The effects of Oesophagostomum dentatum infection and dietary carbohydrates on the morphology and epithelial cell proliferation in the gastrointestinal tract of pigs were investigated experimentally. Thirty-two worm-free pigs (n=32) from a specific pathogen-free farm were randomly divided into four groups (A-D), of eight animals each. Pigs in groups A (control) and B (infected) were fed Diet 1, and pigs in groups C (control) and D (infected) were fed Diet 2. The two diets were formulated: Diet 1 (%) contained barley flour, oat husk meal plus soya bean meal (55:21:24) and Diet 2 (%) contained barley flour, inulin and sugar beet fibre (SBF) (80.1:7:12.9) plus soya bean meal (3:1) to contain carbohydrates from inulin and sugar beet fibre (SBF) that were readily fermentable in the large intestine. The two infected pig groups (16 pigs total) were inoculated with 6000 infective larvae of O. dentatum and all pigs, including the controls, were slaughtered 12 weeks p.i. The combination of O. dentatum infection and highly fermentable dietary carbohydrates affected the mucosal architecture, the epithelial cell proliferation and mucin secretion of the large intestine. Infection had a significant influence on the crypt volume, height and density, and on muscularis externa at the proximal and middle colon. The changes in the affected gut sections were proportional to the number of worms present. However, these parameters appeared unaffected by those diets alone. Reliable ELISAs were investigated with the aim to select hen lines resistant to Salmonella Enteritidis and producing high levels of antibodies. In the first experiment, the relation between the humoral response and the bacteriological results was assessed on hens from the Y11 resistant line and the L2 susceptible line, orally inoculated with 10(8) CFU S. Enteritidis per animal. Anti-lipopolysaccharide (LPS) IgG titres were higher but the liver and spleen were less contaminated in hens from the Y11 line than in hens from the L2 line (p = 0.013, 0.031 and 0.026 respectively). In the second experiment, the hens were inoculated orally with 1.7 x 10(8) CFU S. Enteritidis per animal in order to select the ELISA methods showing the more significant differences. ELISAs were based on LPS, flagella, LPS from rough (LPS-R) and smooth strains (LPS-S) and detected IgG and IgM antibodies from sera and yolks. No between-line host response variation was observed in the yolk, with LPS-S and R antigens nor with anti-LPS IgM in the sera. Otherwise, significant differences were encountered between hen lines with the ELISAs performed on the sera detecting anti-LPS IgG, anti-flagella IgG or IgM (p = 0.017, 0.017 and p < 0.001 respectively). When comparing the kinetics of the selected ELISAs, the IgG antibodies against LPS detected between-line variations as early as 1 to 4 weeks pi, whereas with IgG against flagella, the differences were only detected at 1 and 2 weeks pi and with IgM against flagella, the differences were significant at 1, 2, 4 and 8 weeks pi. The concentration and composition of short-chain fatty acids (SCFA) in gastrointestinal and faecal samples were studied in weaning pigs fed different amounts and composition dietary fibre (DF). In Trial 1 a total of 50 castrated piglets were fed five experimental diets with varying contents of citrus pectin (soluble fibre) and barley hulls (insoluble fibre) and gastrointestinal contents were collected at euthanasia 9 days after weaning. In Trial 2, 120 pair-wise penned piglets were allocated to the same experimental diets as in Trial 1 (24 piglets per treatment), and fresh faecal samples were collected 5 and 32 days after weaning.
There was no difference in the concentrations of SCFA caused by dietary treatments, but across treatments there was a correlation between concentration of SCFA and the proportion of butyrate. The correlation was lowest in the caecum (r2 = 0.22, P = 0.0007) and highest in the distal colon (r2 = 0.45, P = 0.0001). In faeces there was no relationship 5 days after weaning (r2 = 0.004. P = 0.64) but a correlation of r2 = 0.25 (P = 0.0001) 32 days after weaning. The results suggest that adaptive changes and differences between piglets in fermentative capacity immediately after weaning are associated not only with a higher concentration of SCFA but also a changed fermentation profile. 相似文献
Sows were fed three diets varying in type and level of dietary fibre (DF). The low fibre diet (LF; 17% DF) was based on wheat and barley. In the two high DF diets (HF1, high in soluble DF and HF2, high in insoluble DF; 44% DF), the cereal part of the diet was substituted with different co-products (sugar beet pulp, potato pulp, pectin residue, pea hull, brewer's spent grain, and seed residue). The diets were fed for a four-week period to 12 sows (4 for each diet). Sows were stunned 4 h post-feeding, and digesta and tissue samples were collected from various parts of the small and large intestines. The carbohydrate load to the large intestine was 538–539 g/d when feeding the high DF diets and 190 g/d when feeding diet LF. Feeding sows the high DF diet containing large proportion of soluble DF resulted in a lower dry matter content of digesta (23 contra 28%), a higher tissue weight (2.9 contra 2.0 kg), and a higher crypt depth (492 contra 330 μm) and area (23,201 contra 15,751 μm2) in the colon compared with the low DF diet. In conclusion, increasing the amount of DF in the diet for sows resulted in an increased amount of digesta entering the large intestine which influences the functional properties of digesta. Furthermore, a high DF diet with a high proportion of soluble DF increases tissue weight, crypt depth and crypt area in the midcolon compared to a low fibre diet. 相似文献
The effects of weaning age, type of creep diet and the classification of piglets as ‘eaters’ or ‘non eaters’ of creep feed on weight gain to weaning were examined. Four antimicrobial-free diets were offered from day 14 of lactation: (i) wheat-soy based diet with animal and vegetable (‘mixed’) protein sources (COMM), (ii) heat-processed rice (HPR) with barley hulls and potato starch and mixed protein sources (RBPS), (iii) HPR with sugar-beet pulp and mixed protein sources (RSBP), and (iv) HPR with mixed protein sources (R). Pigs were weaned at an average of either 27 (early) or 33 (late) days of age. A dye (indigo carmine) was added to each diet at day 24 of lactation in both groups to classify piglets as ‘eaters’ or ‘non eaters’ of creep feed, based on appearance or non-appearance, respectively, of dye in faeces. Pigs weaned late weighed 1.4 kg more (P < 0.001). Weaning age interacted with diet type and eating classification for weaning weight (P < 0.001). Of the 1067 piglets where classification of the faecal dye was possible, 829 (77.7%) were ‘eaters’ and 238 (22.3%) were ‘non eaters’. This qualitative estimate of creep feed consumption did not influence weight gain to weaning (P > 0.05). 相似文献
