The rape stem weevil, Ceutorhynchus napi Gyll., is a serious pest of winter oilseed rape (Brassica napus L.) crops in Europe that causes stem distortion, severe disruption of stem growth, and significant yield loss. No resistance has been identified in current B. napus breeding material. Resynthesized lines of oilseed rape can be used to introduce genetic material from the parent species Brassica oleracea L. and B. rapa L. The susceptibility to C. napi of five resynthesized lines and three cultivars of oilseed rape was compared in a multi-choice field plot experiment. Morphological stem traits and stem levels of glucosinolates, carbon, and nitrogen were assessed as potential mediators of resistance. Eggs and larvae of C. napi in stems were counted weekly from the beginning of oviposition to larval maturation and stem pith degeneration in response to oviposition was assessed. Egg counts per stem were negatively dependent on stem length and positively dependent on the levels of five glucosinolates, glucobrassicanapin, glucoalyssin, glucobrassicin, neoglucobrassicin, and gluconasturtiin. The resynthesized line S30 was significantly more resistant to C. napi than the commercial cultivar Sollux, having fewer eggs and larvae. This resistance appeared to be largely antixenotic, due to the combination of a long stem and a paucity of the five key glucosinolates. We propose that resynthesized lines such as S30 should be used to introduce genes determining traits such as long stem length and glucosinolate profile into breeding programs to enhance resistance against C. napi in oilseed rape.  相似文献

The present study investigated genetic diversity, structure and hybridization in a collection of the endangered wild pear species Pyrus pyraster (L.) Burgd. A total of 278 putative ‘true type’ P. pyraster trees originating from seven populations in the federal state of Saxony in Germany were analyzed along with 35 pear cultivars commonly cultivated in Saxony. The genetic analysis was performed using nine nuclear microsatellite markers (ncSSR) and two paternally inherited chloroplast marker (cpDNA) amplifying in the intergenic spacer region trnQrps16 and the intron region rps16. On basis of the ncSSR dataset after STRUCTURE analysis 80 % of the wild pear individuals were assigned as ‘true type’ P. pyraster genotypes. The cpDNA analysis showed shared haplotypes in P. pyraster and P. communis but with an unequal frequency in both species. The analysis of molecular variance resulted in a moderate (ncSSR) and great (cpDNA) variation among ‘true type’ P. pyraster and the pear cultivars. The genetic diversity in the ‘true type’ P. pyraster populations was still high and the genetic structure between the populations low (ncSSR and cpDNA) indicating a genetic exchange between the populations by pollen and seeds. The clear discrimination between the P. pyraster and P. communis confirms our expectation of the existence of ‘true type’ P. pyraster individuals in the study area. The existing genetic integrity and the high genetic diversity argue for the implementation of preservation measures in P. pyraster.  相似文献

Objective : To determine the feasibility of sonographical examination of hip joints in non-sedated puppies and to assess the value of static and dynamic ultrasonography for the early diagnosis of canine hip dysplasia (CHD). Methods : Prospective study was carried out. Five hundred and sixty-six puppies between the ages of 16 and 49 days underwent sonographical determination of the α-angle, joint laxity and distraction value (DV). Sonographical parameters were correlated to conventional radiographic CHD classification in the adult dogs. Results : α-Angles between 74° and 89° (X ±sd=82·8°±2·31°) were measured. Despite the feasibility of sonographical hip assessment in puppies, no statistically significant correlation was found between sonographical determinations of the α-angle, joint laxity, DV and conventional radiographic hip joint classification at the age of 12 to 24 months. Clinical Significance : Results of our study suggest that static and dynamic ultrasonography of hip joints in puppies between 16 and 49 days of age is technically feasible but cannot be recommended for detecting puppies that will develop CHD between the ages of 12 and 24 months.  相似文献

The principal aim of this research was to determine the influence of wildfires on soil organic matter (SOM) content and composition in soils located on the northern slope of the Cantabrian Cordillera, an Atlantic mountainous region in the North West of Spain, where wildfires are frequent. Samples from soils with similar aspect, slope, elevation and vegetation characteristics, but with different wildfires histories were collected. Total organic carbon and total nitrogen contents were determined as well as the C/N ratio. Furthermore, a qualitative characterization of the soil organic carbon (SOC) was carried out by 13C variable amplitude cross polarization magic angle spinning (VACP/MAS) Nuclear Magnetic Resonance (NMR) spectroscopy. Our results show that, on the one hand, all the sampled soils can be considered important pools of carbon in this Atlantic mountainous region, especially in the heath areas. On the other hand, the fire-affected soils present higher SOM contents than their unburnt counterparts. This could be attributed to an important reaccumulation of fresh vegetal material, which is probably a consequence of the decrease of SOM decomposition rates after fire. Moreover, charred organic compounds are not found in all the burnt soils, which could be due to the long time since the last fires events took place, to different fire severities, or to different post-fire erosion processes in the studied soils.  相似文献

In CMS (cytoplasmic male sterility)‐based hybrid rye (Secale cereale L.) breeding, effective pollen‐fertility restoration is an essential prerequisite for achieving maximum grain yield on the one hand and for minimizing ergot (Claviceps purpurea) infestation on the other. Restorer genes for the CMS‐inducing ‘Pampa’ cytoplasm derived from landraces collected in Iran and Argentina are used by breeders for achieving this goal. Here, restorer genes from four germplasm sources (‘Altevogt 14160’, ‘IRAN III’, ‘Trenelense’ and ‘Pico Gentario’) were analysed by producing three‐way cross hybrids between an elite CMS single cross and pollinators with and without a given restorer gene. Materials were evaluated on large drilled plots for restorer index (RI), grain yield, plant height and other traits in six environments. In experiment 1, a restorer gene from ‘Altevogt 14160’ was used. Seven pairs of marker‐selected carrier and non‐carrier backcross lines served as pollinators. In experiment 2, the pollinators were 17 backcross line pairs from the other three germplasm sources. These lines were grouped as high (RI > 67%) and low restorers (RI < 30%), respectively, using testcrosses with a highly diagnostic CMS tester. Hybrids carrying an exotic restorer gene suffered from a significant grain yield reduction by 4.4% and 9.4% and were 9.3 and 4.8 cm taller in experiments 1 and 2, respectively. Thousand‐kernel weight was reduced, whereas quality traits were only slightly affected. For all traits, significant genetic variance existed among the testcrosses to the presence vs. absence of a given exotic restorer gene. This offers a chance for the breeder to reduce or ultimately overcome the presently observed performance reductions brought about by exotic restorer genes.  相似文献

Several caliciviruses are known to cause diseases in animal and man. Amongst them are vesicular exanthema virus of swine (VESV), feline calicivirus, rabbit hemorrhagic disease virus (RHDV), and Norwalk and Sapporo-like viruses; the latter viruses cause gastroenteritis in man. In this paper, an overview of caliciviruses is presented, with particular emphasis on the molecular aspects. The unique properties as compared to other related families of positive-stranded RNA viruses will be discussed. Many findings about the molecular biology of caliciviruses came from systems that are difficult to work with, such as RHDV, which can thus far only be propagated either in animals or in primary rabbit hepatocyte cultures. Therefore, the review will concentrate on RHDV.  相似文献

The national antimicrobial resistance monitoring determines the current quantitative resistance level of life-stock pathogens, in order to permit the evaluation and surveillance of the distribution of resistances on a valid basis. During the examination period from June 2002 to July 2003, a total of 1849 pathogens was collected, following a representative German-wide pattern in collaboration with 29 laboratories. The selection of examined bacterial strains included different specimen causing respiratory diseases in fattening pigs (Pasteurella multocida, Bordetella bronchiseptica) and cattle (Pasteurella multocida, Mannheimia haemolytica), respectively, as well as strains causing mastitis in dairy cows (Staphylococcus spp., Streptococcus spp., E. coli). Determination of the in-vitro susceptibility (minimal inhibitory concentration) to at least 17 antimicrobial agents was performed centrally by the BVL using the microdilution broth method. The findings approximately match results of an analogous study conducted in 2001, and correspondingly revealed significantly lower resistance-values in comparison to data published for Germany so far. No correlation could be established between the incidence of resistance and differing stock densities. By means of the resistance monitoring data gathered by the BVL, the risk potential of antimicrobials applied in Germany can be reliably defined. This valuable information about the epidemiological situation of resistance in Germany can be helpful to veterinarians as a decision guidance when choosing appropriate means of therapy. Accordingly, the results stated by the BVL are apt to provide an important contribution to improving the safety of food-animal products. The experiences obtained from the monitoring also show that valid data about the antimicrobial susceptibility can only be raised on an interdisciplinary approach (federal agencies, county diagnostic laboratories, universities, industry). Currently, the BVL put into practice a study with an extended spectrum of bacterial species and indications.  相似文献

The purpose of this study was to elaborate on the advantages and limits of computed tomography (CT) in the differentiation of thoracic lesions, in particular neoplasias. In the course of the investigation CT-scans of the thorax of 31 dogs with lesions in the area of the lungs or mediastinum were evaluated. The lesions were rated by morphology, distribution pattern, attenuation values and contrast-enhancement. Biopsies or the whole body underwent a pathohistological examination. Of the 31 dogs 17 had neoplastic and 14 had inflammatory lesions in the thoracic region. With help of the CT, the exact localisations of the different lesions was possible in most cases. Due to their characteristic morphologies, distribution patterns and attenuation values the differentiation between inflammatory and neoplastic lesions was possible in most cases (n=25/31) on the basis of the CT-scans. Mean non-enhanced CT attenuation values of the neoplastic lesions ranged between 31 and 50 HU, of the inflammatory lesions between -251 and 9 HU. Both neoplastic and inflammatory lesions showed contrast enhancement (between 14 and 38 HU and between 2 and 95 HU respectively). The mediastinal abcesses enhanced mainly on the periphery of the lesion. A differentiation of the various types of neoplastic lesions based on the non-enhanced attenuation values was not successful. Only metastasis could be differentiated because of their distribution pattern.  相似文献

Lymph nodes are essential structures to be evaluated in an ultrasonographic examination of the feline abdomen. It was hypothesized that current technical proficiency would allow all feline abdominal lymph nodes to be identified ultrasonographically. Ten clinically normal, adult, domestic shorthair cats were examined using real-time compound ultrasonographic imaging. The medial iliac lymph nodes were visible in 100% of the cats, the jejunal lymph nodes in 90%, the hepatic lymph nodes in 70%, the aortic lumbar, the splenic, and the pancreaticoduodenal lymph nodes in 60% each, the ileocecal and the colic lymph nodes in 50% each, and the renal, the gastric, the sacral and the caudal mesenteric lymph nodes in 40%, 30%, 20%, and 10% of the cats, respectively. The inconsistent presence of lymph nodes, their poor echocontrast and interposed gas of the gastrointestinal tract explain the lower percentages of identification. The ultrasonographic length and diameter of the lymph nodes were determined. The majority of these measurements corresponded to those in the literature. We conclude that ultrasonography is a valuable tool for the identification and evaluation of most abdominal lymph nodes in the normal cat. Average ultrasonographic measurements are presented as a preliminary guideline for normal feline abdominal lymph nodes. ete  相似文献
