Dogs can be infected by a wide variety of Bartonella species. However, limited data is available on experimental infection of dogs with Bartonella strains isolated from domestic animals or wildlife. We report the inoculation of six dogs with Bartonella henselae (feline strain 94022, 16S rRNA type II) in three sets of two dogs, each receiving a different inoculum dose), four dogs inoculated with B. vinsonii subsp. berkhoffii type I (ATCC strain, one mongrel dog) or type II (coyote strain, two beagles and one mongrel) and B. rochalimae (coyote strain, two beagles). None of the dogs inoculated with B. henselae became bacteremic, as detected by classical blood culture. However, several dogs developed severe necrotic lesions at the inoculation site and all six dogs seroconverted within one to two weeks. All dogs inoculated with the B. v. berkhoffii and B. rochalimae strains became bacteremic at levels comparable to previous experimental infections with either a dog isolate or a human isolate. Our data support that dogs are likely accidental hosts for B. henselae, just like humans, and are efficient reservoirs for both B. v. berkhoffii and B. rochalimae.  相似文献

The antinociceptive and behavioral effects of methadone (MET) alone or combined with detomidine (DET) were studied in horses. Intravenous treatments were randomly administered in a two-phase crossover study. In phase 1, six horses were treated with saline (control) or 0.2 or 0.5 mg/kg methadone (MET0.2; MET0.5, respectively). In phase 2, six horses were treated with 0.01 mg/kg DET alone or with DET combined with 0.2 mg/kg MET (DET/MET0.2). Thermal nociceptive threshold (TNT) and electrical nociceptive thresholds (ENT) were recorded by using a heat projection lamp and electrodes placed in the coronary band of the thoracic limbs, respectively. Spontaneous locomotor activity (SLA) was studied by movement sensors in the stall (phase 1). Chin-to-floor distance was assessed in phase 2. In phase 1, the TNT increased significantly for 30 minute after MET0.5 but not after saline or MET0.2. Hyperesthesia and ataxia were observed in 2 of 6 and 6 of 6 horses after MET0.2 and MET0.5, respectively. SLA increased significantly for 120 minutes after MET in a dose-dependent way, but not after placebo. In phase 2, DET and DET/MET0.2 significantly increased the TNT and ENT above baseline for 15 and 30 minutes, respectively; thresholds were significantly higher with DET/MET0.2 than with DET at the same times. Chin-to-floor distance decreased significantly from baseline for 30 minutes, and no excitatory behavior was observed in both treatments. Although the higher dose of MET induced short-acting antinociception, the associated adverse effects may contraindicate its clinical use. The lower dose of MET potentiated DET-induced antinociception without adverse effects, which might be useful under clinical circumstances.  相似文献

Ageing of drip irrigation systems due to clogging of emitters is considered the largest maintenance problem in microirrigation and this problem is enhanced in subsurface irrigation systems. Whatever the source of the clogging problem, a methodology for early detection of clogging in the field can be useful in decision-making about deploying cleaning processes (flushing or injection of chemicals) and avoiding replacement of laterals. This work presents a methodology for simulating clogging conditions able to reproduce the effects of clogging on pressure profiles, head loss, and emitters flow rate distribution along a single levelled lateral with constant inlet pressure. This methodology was validated by several experiments conducted under controlled conditions of clogging induced by changes in the flow rate of emitters. The effects of clogging intensity and position on hydraulic parameters of a single lateral were analysed in detail and aspects relating to pressure, head loss, and flow rate measurements were discussed. For a given lateral set-up, it is possible to draw a chart relating flow rate and head loss for various levels and positions of clogging. Assuming that measurements of head loss and flow rate are available, this diagram enables immediate estimation of the level and location of clogging, which is useful for deploying maintenance routines. Finally, a protocol was proposed to detect the level of clogging and estimate its position along a levelled lateral under constant inlet pressure. Although constant lateral inlet pressure is atypical in large agricultural settings, it may occur in greenhouse or experimental settings.  相似文献

  1. Extreme climate changes during the Cenozoic Era strengthened different biogeographical barriers that decreased the connectivity among populations, triggering lineage diversification of different species worldwide.
  2. The mitochondrial DNA control region was employed to explore the phylogeography of Sphyrna zygaena, a globally distributed species threatened by unsustainable, illegal, unreported and unregulated fisheries triggered by the international shark fin trade. It is listed as ‘Vulnerable’ by the IUCN Red List and its trade is regulated by CITES Appendix II.
  3. Only 13 haplotypes were found with low genetic diversity levels (hd = 0.686 ± 0.014; π = 0.00206 ± 0.00004) compared with other species of the Sphyrnidae family. The species has a very strong phylogeographic population structure among the Atlantic, Indian and Pacific Oceans (ΦST = 0.79132). Worldwide, there are six distinct populations with some haplotype sharing.
  4. These populations are probably connected by a stepping-stone dispersal of a small number of migrants per generation from the Indo-Pacific towards the Atlantic. Modelling suggests that S. zygaena diverged into two lineages around 6.96 million years ago which have been isolated in glacial refuges in the Atlantic and Indo-Pacific oceans; and after deglaciation, a population expansion probably permitted secondary contact.
  5. Conservation plans to establish differentiated management units should be adopted in each of the identified populations. Among these, the Eastern Central Atlantic and West Indo-Pacific are the most important areas for the species considering the historical migration routes that act as a bridge connecting the Atlantic and Indo-Pacific Oceans while the Gulf of Guinea connects the Atlantic populations. Still, further studies are required to know if these populations are also linked with nursery areas for the species.
  6. The results herein can help to delimit the main evolutionarily significant units to implement effective policies to establish differentiated management units as starting points to genetic monitoring programmes for Sphyrna zygaena.

Summary Broadening agronomic adaptation will improve yield stability in the grain legume Vicia faba L. We gathered information on the adaptation of European and Mediterranean material to European and Mediterranean environments. The material comprised 20 inbred lines (12 European and 8 Mediterranean lines) and 99 intra- and interpool-crosses in generation F1. These were evaluated in 9 environments: two spring-sown Southern German environments (SGermE), and seven autumn-sown Mediterranean environments (MedE) in Sicily, Puglia, Andalucia and South Africa. Standard ANOVA, stability analyses and AMMI analysis were performed. Mean yield in F1 was 257 g/row, the overall parental mean was 144 g/row. The range of environmental means was from 94 g/row to 411 g/row. The average regression coefficient in F1 was b i =1.07, being significantly greater than for the parents (b i =0.68). The opposite was true for the relative magnitude of the deviations from the regressions, which were highly correlated to the AMMI-PC1-results. The AMMI analysis clearly separated the SGermE from the MedE, as well as the germplasm pools. Though the superiority of the F1-hybrids over their parents was striking, their pattern of interactions with the environments strictly reflected that of their parents. A number of promising crosses was identified as a nucleus of a widely adapted faba bean genepool.  相似文献

Purpose

Understanding the ability of ecosystem processes to resist to and to recover from disturbances is critical to sustainable land use. However, the spatial variability of the stability has rarely been addressed. Here, we investigated the functional stability of a soil microbial process for 24 soils collected from adjacent locations from a 0.3?ha tropical rainforest plot in Paracou, French Guiana.

Materials and methods

The 24 locations were characterized regarding soil chemical and biological (microbial diversity) parameters and forest structure. The corresponding soils were submitted to an experimental transient heat disturbance during a microcosm experiment. The response of the respiration process was followed using substrate-induced respiration (SIR).

Results and discussion

The response of soil SIR to heat disturbance varied widely between samples. The variability of the SIR response increased just after the disturbance, and a global rather homogeneous decrease in SIR rates was observed 15 and 30?days after. The stability of SIR in response to heat disturbance could not be related to either the genetic or the metabolic diversity of the microbial community. The initial level of SIR before the disturbance was the soil variable that best correlated with the impact of the disturbance: the soil locations with the highest initial SIR rates were the most affected 15 and 30?days after the heat disturbance.

Conclusions

Such a heterogeneous response suggests that the response of soil processes to a disturbance will be difficult to assess from only local-scale analyses and highlights the need for spatial explicitness in understanding biogeochemical processes.  相似文献

Genetically modified (GM) maize containing genes from the soil bacterium Bacillus thuringiensis (Bt) was cultivated on 29% of the total maize production area worldwide in 2009. Most studies to date compare Bt-maize varieties with their near isogenic lines; however, there is little information on the variability of conventional maize breeding lines and how the effects of Bt varieties are ranked within. In our study on the potential risks of Bt-maize varieties, we analyzed tissue quality and compared the effects of ten conventional and GM maize varieties on soil microbiological properties in a replicated climate chamber experiment. All maize varieties were cultivated twice in the same soil microcosm. Shoot yields and soluble C in leaf tissue of Bt varieties were higher than the ones of non-Bt. Soil dehydrogenase activity was reduced by 5% under Bt varieties compared to non-Bt, while most of the other soil microbial properties (soil microbial biomass, basal respiration) showed no significant differences between Bt and non-Bt varieties. The leaves and roots of one Bt variety were decomposed to a greater extent than the ones of its near isogenic line; the conventional breeding lines also showed higher values. Changes in crop and soil parameters were found when comparing the first and the second crops, but the effects of repeated cropping were the same for all tested varieties. For the studied parameters, the variation among non-Bt-maize varieties was similar to the difference between Bt and non-Bt varieties.  相似文献

A new route for the synthesis of fatty alcohol derivatives of hydroxytyrosol and other olive oil phenolic compounds was developed to allow the preparation of unsaturated derivatives. The biological activity of synthesized compounds was evaluated. Most of the compounds presented a significant antioxidant activity on low-density lipoprotein (LDL) particles. The activity of the tested products was significantly influenced by the number and position of unsaturations as well as modifications on the polar head of the synthesized compounds. Some of them presented modulation of food intake in rats and, due to their molecular similarity with CB(1) endogenous ligands, the endocannabinoid system and PPAR-α were also evaluated as potential targets. The pharmacodynamics could not be totally explained by CB(1) and PPAR-α receptor interactions because only two of the four compounds with biological activity showed a CB(1) activity and all of them presented low PPAR-α affinity, not justifying its whole in vivo activity. The hydroxytyrosol linoleylether (7) increased LDL resistance to oxidation with a capacity similar to that of hydroxytyrosol and was the most active in vivo compound with a hypophagic effect comparable to that of oleoylethanolamine. We consider that this compound could be a good lead compound for future drug development in obesity treatments.  相似文献

Northern corn leaf blight (NCLB), caused by Exserohilum turcicum, is one of the most devastating diseases affecting maize yield worldwide. A foliar spray of nickel (Ni) to potentiate maize resistance against NCLB was investigated by examining alterations in the photosynthetic apparatus (leaf gas exchange and chlorophyll [Chl] a fluorescence parameters), production of ethylene and reactive oxygen species as well as activities of defence and antioxidant enzymes. Mycelial growth of E. turcicum was inhibited by Ni in vitro. Inoculated plants sprayed with Ni exhibited higher foliar Ni concentration, reduced NCLB symptoms, and lower concentrations of malondialdehyde and hydrogen peroxide. In inoculated leaves of plants not sprayed with Ni, concentrations of Chl a, Chl b, and carotenoids were lower and the photosynthetic apparatus was impaired at the biochemical level due to high NCLB severity. The activities of antioxidant enzymes were not affected by Ni, except an increase in glutathione reductase activity for noninoculated plants sprayed with Ni. High lipoxygenase and polyphenoloxidase activities, lower ethylene production, as well as elevated production of phenolics and lignin helped decrease NCLB severity in the leaves of Ni-sprayed plants.  相似文献
