The pregnancy rates obtained after the transfer of cryopreserved in vitro‐produced (IVP) embryos are usually low and/or inconsistent. The objective of this study was to evaluate the pregnancy rates of Holstein, Gyr and Holstein × Gyr cattle after the transfer of vitrified IVP embryos produced with X‐sorted sperm. Seventy‐two Gyr and 703 Holstein females were subjected to ovum pickup (OPU) sessions, followed by in vitro embryo production using semen from sires of the same breeds. Embryos (1636 Holstein, 241 Gyr and 1515 Holstein × Gyr) were exposed to forskolin for 48 h prior to vitrification. The pregnancy rate achieved with Gyr dam and sire was 46.1%, which was similar (p = 0.11) to that of Holstein dam and Gyr sire (40.3%). Crossing Gyr dams with Holstein sires resulted in a pregnancy rate of 38.9% and did not differ (p = 0.58) from the pregnancy rate obtained with the cross between Holstein dams and Gyr sires. The rate obtained with Holstein dam and sire was 32.5%. The average pregnancy rate was 36.6%, and no difference was found in the proportion of female foetuses (88.8%, in average) among breeds (p > 0.05). In conclusion, transfer of cryopreserved X‐sorted embryos represents an interesting choice for dairy cattle. Despite the small differences between pregnancy rates, we highlight the efficiency of this strategy for all of the racial groups studied.  相似文献

Soil surrounding a growing root must be displaced to accommodate the increased root volume. To ease soil penetration, root caps produce border cells and mucilage that lubricate the root surface, decreasing friction at the root‐soil interface. Rhizosphere deformations caused by roots with or without a functional root cap were compared to determine the effects of the root cap on sand displacement and penetration. Intact (KYS wild type) and decapped (agt1dec mutant) primary maize roots were grown in observation chambers filled with sand. Non‐destructive time‐lapse micro‐imaging combined with particle image velocimetry was used to visualize and quantify sand displacements as small as 0.5 µm caused by growing roots. Decapped (agt1dec) roots displayed typical responses of mechanically impeded roots at sand densities that did not affect intact KYS roots. Sand displacement decreased exponentially with distance from the root and extended four to eight root radii into the sand. The calculated mean sand density increase and the compressed sand area were doubled by decapping. Maximum density often occurred in front of the apex of decapped roots whereas it occurred along the sides of intact roots. Periodic variation in sand deformation was observed, probably associated with root circumnutation, which may also facilitate soil penetration. Sand particles moved alongside KYS roots more easily than they did alongside agt1dec roots. A functional exuding cap was therefore essential for efficient rhizosphere deformation and penetration by roots. Manipulating root tip, and specifically root cap, properties is a possible target for improving root penetration in hard soil.  相似文献

The way soil is disrupted and deformed by earthworm movement is hard to quantify non-destructively. Two anecic earthworm species, Aporrectodea longa (Ude) and Lumbricus terrestris L., were compared in their effect on the soil around them as they burrow. Image analysis (particle image velocimetry, PIV) was used successfully to quantify the distance and direction of soil particle displacement by earthworm locomotion giving a unique insight into their effect on the soil structure development. The data collected from both species using PIV show a decline in soil displacement at increasing distance from the earthworm’s body. The quantity of soil being displaced was more to the sides of the earthworms (radially) than in front of the prostomium (axially). Also, L. terrestris displaced more soil than A. longa both axially and radially. The findings from this study suggest that PIV image analysis is a viable tool for examining soil displacement by earthworms and the method used has the potential to be applied to other soil organisms.  相似文献

ABSTRACT

Aim: To determine if vitamin D3 treatment reduced the incidence of vaginal prolapse in pregnant sheep on a North Canterbury sheep breeding property.

Methods: Pregnant ewes from a single farm were allocated to three treatment groups in May 2018. At this time, the first group (EarlyVitADE; n?=?512) received an I/M 1?mL dose of 500,000?IU/mL vitamin D3, 60,000?IU/mL vitamin A, and 25?mg/mL vitamin E. This was repeated in July 2018, when the second group (LateVitADE; n?=?695) also received the same treatment. The third group (n?=?737) were untreated controls. All cases of vaginal prolapse on the property were recorded from pregnancy diagnosis in June 2018 until ewes were set-stocked in August 2018. The planned start of lambing was 10 August 2018.

Results: During the period of observation, vaginal prolapses were recorded in 3/699 (0.4%) 2-year-old ewes, and the odds of vaginal prolapse were not associated with treatment group in these ewes (p?>?0.3). Amongst ewes aged ≥3 years, during the same period, there were 6/333 (1.8%), 6/443 (1.4%) and 25/469 (5.3%) cases in the EarlyVitADE, LateVitADE and control groups, respectively. Compared to control ewes, the odds of vaginal prolapse were reduced in both the EarlyVitADE (OR?=?0.37; 95% CI?=?0.15–0.92) and LateVitADE (OR?=?0.25; 95% CI?=?0.10–0.62) treatment groups.

Conclusions and clinical relevance: In this preliminary study, administration of injectable vitamins A, D3, and E to pregnant ewes reduced the incidence of vaginal prolapse during the period from pregnancy diagnosis to set-stocking on one North Canterbury hill-country farm. Due to the restricted data collection period, this investigation should be replicated to better quantify the repeatability of the observed treatment effect over the complete lambing period.  相似文献

Splitting of the sun's global oscillation frequencies by large-scale flows can be used to investigate how rotation varies with radius and latitude within the solar interior. The nearly uninterrupted observations by the Global Oscillation Network Group (GONG) yield oscillation power spectra with high duty cycles and high signal-to-noise ratios. Frequency splittings derived from GONG observations confirm that the variation of rotation rate with latitude seen at the surface carries through much of the convection zone, at the base of which is an adjustment layer leading to latitudinally independent rotation at greater depths. A distinctive shear layer just below the surface is discernible at low to mid-latitudes.  相似文献

The paper wasp Polistes atrimandibularis is an obligatory social parasite of another Polistes species, P. biglumis bimaculatus. To control the host nest, the parasite sequentially changes the composition of its chemical signature, the cuticular hydrocarbons, during the colonial cycle. Gas chromatography-mass spectrometry of the cuticular hydrocarbons at every stage of the cycle showed that the parasite can switch on and off an entire chemical family, namely, the unsaturated hydrocarbons. In this way the parasite can match the host signature at a critical moment of the colonial cycle.  相似文献

Understanding of the detailed mechanisms of how roots anchor in and reinforce soil is complicated by the variability and complexity of both materials. This study controlled material stiffness and architecture of root analogues, by using rubber and wood, and also employed real willow root segments, to investigate the effect on pullout resistance in wet and air‐dry sand. The architecture of model roots included either no laterals (tap‐root) or a single pair at two different locations (herringbone and dichotomous). During pullout tests, data on load and displacement were recorded. These studies were combined with Particle Image Velocimetry (PIV) image analysis of the model root‐soil system at a transparent interface during pullout to increase understanding of mechanical interactions along the root. Model rubber roots with small stiffness had increasing pullout resistance as the branching and the depth of the lateral roots increased. Similarly, with the stiff wooden root models, the models with lateral roots embedded deeper showed greatest resistance. PIV showed that rubber model roots mobilized their interface shear strength progressively whilst rigid roots mobilized it equally and more rapidly over the whole root length. Soil water suction increased the pullout resistance of the roots by increasing the effective stress and soil strength. Separate pullout tests conducted on willow root samples embedded in sand showed similar behaviour to the rigid model roots. These tests also demonstrated the effect of the root curvature and rough interface on the maximum pullout resistance.  相似文献
