Weaned wether goats (n = 144) approximately 6 months of age were placed in a 2 x 3 factorial design experiment for 5 months to test the main effects and interaction of two levels of nutrition (growth + maintenance, NUT1; twice growth + maintenance, NUT2) and three levels of Haemonchus contortus burden (0, 500 and 2000 larvae administered every 2 weeks: W0, W500 W2000, respectively) on weight, feed intake, level of infection and packed cell volume (PCV). The rationale for the experimental design was based on the lack of information concerning the interaction between nutritional status and worm burden. Results indicated significant effects of worm burden levels on PCV, faecal egg contents (eggs per gram of feces (EPG)), actual worm numbers, feed intake and efficiency of feed utilization. Nutrition x worm burden interactions were also significant for PCV and EPG. However, the differences detected for PCV and actual worm numbers did not translate into large or consistent differences in body weight. Goats on NUT2, after an initial period, showed little difference in body weight, irrespective of worm burden. Within the NUT1 level, W0 kids weighed more than W500 or W2000 kids throughout the study. Although not statistically significant, this constitutes a trend towards an interaction between nutrition and worm burden. In both nutrition levels, there were no body weight differences between W500 and W2000 until the last 14 days. Feed intake was depressed in the first 3 months of the experiment for infected animals, but was subsequently followed by a compensatory reaction. Lower establishment rates, based on actual worm counts, were observed for the higher infection level, but in both infection levels establishment rates tended to decrease with time. Nutrition was found to be more important to counteract the consequences of a parasitic infection than to counteract the establishment of that same infection.  相似文献

Somatic hybrids (SH) obtained by protoplast fusion were investigated for meiotic behavior in order to determine their possible use in breeding programs. Flower buds from four genotypes of SH between P. edulis f. flavicarpa (E) and the wild species P. amethystina (Am) denoted SH (E + Am) # 12, # 13, # 28 and # 35 were collected and the microsporocytes analysed. Meiotic phases showed the presence of abnormalities such as univalents, bivalents and quadrivalents, laggard chromosomes and anaphase bridges. At least 14 bivalents were observed in most of the cells of the hybrid plants. The percentage of cells with quadrivalents ranged from 73.3 in (E + Am) # 13 to 93.3 in (E + Am) # 28 and # 35. Analysis of pollen viability (V) indicated V = 88.23% for # 12, V = 83.86% for # 13, V = 84.20% for # 28, and V = 72.90% for # 35, and fruit development was observed. The high pollen viability, together with the multivalent pairing observed in the four somatic hybrids indicates that these materials can be used for the genetic improvement of yellow passion fruits aiming at introgressions of genes for resistance to diseases and pests. BR-105 and BR-106 are important tropical maize populations, which were submitted to a high-intensity reciprocal recurrent selection, generating the IG-3 and IG-4synthetics. Using 30-microsatellite loci,we measured and compared the genetic diversity of these populations and their synthetics. The populations did not differ significantly regarding the amount of genetic diversity. As a consequence of selection, genetic variability losses, in terms of mean number of alleles per locus,proportion of polymorphic loci, and gene diversity did occur and were greater in the synthetic IG-3 than IG-4. In the synthetics, the number of loci in adherence to Hardy-Weinberg proportions was superior to that observed in the populations. The Wright's mean fixation index was higher than the mean value expected for outcrossing species (5%) indicating as light excess of homozygotic individuals in both populations. The genetic distances confirmed the favourable effects of one cycle of recurrent selection, as the synthetics became more isolated in comparison to the original populations. Zhao and Running (Reports, 20 August 2010, p. 940) reported a reduction in global terrestrial net primary production (NPP) from 2000 through 2009. We argue that the small trends, regional patterns, and interannual variations that they describe are artifacts of their NPP model. Satellite observations of vegetation activity show no statistically significant changes in more than 85% of the vegetated lands south of 70°N during the same 2000 to 2009 period.  相似文献

Cytokine synthesis inhibitory factor (CSIF; interleukin-10), a product of mouse TH2 T cell clones that inhibits synthesis of cytokines by mouse TH1 T cell clones, exhibits extensive sequence similarity to an uncharacterized open reading frame in the Epstein-Barr virus BCRF1. Recombinant BCRF1 protein mimics the activity of interleukin-10, suggesting that BCRF1 may have a role in the interaction of the virus with the host's immune system.  相似文献

The purpose of this study was to evaluate the physico-chemical characteristics and the post-harvest loss of mass of curly lettuce (Lactuca sativa L.) cv. Verônica that was grown in a protective environment with applications of biopesticides. The curly lettuce was treated by spraying with neem oil, cassava flour wastewater (manipueira), biofertiliser or the control treatment of deltamethrin. We determined the weight, circumference, dry matter, ash, crude protein, total phenols and minerals profile for the heads of lettuce. After harvesting, the lettuce was packed into opened plastic bags and kept at 5 °C for 5 days to evaluate its weight loss. No differences were observed among the treatments for the physico-chemical parameters. The lettuce that was sprayed with manipueira or biofertiliser had the greatest weight loss during storage. The achievement of similar yields with cheaper products suggests a benefit of using organic bioinsecticides.  相似文献

Xanthomonas axonopodis pv. passiflorae causes bacterial spot in passion fruit. It attacks the purple and yellow passion fruit as well as the sweet passion fruit. The diversity of 87 isolates of pv. passiflorae collected from across 22 fruit orchards in Brazil was evaluated using molecular profiles and statistical procedures, including an unweighted pair-group method with arithmetical averages-based dendrogram, analysis of molecular variance (AMOVA), and an assigning test that provides information on genetic structure at the population level. Isolates from another eight pathovars were included in the molecular analyses and all were shown to have a distinct repetitive sequence-based polymerase chain reaction profile. Amplified fragment length polymorphism technique revealed considerable diversity among isolates of pv. passiflorae, and AMOVA showed that most of the variance (49.4%) was due to differences between localities. Cluster analysis revealed that most genotypic clusters were homogeneous and that variance was associated primarily with geographic origin. The disease adversely affects fruit production and may kill infected plants. A method for rapid diagnosis of the pathogen, even before the disease symptoms become evident, has value for producers. Here, a set of primers (Xapas) was designed by exploiting a single-nucleotide polymorphism between the sequences of the intergenic 16S-23S rRNA spacer region of the pathovars. Xapas was shown to effectively detect all pv. passiflorae isolates and is recommended for disease diagnosis in passion fruit orchards.  相似文献

The effect of microwave heating on the oxidative stability of corn oil was determined by absorptivity in the UV spectrum and by peroxide and acid values. Oil samples with antioxidants BHA/BHT (1:1; 200 mg kg(-)(1)), with and without citric acid, were heated in a microwave oven (800 W, 2450 MHz) for 0-36 min. Absorptivity at 232 and 270 nm increased during microwave exposure. Control values of absorptivity at 232 nm increased from 3.568 to 12.874 after 36 min of heating. Peroxide value showed a significant difference in the initial stage of heating (0-6 min), but after this time, the peroxide value decreased due to the instability of hydroperoxides at high temperatures. Control 232 nm absorptivities after 6 days in the oven test were similar to those after 32-36 min of microwave heating. Effective antioxidants in the oven test did not show any protection during microwave heating. UV spectrophotometry is a suitable tool for microwave oxidation monitoring.  相似文献

In the search for new routes in the synthesis of hydroxylated kaurane diterpenoids, Verticillium lecanii, grown in a mineral liquid medium for 48 h, was fed with ent-17,19-dihydroxy-16betaH-kaurane, obtained by hydroboration/oxidation of kaurenoic acid, a natural product easily isolated from plants of the genera Xylopia (Annonaceae) and Wedelia (Asteraceae). After 14 days, the culture medium was extracted with ethyl acetate, and the metabolites were purified by column chromatography on silica gel. The results show that V. lecanii biotransformed the starting material into three novel compounds: ent-11alpha,17,19-trihydroxy-16betaH-kaurane; ent-7alpha,17,19-trihydroxy-16betaH-kaurane; and ent-7beta,17,19-trihydroxy-16betaH-kaurane, the structures of which were fully elucidated by using classical and modern two-dimensional NMR techniques.  相似文献

A method of determining total penicillins begins with an enzymatic hydrolysis of the beta-lactam ring to form their respective penicilloate product. Acetonitrile precipitates much of the casein and protein, which are then separated from the liquid by centrifugation. The lipids are removed from the aqueous fraction with methylene chloride. Mercuric chloride is added, which reacts with the penicilloate to liberate the side chain that has a terminal aldehyde. These penilloaldehyde products are extracted with methylene chloride and are subsequently reacted with dansyl hydrazine. The resulting fluorolabeled side chains are separated by liquid chromatography on a C18 column with acetonitrile-water as mobile phase. The fluorescence is measured by the mercury line at 254 nm excitation wavelength and a 500 nm filter on the emission side. The overall average recoveries from milk spiked at 25, 50, and 100 ppb are benzyl penicillin 79.4%; phenoxymethyl penicillin 59.7%; phenethicillin 75.9%; nafcillin 87.7%; methacillin 47.5%; oxacillin 57.6%; cloxicillin 37.3%; and dicloxicillin 26.4%.  相似文献
