High-grade glioma is an aggressive cancer that occurs naturally in pet dogs. Canine high-grade glioma (cHGG) is treated with radiation, chemotherapy or surgery, but has no curative treatment. Within the past eight years, there have been advances in our imaging and histopathology standards as well as genetic charactereization of cHGG. However, there are only three cHGG cell lines publicly available, all of which were derived from astrocytoma and established using methods involving expansion of tumour cells in vitro on plastic dishes. In order to provide more clinically relevant cell lines for studying cHGG in vitro, the goal of this study was to establish cHGG patient-derived lines, whereby cancer cells are expanded in vivo by injecting cells into immunocompromized laboratory mice. The cells are then harvested from mice and used for in vitro studies. This method is the standard in the human field and has been shown to minimize the acquisition of genetic alterations and gene expression changes from the original tumour. Through a multi-institutional collaboration, we describe our methods for establishing two novel cHGG patient-derived lines, Boo-HA and Mo-HO, from a high-grade astrocytoma and a high-grade oligodendroglioma, respectively. We compare our novel lines to G06-A, J3T-Bg, and SDT-3G (traditional cHGG cell lines) in terms of proliferation and sensitivity to radiation. We also perform whole genome sequencing and identify an NF1 truncating mutation in Mo-HO. We report the characterization and availability of these novel patient-derived lines for use by the veterinary community.  相似文献

A 4-year-old, neutered male, British shorthair cat was presented with inappetence, vomiting, hyperproteinemia, and hyperglobulinemia. An exploratory celiotomy identified enlarged mesenteric lymph nodes. Immunohistochemistry of lymph node biopsies confirmed feline infectious peritonitis. This study compared incisional complications after ventral midline laparotomy using 2 absorbable suture materials for apposition of the linea alba in sheep. The linea alba of 93 yearling sheep was sutured by 3 veterinarians in a simple continuous pattern using either polyglactin 910 (PG910; group PG) or polydioxanone (PDS; group PD). A blinded observer assessed surgical sites at the time of suture removal. Multivariate logistic regression was used to assess the association between incisional complications and variables (suture material used, veterinarian, skin suture removal time). The odds of incisional complications did not vary significantly with the type of suture material used (P = 0.11), veterinarian (P = 0.61) or skin suture removal time (P = 0.36). Most incisional complications were cutaneous suture sinus formation. Either PG910 or PDS may be used for linea alba closure in sheep.  相似文献

Following the severe drought in 1999–2000 there was a widespread outbreak of oak decline in the Ozark Highlands. Over 400,000 ha of dead and dying oak trees were observed by the USDA Forest Service in this region. Although oak forests that are dead can be easily interpreted from air photos or classified from satellite images, it is difficult to detect dying trees that are still green but will die back or recover in the following years. In this study, we applied a normalized difference water index (NDWI) to map the continuous forest dynamics related to oak decline. The Landsat TM image in 1992 and the ETM+ image in 2000 were processed to calculate the differential NDWI which revealed moisture variation primarily caused by the drought and the associated red oak borers. A simple thresholding method was used to map oak dying back, recovery and non-change areas in the study area. The died-back areas were extracted from the modified land use/land cover maps created by the Missouri Resource Assessment Partnership (MoRAP). The forest dynamics map was compared with the online FIA database in which tree species at randomly selected sites were recorded in 1989 and 2003. The overall accuracy of forest dynamics mapping with remote sensing imagery was 75.95%. The user's accuracy of dying/recovery area mapping was also high although the producer's accuracy is questionable because of the limitation in ground data collection. The continuous dying/recovery map in this study could provide valuable information on the prediction of oak decline and evaluation of damage when another period of environmental stresses occurs.  相似文献

A receptor-like protein kinase gene (Ppsrkl1) was isolated from a peach (Prunus persica (L.) Batsch.) bark cDNA library prepared with RNAs isolated from bark collected in December (cold acclimated). Sequence analysis indicated that this gene is related to the S-locus family of receptor protein kinases (SRKs) and that it shares greatest homology with ZMPK1 from maize and At4g32300 from Arabidopsis, both of which are intron-less genes. In bark tissues, Ppsrkl1 is induced by water deficit treatment, repressed by short-day photoperiods and showed no response to cold treatment. The Ppsrkl1 mRNA also increased in roots in response to water deficit. In fruit, Ppsrkl1 shows no response up to 6 h after wounding, but at 12 and 24 h after wounding, Ppsrkl1 mRNA shows an abrupt decline. This decline was prevented by the addition of salicylic acid to the wound site. The Ppsrkl1 mRNA rapidly decreased in fruit after 10-min exposure to UV-C radiation, followed by a return to normal levels within 1.5 h. Taken together, these experiments indicate that Ppsrkl1 is negatively regulated by light and positively influenced by salicylic acid treatment in fruit and water stress in bark and roots.  相似文献

We used three methods to measure boundary layer conductance to heat transfer (g(bH)) and water vapor transfer (g(bV)) in foliated branches of Abies amabilis Dougl. ex J. Forbes, a subalpine forest tree that produces clumped shoot morphology on sun-formed branches. Boundary layer conductances estimated in the field from energy balance measurements increased linearly from approximately 10 mm s(-1) at low wind speeds (< 0.1 m s(-1)) to over 150 mm s(-1) at wind speeds of 2.0 m s(-1). Boundary layer conductances measured on shoot models in a wind tunnel were consistently higher than field measurements. The difference between wind tunnel values and field measurements was attributable to variation in path length between the two experimental environments. Boundary layer conductance estimated by subtracting stomatal resistance (r(sV)) measured with a porometer from the total branch vapor phase resistance were unusually small. Sensitivity analysis demonstrated that this method is not suitable for coniferous foliage or when stomatal conductance (g(sV)) is small compared with g(bV). Analysis of the relative magnitudes of g(sV) and g(bV) revealed that, under most conditions, A. amabilis branches are well coupled (i.e., g(sV) is the dominant controller of transpiration). The boundary layer conductance to heat transfer is small enough that leaf temperature can become substantially higher than air temperature when radiation is high and wind speed is low. Over a two-month period, the maximum difference between leaf and air temperatures exceeded 6 degrees C. Leaf temperature exceeded air temperature by more than 2 degrees C on 10% of the daylight hours during this period. Consideration of both the photosynthetic temperature response of A. amabilis foliage as well as the summer air temperature conditions in its habitat suggests that these elevated leaf temperatures do not have a significant impact on carbon gain during the growing season.  相似文献

Summary Winter-type experimental lines from crosses of two winter-type barley cultivars with a spring-type, barley yellow dwarf (BYD) tolerant cultivar were compared in inoculated and uninoculated hill plots. Mature plants traits-winter survival, height, number of spike-bearing tillers, and biomass-were examined. The experimental lines could not be compared with the BYD tolerant parent using these diagnostic symptoms since the spring-type parent would be winterkilled. Two criteria were used to rate experimental lines for BYD tolerance: (1) nonsignificant difference between BYD-inoculated and BYD-uninoculated treatments for all traits, and (2) a BYD injury score obtained by averaging percent reduction from BYD infection for each trait.Missouri Agricultural Experiment Station Journal Paper No. 10059. BACKGROUND: Aminocyclopyrachlor is a new herbicide proposed to control broadleaf weeds and shrubs in non‐crop and rangeland systems. To gain a better understanding of observed field efficacy, the uptake and translocation of foliar‐applied aminocyclopyrachlor (DPX‐MAT28) and aminocyclopyrachlor methyl ester (DPX‐KJM44) were evaluated in two annuals, prickly lettuce (Lactuca serriola L.) and yellow starthistle (Centaurea solstitialis L.), and one perennial, rush skeletonweed (Chondrilla juncea L.). RESULTS: Absorption and translocation varied between species. While absorption of DPX‐KJM44 was greater than absorption of DPX‐MAT28, rush skeletonweed absorbed the most, followed by yellow starthistle and prickly lettuce. Overall, the total translocation of either herbicide was highest in yellow starthistle, followed by rush skeletonweed and prickly lettuce. Proportional herbicide movement between species was similar, with the majority translocating to developing shoots. However, in rush skeletonweed, early translocation was directed to root tissue. In rush skeletonweed, no DPX‐MAT28 metabolism occurred, while DPX‐KJM44 was rapidly de‐esterified and translocated as DPX‐MAT28. CONCLUSION: Aminocyclopyrachlor absorption and translocation are dependent on active ingredient structure and species sensitivity. Highly sensitive species such as prickly lettuce absorb and translocate less material than relatively less sensitive species such as rush skeletonweed. De‐esterification of DPX‐KJM44 appears to delay translocation of the resulting acid in yellow starthistle and rush skeletonweed. Copyright © 2011 Society of Chemical Industry  相似文献

Pine nuts are a part of traditional cooking in many parts of the world and have seen a significant increase in availability/use in the United States over the past 10 years. The U.S. Food and Drug Administration (US FDA) field offices received 411 complaints from U.S. consumers over the past three years regarding taste disturbances following the consumption of pine nuts. Using analysis of fatty acids by gas chromatography with flame ionization detection, previous reports have implicated nuts from Pinus armandii (Armand Pine) as the causative species for similar taste disturbances. This method was found to provide insufficient species resolution to link FDA consumer complaint samples to a single species of pine, particularly when samples contained species mixtures of pine nuts. Here we describe a DNA based method for differentiating pine nut samples using the ycf1 chloroplast gene. Although the exact cause of pine nut associated dysgeusia is still not known, we found that 15 of 15 samples from consumer complaints contained at least some Pinus armandii, confirming the apparent association of this species with taste disturbances.  相似文献
