilian legislation. The sensory acceptance of the liquid smoked sausages was higher (p < .05) when compared to the traditionally smoked sausages for the attributes of color, aroma, texture, flavor, and overall acceptance, with scores ranging from 6 (liked slightly) to 7 (liked moderately). Between the two smoking methods tested, liquid smoking presented better sensory acceptance, easier execution, greater process control, and less environmental impact; thus, it is a viable alternative to the traditional smoking of sea catfish (Sciades herzbergii) sausages.  相似文献

Summary

Until recently, olive breeding programmes have been based exclusively on cultivated olive germplasm (Olea europaea subsp. europaea var. europaea), in contrast to breeding in other fruit crops where the use of wild relatives has been widely reported. In this study, ten agronomic traits were evaluated in two progenies derived from controlled crosses between the Spanish olive cultivar ‘Picual’ (female) and two wild (O. europaea subsp. europaea var. sylvestris) genotypes (males). The results of this evaluation were compared with the progenies of crosses between ‘Picual’ (female) and the three cultivars, ‘Sikitita’, ‘Meski’, and ‘Zaity’ (male). The two ‘Picual’ × wild genotype progenies had the highest mean values for vigour traits (i.e., tree height and trunk diameter). Progenies from both ‘Picual’ × wild genotype crossess also showed the highest percentages of trees (53.8% and 37.5%) with a short juvenile period, compared to the progenies from crosses between ‘Picual’ and each of the three cultivars ‘Sikitita’, ‘Meski’, and ‘Zaity’ (0%, 5%, and 3.6%, respectively). Significant differences were also observed between progenies in fruit traits as well as in oil contents and fruit weights. Progenies from both crosses with wild olive showed lower values for the fruit traits evaluated than the cultivated olive progenies. However, significant improvements were achieved compared to fruit traits in the wild parents. The implications of these results for the future use of wild germplasm in olive breeding programmes are discussed.  相似文献

This work aims to identify and characterize heavy metal contamination in a fluvial system from Cartagena–La Unión mining district (SE Spain). In order to assess the dynamics of transport and the accumulation of heavy metals, sediments, surface water and vegetation, samples along “El Avenque” stream were collected. The former direct dumps of wastes and the presence of tailing ponds adjacent to the watercourse have contributed to the total contamination of the stream. Total Cd (103 mg kg−1), Cu (259 mg kg−1), Pb (26,786 mg kg−1) and Zn (9,312 mg kg−1) in sediments were above the limits of European legislation, being highest where tailing ponds are located. Bioavailable metals were high (3.55 mg Cd kg−1, 6.45 mg Cu kg−1, 4,200 mg Pb kg−1 and 343 mg Zn kg−1) and followed the same trend than total contents. Metals in water were higher in sampling points close to ponds, exceeding World Health Organization guidelines for water quality. There is a direct effect of solubilisation of sediment metals in water with high contents of SO42−, product of the oxidation of original sulphides. The mobility of metals varied significantly with shifts in pH. Downstream, available and soluble metals concentrations decreased mainly due to precipitation by increments in pH. As a general pattern, no metal was bioaccumulated by any tested plant. Thus, native vegetation has adopted physiological mechanisms not to accumulate metals. This information allows the understanding of the effect of mining activities on stream contamination, enforcing the immediate intervention to reduce risks related to metals’ mobility.  相似文献

Despite the low timber productivity of Mediterranean Pinus halepensis Mill. forests in south-eastern Spain, they are a valuable carbon sequestration source which could be extended if young stands and understories were considered. We monitored changes in biomass storage of young Aleppo pine stands naturally regenerated after wildfires, with a diachronic approach from 5 to 16 years old, including pine and understory strata, at two different quality sites (dry and semiarid climates). At each site, we set 21 permanent plots and carried out different thinning intensities at two ages, 5 and 10 years after fires. We found similar post-fire regeneration capacity at both sites in terms of total above-ground biomass storage ~6 Mg ha?1 (3 Mg ha?1 of the above-ground pine biomass plus 3 Mg ha?1 of the above-ground understory biomass), but with a contrasting pine layer structure. Generally, across the diachronic study, the earlier thinning reduced biomass stocks at both sites, except for the best quality site (the dry site), where the earliest thinning (applied at post-fire year 5) enlarged carbon storage by 11 % as compared to non-thinned plots. We found root:shoot ratios of an average 0.37 for the pine layer and 0.45 for the understory layer. These results provided new information which not only furthers our understanding of carbon sequestration in low timber productivity Mediterranean forests, but will also help to develop new guidelines for sustainable management adapted to the high-risk terrestrial carbon losses of fire-prone areas.  相似文献

The proteolytic processing of amyloid precursor protein (APP) by β-secretase (BACE1) and γ-secretase releases amyloid-β peptide (Aβ), which deposits in amyloid plaques and contributes to the initial causative events of Alzheimer’s disease (AD). In the present study, the regulatory mechanism of APP processing of three phlorotannins was elucidated in Swedish mutant APP overexpressed N2a (SweAPP N2a) cells. Among the tested compounds, dieckol exhibited the highest inhibitory effect on both intra- and extracellular Aβ accumulation. In addition, dieckol regulated the APP processing enzymes, such as α-secretase (ADAM10), β-secretase, and γ-secretase, presenilin-1 (PS1), and their proteolytic products, sAPPα and sAPPβ, implying that the compound acts on both the amyloidogenic and non-amyloidogenic pathways. In addition, dieckol increased the phosphorylation of protein kinase B (Akt) at Ser473 and GSK-3β at Ser9, suggesting dieckol induced the activation of Akt, which phosphorylated GSK-3β. The specific phosphatidylinositol 3-kinase (PI3K) inhibitor LY294002 triggered GSK-3β activation and Aβ expression. In addition, co-treatment with LY294002 noticeably blocked the effect of dieckol on Aβ production, demonstrating that dieckol promoted the PI3K/Akt signaling pathway, which in turn inactivated GSK-3β, resulting in the reduction in Aβ levels.  相似文献

Significant amounts of enolase have recently been found in the saliva of the argasid tick Ornithodoros moubata, raising the question as to what the function of enolase in the tick–host interface is. Enolase is a multifunctional glycolytic enzyme known to act as a plasminogen receptor on cellular surfaces, promoting fibrinolysis and extracellular matrix degradation. Fibrinolysis could be important for ticks to dissolve clots that might be formed during feeding as well as to prevent clotting of the ingested blood meal in the tick midgut. Additionally, enolase-mediated extracellular matrix degradation could contribute to the tick feeding lesion. Moreover, previous observations suggested an additional antihaemostatic role for O. moubata enolase as a P-selectin antagonist ligand. Accordingly, the aim of the present study was to investigate the potential role of the O. moubata salivary enolase as a plasminogen receptor and P-selectin ligand, and to evaluate its potential as an antigen target for anti-O. moubata vaccines. The study included the cloning, sequencing and recombinant production of the O. moubata enolase, plasminogen binding and activation assays, P-selectin binding assays, animal immunization trials, and RNAi knockdown of the enolase gene. Here we confirmed that enolase is secreted to the saliva of the tick and provide convincing evidence for a role of this salivary enolase as a plasminogen receptor, most likely stimulating host fibrinolysis and maintaining blood fluidity during tick feeding. The RNAi experiments and immunization trials indicated that enolase could be also involved in the regulation of tick reproduction, suggesting new potential control strategies. Finally, the P-selectin binding experiments demonstrated that this enolase is not a P-selectin ligand.  相似文献

Abstract

The infectious pancreatic necrosis virus (IPNV; genus Aquabirnavirus) affects salmon and trout, causing high mortality in first-feeding fry. The classification of this virus includes nine serotypes and seven genogroups. In Mexico, two different isolates were identified in 2000 and 2008, respectively. Both isolates were classified into genogroup I according to the RNA genome of this virus. As Mexico is importing rainbow trout Oncorhynchus mykiss eggs from different countries, the aim of this study was to genotype IPNV isolates obtained from four rainbow trout producer regions within the state of Mexico. We utilized a fragment of the VP2* (outer capsid protein) gene sequence of Mexican IPNV isolates as a molecular marker to determine the genogroup to which they belong. Although all Mexican IPNV isolates were grouped into genogroup I, we identified genetic diversity among these isolates, and 14 unique nucleotide sequence types were associated with the four producer regions in Mexico State.

Two experiments were conducted to determine the dietary value of pellets containing kenaf (Hibiscus cannabinus cv. 'Everglade 41') hay. Averaged across both experiments, kenaf pellets contained 82.6% kenaf hay, 16.6% liquid molasses, and 0.8% mineral oil. The chemical composition of the kenaf pellet was 12.6% crude protein (CP), 41.2% neutral detergent fiber (NDF), and 14.4% acid detergent fiber (ADF). In Exp. 1 (digestion and N balance trial), 18 lambs (body weight [BW] = 36.4 kg) were blocked by BW. Lambs were randomly assigned within a block to Diet 1 (59.5% corn and 40.5% alfalfa pellet), Diet 2 (59.7% corn, 28.4% alfalfa pellets, and 11.9% kenaf pellets), or Diet 3 (59.6% corn, 16.5% alfalfa pellets, and 23.9% kenaf pellets). Diets were formulated so that CP was the first-limiting nutrient. Each diet was limit-fed at 2.4% of BW. Replacing alfalfa pellets with kenaf pellets tended to decrease (P = 0.10) CP and ADF intakes, but increased (P = 0.01) DM digestibility. Diet had no effect (P = 0.33) on N balance. In Exp. 2 (dry matter [DM] intake trial), 32 lambs (BW = 30.4 kg) were blocked by gender and BW. Within a block, lambs were randomly assigned to one of four diets in a 2 x 2 factorial arrangement. Main effects were hay (bermudagrass or fescue) and supplemental protein source (kenaf or alfalfa pellets). Lambs were housed in individual pens with ad libitum access to the assigned hay. Supplemental protein was fed (185 g of DM) once daily. Hay intake was measured weekly for 8 wk. Lambs consumed more (P = 0.002) fescue than bermudagrass hay (743 vs 621 g/ d). Lambs fed fescue hay gained weight more rapidly (P = 0.001) than lambs fed bermudagrass hay (120 vs 72 g/d). Hay intake and ADG were similar (P = 0.90) for lambs fed alfalfa or kenaf pellets. Kenaf hay mixed with molasses and mineral oil can be formed into a pellet. In the diets used in this experiments, kenaf pellets can replace alfalfa pellets in diets fed to lambs without altering forage intake, gain, or N retention.  相似文献
