Since 1993, Rhizoctonia crown and root rot (Rhizoctonia solani AG 2–2 IIIB) has represented an increasing problem for sugar beet production in Germany. Up to now, the outbreak of the infection and the spread of the disease within a field cannot be predicted and effective countermeasures are not available. Although little is known about the living conditions of R. solani in soils, abiotic soil properties are likely to influence the disease occurrence. Investigations were carried out based on 60 pairwise comparisons, each consisting of a disease‐affected and an adjacent nonaffected patch on farmers' fields in 2002 and 2003. Soil samples from the top soil layer (0–30 cm) were collected before harvest, and eight of the most frequently mentioned soil properties potentially influencing Rhizoctonia crown and root rot infection were examined: bulk density, texture, carbonate carbon, potassium, phosphorus, organic carbon, total nitrogen, and pH. The occurrence of the disease was significantly related to the soil C : N ratio, indicating the influence of soil organic matter on the disease occurrence. Examinations of soil thin sections showed that organic‐matter particles in the soil serve as a substrate for R. solani. All other soil physical and chemical properties examined did not differ between the disease‐affected and nonaffected patches and seem to be of minor importance. 相似文献

Knowledge about spatial soil variation in terms of measured pedodiversity, as well as the spatial distribution of soils in terms of spatial subset representativity, offers the possibility to estimate the quality and variance within a soil map. Additionally, it can help to identify representative sample locations. Demonstrated at the German soil map at a scale of 1:1,000,000, this study describes a methodology to analyze the distribution of taxonomical pedodiversity using the Simpson index and a new approach to derive representative spatial subsets based on a modified χ2‐test (χm2), which can be used as monitoring areas. To analyze the spatial composition of the soil map and to detect differences in the underlying mapping schemes of the German soil map 1:1,000,000, three different spatial data structures were studied: (1) the entire soil map, (2) the soil map segmented into geomorphological regions, and (3) the soil map segmented into the Federal States of Germany. Representative patches of varying sizes were statistically derived for all spatial subsets as well as the entire soil map ranging from 20 km × 20 km up to 70 km × 70 km. The results show that the measured pedodiversity is linked to both the geomorphology as well as the political borders of the Federal States. On the one hand, this reveals the uncertainty of measuring pedodiversity on the basis of soil‐class maps as the spatial representation of pedodiversity is influenced by the different mapping traditions and methods applied in the 16 Federal States of Germany. On the other hand, it allows the analysis of the aggregation schemes of different landscapes. The presented approach helps to understand large soilscapes and to compare different soil maps of different states and countries as well as to enhance the soil map with additional information. Suitable predictors of degradability are sought to support the identification of biochars with large potential to increase C sequestration in soils. We determined the biodegradation of 9 chars from hydrothermal carbonization and pyrolysis in two agricultural soils. The 200‐ and 115‐day degradation correlated strongly with the O:C‐ and slightly with the H:C‐atomic ratio of 9 and 14 biochars, respectively. ABSTRACT: Single nucleotide polymorphisms (SNPs) in the genes for interleukin-4, -13 and interferon-gamma, and 21 additional SNPs which previously had been significantly associated with immune traits in the chicken, were genotyped in white and brown layer hens and analyzed for their association with helminth burden following natural infections. A nucleotide substitution located upstream of the promoter of the interferon-gamma gene was significantly associated with the log transformed number of Ascaridia galli in the brown layer line (genotype CC: 6.4 ± 1.0 worms; genotype CT: 11.7 ± 2.2 worms). Therefore, IFNG seems to be a promising candidate gene for further studies on helminth resistance in the chicken. 相似文献

In this pilot study, we tested whether a soft mat (foam covered with a heat-sealed thermoplastic) reduces alterations and injuries at the skin and the leg joints.The soft mat in the lying area of partly slatted pens was compared to a lying area consisting of either bare or slightly littered (100 g straw per pig and day) concrete flooring. In this study we focused on skin lesions on the legs of finishing pigs as indicators of impaired welfare. Pigs were kept in 19 groups of 8-10 individuals and were examined for skin lesions around the carpal and tarsal joints either at a weight of <35 kg, or at close to 100 kg. The likelihood of hairless patches and wounds at the tarsal joints was significantly lower in pens with the soft lying mat than in pens with a bare concrete floor. Pens with a littered concrete floor did not differ compared to pens with a bare concrete floor. The soft lying mat thus improved floor quality in the lying area in terms of preventing skin lesions compared to bare and slightly littered concrete flooring. Such soft lying mats have thus the potential to improve lying comfort and welfare of finishing pigs. 相似文献

In this study 595 lactating cows originating from 31 carinthian farms were investigated in accordance of liver fluke infection using individual and tank milk as well as individual blood and faecal samples. Two commercial ELISAs were used to test the milk and blood serum, and the results were compared with coproscopy and a commercial copro-antigen ELISA. Receiver operating characteristics (ROC) and two-graph operating characteristics (TG ROC) of tank milk results were conducted based on the individual milk to determine the minimum reliable in-herd antibody prevalence for the predominant condition in the investigation area. In 17.8% of the examined individuals located in 64.5% of the farms eggs were detected by coproscopy. The copro-antigen ELISA delivered 13.4% positive individuals from 54.8% of the farms. The milk ELISAs showed 42.7% (Euroclone) and 44.2% (Pourquier) positive cows on 90.3% of the farms. The blood samples were positive in 43% (Euroclone) and 45.2% (Pourquier) of the individuals from 90.3% to 96.8% of the herds, respectively. Based on the milk and the blood an average in-herd prevalence of 30-45% can be assumed. The serum and milk samples delivered correlating results with kappa values between 0.94 and 0.97, whereas the coproscopy and copro-antigen ELISA did not correlate well with the ELISA results. The two different ELISA tests highly correlated on individual and on herd level. Both showed a reliable minimum in-herd prevalence of ~20%, meaning that one fifth of the individuals in a herd have to be positive to obtain a positive bulk tank milk result. In the investigated area a higher in-herd prevalence is expected, therefore the tank milk is useful as a monitoring tool and can be used as a basis for intervention strategies. 相似文献

The aim of this work was to evaluate the potential of lufenuron, a benzylphenylurea with ability to interfere with the formation of insect exoskeleton, as a therapeutic drug for larval echinococcosis (hydatid disease). For this purpose lufenuron, alone or in combination with albendazole, was administered to CD1 mice bearing Echinococcus granulosus hydatid cysts in the peritoneal cavity. Neither of the drugs alone was able to exert parasiticidal effects. However, in combination with albendazole, lufenuron reduced the growth of cysts (30-40% in cyst diameter respect to control, p<0.05). This effect was associated with ultrastructural alterations of the hydatid cyst wall and a reduction of the content of myo-inositol-hexakisphosphate, the major component of the electron dense granules of the laminated layer. Overall, this work provides evidence that lufenuron could represent a useful compound for the use in chemotherapy against larval echinococcosis, by enhancing albendazole parasiticidal activity. 相似文献

Macrocyclic lactone (ML) molecules have been used for heartworm control for more than 25 years. However, in recent years, there have been reports of loss of efficacy of ML heartworm preventatives against Dirofilaria immitis in some locations in the United States. Macrocyclic lactone resistance is a common problem in nematode parasites of livestock, and more recently, evidence of ivermectin resistance has been reported in the human filarial nematode Onchocerca volvulus. In this study, four D. immitis sample groups from the United States with different treatment histories were investigated for evidence of ML-driven genetic selection. DNA from individual adult worms and microfilariae was amplified by polymerase chain reaction to investigate a gene encoding a P-glycoprotein, a protein class known to be involved in ML pharmacology. A significant correlation of a GG-GG genotype with ivermectin response phenotype was found. Moreover, a significant loss of heterozygosity was found in a low responder group; loss of heterozygosity is commonly seen in loci when a population has been under selection. Further studies are required to confirm ML resistance in heartworm populations. However, the genetic changes observed in this study may be useful as a marker to monitor for ML resistance in D. immitis. 相似文献

Amino sugars are increasingly used as indicators for the accumulation of microbial residues in soil and plant material. A
reverse-phase high-performance liquid chromatography method was improved for the simultaneous determination of muramic acid,
mannosamine, glucosamine and galactosamine in soil and plant hydrolysates via ortho-phthaldialdehyde (OPA) pre-column derivatisation
and fluorescence detection. The retention time was reduced, and the separation of muramic acid and mannosamine was optimised
by modifying the mobile phase. The effects of excitation wavelength, OPA reaction time, tetrahydrofuran concentration and
pH value of the mobile phase on the amino sugar separation were tested. Quantification limits were in the range of 0.13 to
0.90 μg ml−1. No interferences exist from amino acids or other primary amines, occurring in soil and plant hydrolysates. 相似文献
