The sequences of a small RNA segment of Aino virus isolates were analyzed to define the molecular epidemiology and genetic relationships to other species in the genus Orthobunyavirus in the family Bunyaviridae. The nucleotide and amino acid sequences of the segment were highly conserved among strains isolated from 1964 to 2002 in Japan. These Japanese isolates were segregated into two distinct lineages, one containing the prototype strain JaNAr28 isolated in 1964 and the other containing strains isolated after 1986, by phylogenetic analysis based on the nucleocapsid gene sequences. Japanese strains isolated after 1986 were rather more closely related to Kaikalur virus isolated in India in 1971 than to strain JaNAr28. On the other hand, an Australian strain, B7974, was closely related to Peaton virus. The B7974 strain might have been generated by inter-serotype genetic reassortment between Aino and Peaton viruses in Australia during their evolution. However, recent Aino virus strains isolated in Japan appear to be genetically stable. 相似文献

To investigate in vitro differentiation of canine adipose tissue-derived stromal cells (ATSCs) into neuronal cells, ATSCs from celiac adipose tissue in clinically healthy beagle dogs were treated with 100 muM dibutyryl cyclic adenosine monophosphate (dbcAMP) and 125 muM isobuthylmethylxanthine (IBMX). ATSCs were morphologically changed into differentiated ATSCs from spindle-shaped cells to neuron-like cells with numerous processes after the treatment. Expression of neuron-specific enolase (NSE) as an early neuron specific marker protein was detected in both ATSCs and differentiated ATSCs, however diachronic increase of NSE expression was observed in differentiated ATSCs after the treatment with dbcAMP/IBMX. In addition, neurofilament-68 (NF-68) as an early to mature neuron specific marker protein was weakly expressed in differentiated ATSCs. Neuron specific glutamate and glucose transporter (EAAC1 and GLUT-3, respectively) mRNAs were strongly expressed in differentiated ATSCs compared with those in ATSCs, although glia specific glutamate transporter mRNA (GLT-1) was also detected in differentiated ATSCs. ATSCs can differentiate into early to mature neuronal cells and are candidate cells for autologous nerve regeneration therapy, although additional research is needed to examine functional characteristics of differentiated ATSCs. 相似文献

We report a case of mixed epithelial and stromal tumor of the kidney (MESTK) in a 32-week-old heterozygous sphingosine 1-phosphate-2 (S1P2) receptor deficient female mouse. A white solid mass replacing the left kidney was observed at the left retroperitoneal wall. Histologically, the tumor mass consisted of dimorphic cellular components of epithelial and stromal cells. Epithelial cells formed various sized irregular-shaped tubular structures resembling renal tubules surrounded by stromal cells. Immunohistochemically, epithelial cells were positive for cytokeratin, while stromal cells showed positive immunoreactivity with alpha-smooth muscle actin as well as vimentin. The oomycete Phytophthora infestans is the causal agent of potato late blight, one of the most destructive and historically significant pathogens in agricultural production. A virus-induced gene silencing-based screening of the solanaceous model plant N. benthamiana resulted in revealing a wide range of resistance mechanisms of solanaceous plants against this pathogen. In this article, we present an overview of the various pathways involved in the N. benthamiana–P. infestans pathosystem, including some of the follow-up work that was triggered by these findings. The purpose of this review is to assemble these findings and integrate them into our current understanding of plant pathogen defense mechanisms and discuss their potential application for the development of potato resistance to P. infestans. 相似文献

There are a number of reports documenting skeletal development in groupers, but surprisingly little is known about muscle development. We have documented muscle development in the seven-band grouper Epinephelus septemfasciatus (Thunberg), focusing primarily on the cranial muscles from 0 days post-hatching (dph) to 28 dph, using a modified whole mount immunohistochemical staining method. The eye muscles developed first followed by the cheek muscles, which completed development prior to first feeding. The muscles associated with the opercle and the levator internus 1 developed gradually and were not linked to first feeding. Later, between 12 and 16 dph, the muscle composition in the dorsal branchial arches changed to the adult form. We observed a number of cultured larvae that exhibited abnormal musculature in the body and jaw. Our results provide the first documentation of muscle development in larval seven-band grouper. This information can be used to detect abnormalities during development and provide a marker to assess the effect of changes in larval rearing techniques. 相似文献

of LDL-C (P = 0.041) and TC (P = 0.037) during the trial. Significant differences between the test and placebo groups were found for the visceral fat (P = 0.039), BMI (P = 0.015), and waist circumference (P = 0.011) at the end point. Aqueous solutions of gellan gum with comparable molecular masses but with different acyl contents were investigated by atomic force microscopy and rheological measurements in the presence or absence of various cations. For a high-acyl sample, no continuous network structures were identified microscopically, except in the presence of Ca (2+), where structural inhomogeneity was the highest in terms of the height distribution of molecular assemblies. Rheological thermal hysteresis between sol-gel transitions was detected in the presence of K (+) and Ca (2+), particularly Ca (2+). The storage modulus at 20 degrees C was larger in the order Na (+) < Ca (2+) < K (+). For a low-acyl sample, continuous network structures were identified in the presence of each cation, involving greater thermal hysteresis than the corresponding data for the high-acyl sample. Structural homogeneity was the highest in the presence of K (+). Thermal hysteresis and elasticity of the system were discussed in terms of continuousness and homogeneity of network structures. 相似文献

The maternal-fetal and neonatal transfers of polychlorinated dibenzo- p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs), and dioxin-like polychlorinated biphenyls (PCBs) were measured in the blood and milk of dams and in the blood of newborn and suckling calves. Calf blood toxic equivalent quantities (TEQs) were drastically increased by suckling. Blood concentrations of individual congeners were greater in suckling calves than in newborn calves, excluding octachlorodibenzo- p-dioxin (O 8CDD); O 8CDD did not readily transfer to milk but was readily absorbed by the gastrointestinal tract and remained in systemic circulation longer than other congeners. Congener concentrations in milk were correlated with maternal blood levels, and those in suckling calf blood were dependent on their concentrations in milk. These results suggest that neonatal calves absorb more lipophilic organochlorine compounds than prenatal fetuses, that those compounds that are transferred from dams to calves are associated not only with lipid transport but also with other carriers, and that the distribution of congeners is structure-dependent. 相似文献
