  1. Devil rays (Mobula spp.) are globally threatened cartilaginous fishes that have attracted global conservation concern owing to their high extinction risk and lack of protection in many countries. Limited resources and data on threatened marine species, including devil rays, impede conservation actions, particularly in developing countries, many of which have high biodiversity.
  2. Devil ray catch is a component of artisanal fisheries in Bangladesh, but data on their fisheries and trade are limited. To characterize devil ray fishing practices, fishers’ perception and trade, 230 fishers and traders were interviewed between 4 June 2018 and 22 June 2019, in four areas of south-east Bangladesh. Catch data were also opportunistically collected at landing sites.
  3. Six devil ray species were documented, caught in an array of gill nets, set-bag nets and longlines. All interviewed fishers reported life-long devil ray bycatch in some numbers, and also noted a decline in catch over the last decade. Bottom trawling, increased bycatch levels, increased demand for devil ray products and, in some cases, ecosystem changes were identified by fishers as threats to devil ray populations.
  4. Unregulated and undocumented trade and retained bycatch, especially by gill nets and set-bag nets, are fuelled by local consumption of devil ray meat and international trade in meat and gill rakers. Compliance with international trade control treaties for all Mobula spp. or the Bangladeshi law protecting Mobula mobular was low, with the majority of fishers (87%, n = 174) unaware of their existence.
  5. To manage devil ray fisheries, and prevent possible population declines, we propose a combination of legally enforced gear modifications, and catch and trade control through community-owned implementation strategies. Additionally, we propose the simultaneous implementation of inclusive, community-based awareness and stewardship projects in conjunction with a coast-wide ray monitoring programme. Finally, we emphasize that more research and action rooted in a sustainable fishery model is urgently needed to protect Bangladeshi devil ray populations.

Banana flour obtained from unripe banana (Musa acuminata, var. Nanicão) under specific drying conditions was evaluated regarding its chemical composition and nutritional value. Results are expressed in dry weight (dw). The unripe banana flour (UBF) presented a high amount of total dietary fiber (DF) (56.24 g/100 g), which consisted of resistant starch (RS) (48.99 g/100 g), fructans (0.05 g/100 g) and DF without RS or fructans (7.2 g/100 g). The contents of available starch (AS) (27.78 g/100 g) and soluble sugars (1.81 g/100 g) were low. The main phytosterols found were campesterol (4.1 mg/100 g), stigmasterol (2.5 mg/100 g) and β-sitosterol (6.2 mg/100 g). The total polyphenol content was 50.65 mg GAE/100 g. Antioxidant activity, by the FRAP and ORAC methods, was moderated, being 358.67 and 261.00 μmol of Trolox equivalent/100 g, respectively. The content of Zn, Ca and Fe and mineral dialyzability were low. The procedure used to obtain UBF resulted in the recovery of undamaged starch granules and in a low-energy product (597 kJ/100 g).  相似文献

BACKGROUND: In most organisms proper reductional chromosome segregation during meiosis I is strongly correlated with the presence of crossover recombination structures (chiasmata); recombination deficient mutants lack crossovers and suffer meiosis I nondisjunction. We report that these functions are separable in the fission yeast Schizosaccharomyces pombe. RESULTS: Intron mapping and expression studies confirmed that Rec12 is a member of the Spo11/Top6A topoisomerase family required for the formation of meiotic dsDNA breaks and recombination. rec12-117, rec12-D15 (null), and rec12-Y98F (active site) mutants lacked most crossover recombination and chromosomes segregated abnormally to generate aneuploid meiotic products. Since S. pombe contains only three chromosome pairs, many of those aneuploid products were viable. The types of aberrant chromosome segregation were inferred from the inheritance patterns of centromere linked markers in diploid meiotic products. The rec12-117 and rec12-D15 mutants manifest segregation errors during both meiosis I and meiosis II. Remarkably, the rec12-Y98F (active site) mutant exhibited essentially normal meiosis I segregation patterns, but still exhibited meiosis II segregation errors. CONCLUSIONS: Rec12 is a 345 amino acid protein required for most crossover recombination and for chiasmatic segregation of chromosomes during meiosis I. Rec12 also participates in a backup distributive (achiasmatic) system of chromosome segregation during meiosis I. In addition, catalytically-active Rec12 mediates some signal that is required for faithful equational segregation of chromosomes during meiosis II.  相似文献

Wheat (Triticum aestivum L.) flour functionality during the dough development and baking processes is an important quality attribute considered by the baking industry. A flour with high water absorption during mixing means more water and less flour is needed, compared to a flour with less water absorption. The objective of this study was to identify quantitative trait loci (QTL) influencing water absorption and dough rheological properties of hard red spring wheat. QTL were mapped on a genetic linkage map that comprised 531 simple sequence repeats (SSRs) and diversity array technology (DArT) marker loci. Composite interval mapping with 139 recombinant inbred lines (RILs) was used to identify QTL within and across two field environments. Six QTL on chromosomes 1A, 1B, 4B, 4D, and 5A were detected for farinograph water absorption. These QTL also confirmed earlier studies that flour water absorption is a function of protein content, starch damage, and gluten strength. In this study, dough rheological properties such as dough development time, dough stability, mixing tolerance index, and time to breakdown were influenced by the high-molecular weight glutenin genes Glu-B1 and Glu-D1.  相似文献

ObjectiveTo evaluate the effectiveness of paravertebral lumbar plexus block combined with parasacral sciatic block to anesthetize one hind limb in awake dogs.Study designRandomized, controlled, blinded experimental study.AnimalsEight healthy mongrel dogs weighing 12.4 ± 4.5 kg and aged 7 ± 2.33 years.MethodsAfter sedation with medetomidine, dogs received B1: bupivacaine 0.25%, 0.2 mL kg?1, B2: bupivacaine 0.5%, 0.2 mL kg?1, B3: bupivacaine 0.25% 0.4 mL kg?1, P1: NaCl 0.2 mL kg?1, P2: NaCl 0.4 mL kg?1. The lumbosacral plexus was blocked through a paravertebral block of the fourth, fifth and sixth lumbar nerves combined with a parasacral block. The relevant nerves were located using a nerve stimulator and injections of each treatment were administered. Degree and durations of sensory blockade were determined through the response to a Halsted clamp pressure on the skin innervated by the saphenous/femoral and lateral cutaneous femoral nerves (lumbar dermatomes) and by the peroneal and tibial nerves. The degree and duration of motor blockade was assessed evaluating the ability to walk normally and proprioception.ResultsP1 and P2 treatments did not show any grade of sensory or motor blockade. The B2 treatment produced a higher degree of sensory blockade compared to B1 and B3 for both lumbar and sciatic dermatomes. There was no significant difference in the degree of sensory blockade comparing B1 to B3. The B2 treatment had greater motor blockade compared to B1 and B3. The duration of sensory and motor blockade was longer in B2 compared to B1 and B3.Conclusion and clinical relevanceWhen the nerve stimulator is used to perform the lumbosacral plexus block, the concentration of the bupivacaine has a more important role than the volume to produce a more solid and longer block.  相似文献

The effects of grain texture on pastamaking and breadmaking quality were studied in three F8 soft-textured durum wheat lines (SDLs) containing wild-type alleles Pina-D1a and Pinb-D1a as compared with their hard durum sister lines (HDLs). SDLs homozygous for a small 5DS segment, less than 14.4 cM in size, accumulated puroindolines A (Pin-A) and B (Pin-B) and showed SKCS values (19.9-23.6) significantly lower than those (72.6-76.8) of their hard-textured counterparts lacking Pin-A and Pin-B. In addition, SDLs exhibited approximately 24% higher flour extraction rates compared with HDLs. Reducing the kernel hardness decreased farinograph water absorption, dough tenacity (P) and, accordingly, alveograph P/L ratio, but increased farinograph stability, mixing tolerance and dough extensibility (L). Spaghetti cooking quality, as determined by the sensory judgment of firmness, stickiness and bulkiness, was unaffected by the kernel hardness, whereas the loaf volume exhibited a 10% increase associated with kernel softening. Flour and semolina, but not spaghetti, from SDLs showed a substantial reduction in yellowness (b*) and brownness (100 − L*) likely due to their finer particle size compared with HDLs. Alleles Pina-D1a and Pinb-D1a may offer new perspectives for breeding dual purpose (pasta and bread) durum wheat varieties.  相似文献

Transgenic Citrus sinensis ‘Hamlin’ and ‘Valencia’ plants containing Citrus tristeza virus (CTV)-derived sequences were propagated and inoculated with CTV. For propagation, selected buds from transgenic and non-transgenic control plants were grafted onto C. aurantium and C. limonia rootstock plants. CTV inoculation was performed via viruliferous aphids (Toxoptera citricida), and viral detection post-inoculation was performed through DASI-ELISA or RT-qPCR. After four inoculations, none of the transgenic lines tested showed complete resistance. However, viral multiplication was undetectable in some of the propagated clones. These resistant clones mainly came from transgenic ‘Valencia’ sweet orange plants grafted onto C. limonia rootstock containing the pCTV-CS gene construct. Although the tested viral inoculation method represents natural field infection conditions, the results did not differ significantly from those previously reported when the same transgenic lines were bud-graft inoculated. This finding indicates that the difficulties in producing CTV-resistant transgenic citrus lines may be unrelated to the inoculation method. Transgene expression level was quantified by RT-qPCR analysis and it was not possible to relate transgene mRNA level with resistance to the pathogen.  相似文献

Objective To evaluate the endotracheal tube cuff pressure achieved by four different inflation methods. Study design Prospective clinical study. Animals Eighty client owned dogs. Methods After anaesthesia induction, endotracheal intubation was performed using plastic or silicone tubes. A clinician unaware of tube type inflated the cuff by simple digital palpation (method A), thereafter the cuff was deflated and inflated again by a second clinician who tried to reproduce a previously learned cuff pressure of between 19 and 24 mmHg (method B). During method C the cuff was inflated to the minimum occlusive volume at an airway pressure of 20 cm H2O, and in method D the cuff was incrementally deflated until an audible air leak could be heard from the oral cavity at an airway pressure of 25 cm H2O. For all the methods, an operator recorded the actual cuff pressure obtained using a manometer. Heart rate, respiratory rate and mean arterial pressure were monitored throughout the procedure. Results The mean inflation pressure for plastic tubes was 56 ± 28 mmHg for method A, 20 ± 9 mmHg for method B, 35 ± 32 mmHg for method C and 46 ± 39 mmHg for method D. Pressures using silicone tubes were significantly higher than for plastic tubes, the mean registered pressures being 79 ± 39, 33 ± 16, 77 ± 50 and 92 ± 56 mmHg for methods A, B, C and D. Conclusions and Clinical Relevance None of the methods evaluated in this study can be considered effective for inflating the endotracheal tube cuff to within the optimal range when using silicone tubes. Direct measurement of the cuff pressure with a manometer is therefore recommended.  相似文献

Purpose

The possibility of using chemical and microbial additives to enhance the phytoextraction of mercury (Hg) and arsenic (As) from a multi-contaminated soil could be very effective, leading to a significant saving in terms of time and costs of the reclamation. The aim of this study was to evaluate the efficacy of the addition of (i) thiosulfate and (ii) metal-tolerant bacteria isolated from the polluted soil having plant growth promotion (PGP) potential to perform As and Hg phytoextraction by Brassica juncea and Lupinus albus.

Materials and methods

A collection of 13 bacterial isolates able to tolerate As and Hg was obtained from the contaminated soil, identified by partial 16S rRNA gene sequencing and tested in vitro for PGP activities. The most promising strains were further tested in vivo for the evaluation of plant growth ability and rhizocompetence on model plants. Pot experiments were conducted in microcosms, with polluted soil vegetated with B. juncea and L. albus. Ammonium thiosulfate and potassium dihydrogen phosphate were used as mobilizing agents, together with a bacterial consortium composed by the most promising PGP isolates.

Results and discussion

Thirteen indigenous metal-tolerant bacterial strains were isolated, and their in vitro characterization highlighted their great potential in assisting the phytoremediation process; most of them tolerated both trace elements and showed, at the same time, multiple PGP traits. The results were confirmed in vivo on model plants and lead to the selection of the most promising PGP strains to be applied in microcosm-scale phytoextraction experiments. Thiosulfate addition significantly increased the mobilization of both elements, promoting bioavailability and phytoextraction. When a selected bacterial consortium was supplemented in addition to thiosulfate, the efficacy of the phytoaccumulation was increased up to 85 % for As and up to 45 % for Hg.

Conclusions

The use of the common fertilizer thiosulfate appeared to have great potential in phytoextraction practices since it was able to facilitate the uptake by plants of both Hg and As. Moreover, the application of a consortium of indigenous PGP bacteria (PGPB) produced a further positive effect on the plant biomass, supporting and enhancing the phytoextraction strategy, thus demonstrating their potential in a microbe-assisted phytoremediation intervention.

Objective

To evaluate intraoperative and postoperative efficacy of ultrasound (US)-guided femoral (FN) and obturator (ON) nerves block, in the iliopsoas muscle compartment (IPM), using an in-plane technique.

Study design

Anatomical research and randomized, prospective, ‘blinded’ clinical study.

Animals

Six dog cadavers and 20 client-owned dogs undergoing tibial plateau levelling osteotomy (TPLO) surgery.

Methods

In phase 1, anatomical dissections and US imaging of the IPM were performed to design an US-guided nerve block involving the FN and ON simultaneously. The technique was considered successful if new methylene blue solution injection (0.1 mL kg?1) stained FN–ON for ≥2 cm. In phase 2, the US-guided nerve block designed in phase 1, combined with US-guided sciatic nerve (ScN) block, was performed in 20 dogs undergoing TPLO surgery. Patients were assigned randomly to one of two treatment groups: ropivacaine 0.3% (R3, n = 10) and ropivacaine 0.5% (R5, n = 10) at a volume of 0.1 mL kg?1 for each nerve block. Intraoperative success rate (fentanyl requirement < 2.1 mcg kg?1 hour?1) and postoperative pain score [Short Form-Glasgow Composite Measure Pain Scale (SF-GCMPS) ≥ 5/20] were evaluated.

Results

In phase 1, the US image of FN–ON was detected between L6 and L7. In-plane needling technique produced a staining of >4 cm in six of six cases. No abdominal or epidural dye spread was found. In phase 2, median fentanyl infusion rates were 0.5 (0.0–0.9) μg kg?1 hour?1 for R3 and 0.6 (0.0–2.2) μg kg?1 hour?1 for R5. At 9 and 11 hours after the peripheral nerve blocks, an SF-GCMPS ≥ 5 was observed for R3 and R5, respectively.

Conclusions and clinical relevance

The US-guided FN–ON block in the IPM, using an in-plane technique, combined with US-guided ScN block, provided sufficient analgesia to minimize the use of fentanyl during TPLO surgery. A longer postoperative analgesia was observed in group R5 compared with R3.  相似文献
