Endothelial cells are important for maintenance of vascular integrity by producing a variety of bioactive molecules such as nitric oxide (NO). Recent evidence has suggested that there are some differences in characteristics between endothelial cells from different origins. Here we examined responses of two typical endothelial cells to hepatocyte growth factor (HGF), which induces endothelium-dependent relaxation of microvessels. Stimulation of human umbilical vein endothelial cells (HUVEC) and bovine aortic endothelial cells (BAEC) with HGF increased endothelial NO synthase activity, accompanied with an increase of activity-related site-specific phosphorylation of protein kinase B/Akt. However, HGF stimulated phosphorylation of p38 mitogen-activated protein kinase (MAPK) only in HUVEC, but not in BAEC, while it induced phosphorylation of p44/p42 MAPK in both cells. These results suggest that HGF transduces different intracellular signals between aortic and umbilical venous endothelial cells, and that the differences might represent divergent endothelial responses to growth factors, especially those that activate receptor-tyrosine kinases.  相似文献

Fertility of dairy cows in Northern Ireland

A comprehensive database was established on the milk production and reproductive performance of dairy cows in 19 selected herds in Northern Ireland, varying in size, management system and genetic merit. Data were obtained for 2471 cows, 1775 of which calved in a second year, and 693 were culled from the herd for specific reasons. The estimated mean rate of heat detection (assessed by the interheat interval during the main breeding season) in all the herds was 71 per cent, with a range from 53 to 92 per cent The average conception rate to first insemination was 37.1 per cent (range 21 to 66 per cent). The average calving interval for the retained cows was 407.2 days (range 359 to 448 days). Twenty-eight per cent of the cows that calved were culled, with infertility being the largest single reason (26.8 per cent of the cows culled). There were major differences in reproductive performance between the herds, but heat detection rate, conception rate and calving interval did not appear to be affeded by a herd's genetic merit. The herds with shorter calving intervals were characterised by better heat detection efficiency (83 v 61 per cent, P<0.01), a shorter interval from calving to first insemination (74 v 97 days; P<0.05), a higher conception rate to first insemination (45 v 34 per cent, P>0.10) and a lower removal rate (23 v 37 per cent, P<0.01). Furthermore, the cows in these herds had lower body condition scores (BCS) in the dry period (3.0 v 3.3; P<0.05) but lost less body condition in early lactation (0.3 v 0.6 BCS units, P<0.05). These results show that dairy herd fertility in Northern Ireland is generally low and similar to that previously reported for England and the USA, but that in some herds changes in herd management practices improved the cows' fertility.  相似文献

Virucidal efficacy was evaluated for four recently available disinfectants: chlorine dioxide, potassium peroxymonosulfate, a quaternary ammonium compound, and citricidal (grapefruit extract). Sodium hypochlorite (3%) and tap water were used as positive and negative controls respectively. Feline herpesvirus, feline calicivirus, and feline parvovirus were exposed to the manufacturers' recommended dilutions of the evaluated disinfectants. Both chlorine dioxide and potassium peroxymonosulfate completely inactivated the three viruses used in this study. These disinfectants can aid in controlling nosocomial transmission of viruses with less of the deleterious effects of sodium hypochlorite. The quaternary ammonium compound evaluated in this study and citricidal were not effective against feline calicivirus and feline parvovirus.  相似文献   

Regional brain monoamine concentrations and their alterations in bovine hypomagnesaemic tetany experimentally induced by a magnesium-deficient diet

Monoamines are important brain neurotransmitters. An investigation was carried out to determine if hypomagnesaemic tetany was associated with alterations in regional brain monoamine concentrations in bovines. The results, established in cows with normal magnesium status, demonstrated that regional differences existed in the distribution and concentration of brain monoamines in the adult bovine, which were similar to those in other species. In magnesium-deficient cows, severe hypomagnesaemia and lowered cerebrospinal fluid (CSF) magnesium concentrations were associated with significant alterations in monoamine concentrations in some brain regions. Alterations in 3,4-dihydroxyphenylalanine (DOPA) and dihydroxyphenylacetic acid (DOPAC) concentrations in the corpus striatum, and dopamine (DA) in the cerebral cortex and cerebellum were recorded. These regions play an important role in both voluntary and involuntary motor function, and therefore these alterations may play a role in the aetiology of hypomagnesaemic tetany. However, there was no significant change in DA concentrations in the corpus striatum (the main dopaminergic region in the brain) associated with hypomagnesaemia. In addition, a significantly lower norepinephrine (NE) concentration in the corpus striatum of hypomagnesaemic animals was also recorded. Norephinephrine is generally excitatory and therefore lowered NE concentrations would be expected to result in depression rather than stimulation of motor function.  相似文献   

Skeletal muscle lymphoma in a bullmastiff

A 16-month-old, neutered male bullmastiff was presented for acute onset of massive swelling of the right hind limb. Primary skeletal muscle lymphoma was diagnosed based on cytopathology, surgical biopsy, and necropsy findings. Cutaneous metastases developed during the hospitalization, and additional metastases were found in the heart and thoracic wall. Primary skeletal muscle lymphoma is a rare form of lymphoma in dogs and should be considered as a differential diagnosis for acute, soft-tissue swelling of the limb.  相似文献

The effects of field aging (0–28 days) and pheromone loading rate on the longevity of red rubber septa loaded with the sex pheromone blend of the oriental fruit mothGrapholita molesta (Busck), were evaluated in North Carolina apple orchards in 2002. Separate field tests examined the influence of trap height and pheromone loading rate of rubber septa on trap catches of adultG. molesta males in an abandoned orchard. The loss of the major pheromone component, (Z)-8-dodecenyl acetate (Z8–12:OAc), from red rubber septa over a 4-week period exhibited a relatively constant release rate with 30, 100 and 300 μg pheromone. Trap catch was significantly higher in pheromone traps placed in the upper canopy than in those in the lower canopy. Pheromone traps baited with 100μg lures caught more moths compared with those loaded with 300 μg. Ninety-eight dogs with lymphoma treated with a 5-drug combination chemotherapy regimen (vincristine, L-asparaginase. cyclophosphamide, doxorubicin, prednisone [VELCAP-L]) were evaluated for pretreatment characteristics predictive for response and remission duration. The complete remission rate was 69%, with a median remission duration of 55 weeks. Dogs with advanced stage of disease, constitutional signs, dogs that were older, and dogs that were dyspneic were less likely to achieve remission. Once in remission, small dogs and dogs without pretreatment thrombocytopenia were likely to have longer remission duration. Toxicoses were frequent, but rarely fatal, and no predictitive factors were found for a dog developing toxicoses. VELCAP-L is an effective treatment for dogs in stage I-III lymphoma, particularly in young, small animals.  相似文献   

Fibrocartilaginous emboli in a tayra (Eira barbara): a case report.

An 8-yr-old male tayra (Eira barbara) was presented with acute onset of pelvic limb paralysis. Radiography was unremarkable. Neurologic examination showed signs consistent with an intramedullary lesion between the second thoracic and fifth lumbar spinal cord segments. The animal's condition did not improve after 4 days of aggressive glucocorticoid therapy, and euthanasia was performed. Histologic examination of the spinal cord showed amorphous emboli suggestive of cartilaginous fragments within spinal veins. A diagnosis of fibrocartilaginous emboli was made, the first known case in a mustelid.  相似文献
