Insertional mutagenesis of the Drosophila genome with single P elements 总被引:58,自引:0,他引:58
A versatile genetic method for identifying and cloning Drosophila melanogaster genes affecting any recognizable phenotype is described. Strains are constructed in which the insertion of a single P transposable element has caused a new mutation, greatly simplifying the genetic and molecular analysis of the affected gene. Mutagenesis is initiated by crossing two strains, each of which contains a specially designed P element. One element (jumpstarter), encoding P element transposase, efficiently mobilizes the second nonautonomous transposon (mutator), whose structure facilitates selection and cloning of new insertion mutations. Random mutator transpositions are captured in individual stocks that no longer contain jumpstarter, where they remain stable. This method was used to construct 1300 single P element insertion stocks which were then screened for recessive mutations. A library of single-element insertion strains will allow the structure and function of Drosophila genes to be readily correlated, and should have many other applications in Drosophila molecular genetics. 相似文献

Kimberlite eruptions bring exotic rock fragments and minerals, including diamonds, from deep within the mantle up to the surface. Such fragments are rapidly absorbed into the kimberlite magma so their appearance at the surface implies rapid transport from depth. High spatial resolution Ar-Ar age data on phlogopite grains in xenoliths from Malaita in the Solomon Islands, southwest Pacific, and Elovy Island in the Kola Peninsula, Russia, indicate transport times of hours to days depending upon the magma temperature. In addition, the data show that the phlogopite grains preserve Ar-Ar ages recorded at high temperature in the mantle, 700 degrees C above the conventional closure temperature. 相似文献

Supplementing diets with l-Arginine (Arg) improves female reproductive performance and reproductive blood flow in other species. The objectives of this study were to investigate uterine artery blood flow changes before and after parturition, and evaluate blood flow in Arg supplemented and control mares by Doppler ultrasonography. Sixteen light-horse mares began Doppler ultrasonography evaluation, 21 days before expected foaling date (EFD) and continued until day 7 postparturition. The mares under treatment (n = 8) were supplemented with 100 g Arg, once daily, beginning with 21 days before EFD. Blood flow measurements were calculated as pulsatility index (PI) and resistance index (RI) for both uterine arteries, either ipsilateral or contralateral; to uterine horn of established pregnancy; defined gravid uterine artery (GUA) and non-gravid uterine artery (NGUA), respectively. The mares under treatment had a shorter gestation length (337 ± 1.7 days) as compared to control (345 ± 2.1 days; P ≤ .05). No differences in gestation length were observed between groups when examined by age, parity, EFD, or sex of foal. Both GUA and NGUA uterine artery diameter decreased from the day before parturition to day 7 after parturition (P ≤ .001). During this time period, both PI and RI increased (P ≤ .01); indicating less blood flow. A treatment effect was observed with Arginine-treated mares having greater blood flow prepartum in the NGUA (P ≤ .001) and postpartum in the GUA (P ≤ .05), for both indices. Understanding how comets work--what drives their activity--is crucial to the use of comets in studying the early solar system. EPOXI (Extrasolar Planet Observation and Deep Impact Extended Investigation) flew past comet 103P/Hartley 2, one with an unusually small but very active nucleus, taking both images and spectra. Unlike large, relatively inactive nuclei, this nucleus is outgassing primarily because of CO(2), which drags chunks of ice out of the nucleus. Detailed data on the momentum-resolved temperature dependence of the superconducting gap of Bi(2)Sr(2)CaCu(2)O(8+x) are presented, complemented by similar data on the intensity of the photoemission superconducting condensate spectral area. The gap anisotropy between the Gamma-Mand Gamma-X directions increases markedly with increasing temperature, contrary to what happens for conventional anisotropic-gap superconductors, such as lead. Specifically, the size of the superconducting gap along the Gamma-X direction decreases to values indistinguishable from zero at temperatures for which the gap retains virtually full value along the Gamma-M direction. Preliminary experiments indicate that tunable Pb(1-x)Sn(x)Te diode lasers will be useful in the identification and sensitive detection of most of the atmospheric pollutant gases. For point-sampling applications, concentrations in the parts-per-billion range should be measurable with very high specificity. For long-range atmospheric transmission techniques, the improved resolution capability and tunability of these diode lasers make them attractive replacements for spectrometers and fixed-frequency laser sources where operation at cryogenic temperatures is not a serious impediment. By using these lasers as tunable local oscillators in the infrared heterodyne configuration, remote passive detection of gases present in smokestack effluent appears possible. Finally, pulsed operation at temperatures available with simple cryogenic coolers permits immediate application to the fast detection of gases present in automobile exhaust and in chemical processing plants. 相似文献
