We describe the efficacy of L-870812, an inhibitor of HIV-1 and SIV integrase, in rhesus macaques infected with the simian-human immunodeficiency virus (SHIV) 89.6P. When initiated before CD4 cell depletion, L-870812 therapy mediated a sustained suppression of viremia, preserving CD4 levels and permitting the induction of virus-specific cellular immunity. L-870812 was also active in chronic infection; however, the magnitude and durability of the effect varied in conjunction with the pretreatment immune response and viral load. These studies demonstrate integrase inhibitor activity in vivo and suggest that cellular immunity facilitates chemotherapeutic efficacy in retroviral infections. 相似文献

The quality of protein concentrates coming from conventional and non-conventional vegetable sources was evaluated. The protein quality was assessed by the multi-point assay RPV. Whole body weight gain, weight gain corrected for the gastrointestinal content (G.I.), total body nitrogen and total liver nitrogen were used as response parameters. The various protein sources ranked differently depending upon the response parameters utilized, but a great majority of the sources had a good nutritional value (over 0.5). The gastrointestinal content is one of the principal factors responsible for the differences found. However, even after correcting for the G.I. content, some differences remained. For the various protein sources, the nitrogen deposition in the liver correlates satisfactorily with the whole body nitrogen deposition, if in the calculation of the regression line, the data for faba bean and tobacco were omitted (respectivelyr=0.610 andr=0.925). On the whole, these data indicate that most plant protein concentrates in suitable combinations and properly processed, may become a promising alternative to animal proteins. 相似文献

Tomato black ring virus (TBRV) is an important pathogen infecting a wide range of plant species worldwide. Phylogenetic studies of TBRV have already been conducted, although limited by the use of short genomic regions or a reduced amount of isolates. In the present study, we carried out an exhaustive phylogenetic and population genetic analysis based on the coat protein gene (CP) sequence of 57 TBRV isolates originating from different host plants and European geographic regions (47 isolates from Poland, 8 from Lithuania, one from the UK, and one from Hungary). Moreover, the selective pressure acting on particular codons and coevolution of amino acid residues in the CP were analysed. The results clearly showed that the TBRV population is being shaped by recombination and both positive and purifying selection. The analyses revealed that the placement of TBRV isolates in the phylogenetic trees was nonrandom, with isolates clustering according to host plant families and geographic origin. 相似文献

Brown alga Bifurcaria bifurcata is a prolific source of bioactive acyclic (linear) diterpenes with high structural diversity. In the continuation of our investigations on Irish brown algae, we undertook an in-depth chemical study on the n-hexanes and chloroform subextracts of B. bifurcata that led to isolation of six new (1–6) and two known (7–8) acyclic diterpenes. Chemical structures of the compounds were elucidated by a combination of 1D and 2D NMR, HRMS, FT-IR, [α]D and vibrational circular dichroism (VCD) spectroscopy. Compounds 1–8, as well as three additional linear diterpenes (9–11), which we isolated from the same seaweed before, were tested against the human breast cancer cell line (MDA-MB-231). Several compounds moderately inhibited the growth of the MDA-MB-231 cell line with IC50 values ranging from 11.6 to 32.0 μg/mL. The present study carried out on the lipophilic extracts of the Irish B. bifurcata shows the enormous capacity of this seaweed to produce a large palette of acyclic diterpenes with diverse oxygenation and substitution patterns and promising bioactivities. 相似文献

of special importance for conservation. Using Rubus idaeus as an example, we propose a compound selection of wild populations for in situ conservation. Twelve raspberry populations
in the Leningrad region have been evaluated from the point of view of their significance for in situ conservation. To exclude the run-wild forms from the evaluation process, we tried to identify them applying a mathematical
model developed by statusmetry method. The constructed model is based on morphological features of leaves of primocanes and
helps to distinguish the wild forms from the cultivated and run-wild ones. The wild raspberry populations were screened for
plants with features useful for cultivated raspberry improvement such as waxy cover presence, yellow fruiting plants, spinelessness,
fruiting primocanes and others. The diversity of wild raspberry populations was assessed by variation of morphological features,
presence of botanical varieties and polymorphism of seed storage proteins. The results of viability status assessment for
the chosen populations reflect the perspectives of their development and permit to determine the strategy and measures for

There is substantial interest in the development of drugs that limit the extent of ischemia-induced cardiac damage caused by myocardial infarction or by certain surgical procedures. Here, using an unbiased proteomic search, we identified mitochondrial aldehyde dehydrogenase 2 (ALDH2) as an enzyme whose activation correlates with reduced ischemic heart damage in rodent models. A high-throughput screen yielded a small-molecule activator of ALDH2 (Alda-1) that, when administered to rats before an ischemic event, reduced infarct size by 60%, most likely through its inhibitory effect on the formation of cytotoxic aldehydes. In vitro, Alda-1 was a particularly effective activator of ALDH2*2, an inactive mutant form of the enzyme that is found in 40% of East Asian populations. Thus, pharmacologic enhancement of ALDH2 activity may be useful for patients with wild-type or mutant ALDH2 who are subjected to cardiac ischemia, such as during coronary bypass surgery. 相似文献

Tomato black ring virus (TBRV), a member of the Nepovirus genus, is a serious plant pathogen distributed worldwide. It causes significant damage to several economically important crops, such as artichoke or strawberry. The TBRV bipartite genome consists of two polyadenylated single-stranded positive-sense RNA molecules, which may be accompanied by subviral particles such as defective interfering RNAs (DI RNAs) and satellite RNAs (satRNAs). In this study, we obtained the complete genome sequence of six TBRV isolates originating from different hosts and determined the presence of eight TBRV satRNAs. Subsequently, genetic variability, recombination, phylogenetic and selection pressure analyses were performed. The results revealed that the TBRV population is genetically diverse. The occurrence of potential recombination events, evidence of positive selection pressure acting on particular codons and the diversification of satRNAs within the TBRV population indicated that the virus mutates and can rapidly adapt to new environmental conditions or hosts. The presented data shed some light on TBRV evolutionary dynamics and epidemiology. 相似文献
