Purpose

Standard assays for phytotoxicity provide a reductionist view on the performance of plants under toxic stress. To address two of the most prominent issues in plant toxicity studies, our aims were (1) to assess how well physiological and biochemical parameters complement standard toxicological endpoints when testing natural soils and (2) to assess the suitability of three commonly used control soils as comparative references.

Material and methods

We compared the performance of Zea mays and Helianthus annuus in three control soils (artificial Organisation for Economic Co-operation and Development (OECD) soil, standard LUFA 2.2 soil, and turf-perlite) against three natural soils representing a gradient of contamination (from a deactivated uranium mine). Standard endpoints (emergence and biomass) were estimated, along with pigment content, photosynthetic parameters, cellular injury, and proline content.

Results and discussion

The toxicological profile of natural soils was highly dependent on the control soil used as reference; also, plant physiological performance was influenced by the soils’ properties. We discuss the need to interpret and combine multiple lines of evidence as a way to increase the degree of confidence one classifies soils based on their ecotoxicity, and this is where the integration of physiological and biochemical parameters bring added value.

Conclusions

When facing large variability in soil characteristics, it is best to collect and integrate as much information possible to strengthen conclusions about phytotoxicity of natural soils. Obviously, this refutes reductionist views and places the final conclusion in the hands of expert judgment.

Soil N mineralization was quantified in two long-term experiments in northern France, in which no-till (NT) and conventional tillage (CT) had been differentiated for 33 years (Site 1) and 12 years (Site 2). Both sites had the same soil type but differed in crop rotation. N mineralization kinetics were assessed in situ in bare soil in both systems for 254 days (Site 1) and 555 days (Site 2) by taking frequent measurements of water and nitrate contents from soil layers and using the LIXIM calculation model. The N mineralization potential was also determined in soil samples incubated under controlled laboratory conditions. Small or non-significant differences in water and nitrate contents between NT and CT were apparent within the soil profiles on both sites. Net mineralization did not differ significantly between sites or tillage treatments. The amount of N mineralized from August 2003 to April 2004 was 67 ± 10 kg N ha−1 on Site 1 and 74 ± 5 kg N ha−1 on Site 2, and 161 ± 6 kg N ha−1 from August 2003 to February 2005 on Site 2. The kinetics of N mineralization versus normalized time (equivalent time at constant temperature of 15 °C and water content at field capacity) were linear during the shorter period (254 days corresponding to 120 normalized days). The slope (N mineralization rate) did not differ significantly between treatments and sites, and the average rate was 0.57 ± 0.05 kg N ha−1 nd−1. The kinetics were non-linear on Site 2 over the longer period (555 days corresponding to 350 normalized days). They could be fitted to an exponential model with a slope at the origin of 0.62 kg N ha−1 nd−1. The N mineralization kinetics obtained in laboratory incubations for 120–150 normalized days were also almost linear with no significant differences between treatments. Assuming that mineralization took place in the ploughed layer (in CT) or over the same soil mass (in NT) they were in good agreement with the kinetics determined in situ on both sites. The calculated water drainage below the sampled profile was slightly greater in NT due to lower evaporation. The calculated leached N was slightly higher in NT than CT on Site 1, but did not differ between treatments on Site 2. It is concluded that N mineralization and leaching in NT and CT were similar, despite large differences in N distribution within the soil profile and a slight difference in organic N stock.  相似文献

Volatile concentrates from the oleo-gum resin of Commiphora myrrha (Nees) Engl. and from the rhizomes of Acorus calamus were isolated by supercritical extraction with carbon dioxide. The volatile oil of myrrh was obtained at 9.0 MPa and 50 degrees C and at a CO2 flow of 1.5 kg/h. Acorus calamus was extracted at 9.0 MPa and 45 degrees C and at a CO2 flow of 1.6 kg/h. In both cases, an oil devoid of cuticular waxes was obtained with a single depressurization stage. The SFE myrrh oil had a yield, Y, of 3.2%. Its main components, identified and quantified by GC/MS, were furanoeudesma-1,3-diene, 34.9%; lindestrene, 12.9%; curzerene, 8.5%; and germacrone, 5.8%. The essential oils from the same starting material by hydrodistillation, HD, (Y = 2.8%) and by steam distillation, SD, (Y = 0.4%) were quite similar to the SFE extract. The main components of the SFE oil of A. calamus (Y = 3.5%) were acorenone, 13.4%; iso-acorone, 11.6%; (Z)-sesquilavandulol, 11.0%; dehydroxy isocalamendiol, 7.7%; and beta-asarone, 5.5%. The comparison with hydrodistilled (Y = 1.8%) and steam distilled (Y = 1.0%) oils revealed large differences in the content of iso-acorone and crypto-acorone.  相似文献

Gluten-glycerol dough was extruded under a variety of processing conditions using a corotating self-wiping twin-screw extruder. Influence of feed rate, screw speed, and barrel temperature on processing parameters (die pressure, product temperature, residence time, specific energy) were examined. Use of flow modeling was successful for describing the evolution of the main flow parameters during processing. Rheological properties of extruded samples exhibited network-like behavior and were characterized and modeled by Cole-Cole distributions. Changes in molecular sizes of proteins during extrusion were measured by chromatography and appeared to be correlated to molecular size between network strands, as derived from the rheological properties of the materials obtained. Depending on operating conditions, extrudates presented very different surface aspects, ranging from very smooth-surfaced extrudates with high swell to completely broken extrudates. The results indicated that extrudate breakup was caused by increasing network density, and some gliadins may have acted as cross-linking agents. Increasing network density resulted in decreasing mobility of polymeric chains, and “protein melt” may no longer have been able to support the strain experienced during extrusion through the die. Increasing network density was reflected in increased plateau modulus and molecular size of protein aggregates. Increasing network structure appeared to be induced by the severity of the thermomechanical treatment, as indicated by specific mechanical energy input and maximum temperature reached.  相似文献

This work studies the influence of two mulching treatments on soil properties and the field performance of afforested holm‐oak seedlings (Quercus ilex L. subsp. ballota (Desf.) Samp.) nine years after outplanting. Mulching treatments composed of stones, forest debris, and an untreated control were randomly applied to 180 seedlings (n = 60) in January 2001 at the bed level (1 × 1 m) in an abandoned agricultural field in SE Spain. Survival, growth measured by means of leaf area, and nutrient concentrations in leaves and soil were measured. Both mulches provided higher survival and greater leaf‐area growth than the control, but did not differ in leaf‐nutrient concentration. Most of the analysed soil variables were not affected by the mulching treatments at the end of the study period, and, therefore, the soil properties changes by mulching might be slower than expected under semi‐arid conditions.
