黄土高原黄土团粒组成及其与碳酸钙关系的研究   总被引:13,自引:1,他引:13       下载免费PDF全文
通过团粒分析、电子显微镜 (SEM/EDX)扫描等元素分析扫描方法和手段 ,对黄土高原西北部自然黄土中的团粒数量及其与碳酸钙的溶解量的关系、团粒表面和内部碳酸钙的分布进行了研究。结果表明 :碳酸钙大约参与了 99%的团粒形成 ,呈现团粒越大 ,碳酸钙含量越多的趋势 ;反之 ,团粒数量与碳酸钙的溶解量成反比 ;随着团粒表面碳酸钙分布的减少 ,附着在团粒表面的微团粒及粉粒、粘粒减少 ;在团粒内部 ,碳酸钙与矿物颗粒之间存在 3种胶结方式 ,分别为碳酸钙附着在矿物颗粒表面、碳酸钙分布于矿物颗粒之间以及碳酸钙表面附着矿物颗粒。实验结果证明了黄土团粒的主要胶结物是碳酸钙这一理论。  相似文献

Viruses exist everywhere on the planet. Recent development in viral genomics confirmed that genomic information is preserved among viral subsets and can be used for phylogenetic classification of viruses and for evaluation of viral diversity in the environment. The capsid gene of T4-type bacteriophages, g23, is the most widely applied gene for evaluating the diversity of the T4-type bacteriophage family. In this study, we applied denaturing gradient gel electrophoresis to PCR products of DNA with g23-specific primers that were extracted from a Japanese paddy field under long-term fertilizer trial and obtained 39 different g23 clones at the DNA level. They showed identities of 27–99% with the clones within the NCBI database at the amino acid level. They were quite distinctive from those obtained in marine environments and most of them formed six phylogenetically novel groups in the T4-type bacteriophage family with the clones obtained from another paddy field. The existence of six novel groups was confirmed from molecular analysis of all the amino acid sequences between the primers, of the amino acid sequences excluding hypervariable region, and of those of conserved regions. These findings indicate that T4-type bacteriophage communities in paddy fields consist of previously uncharacterized members phylogenetically distant from those in marine environments. The type of fertilizers and the stage during rice cultivation were not the major factors in determining T4-type bacteriophage communities in the paddy field.  相似文献

Potassium chlorate is widely used as an active substance for flower induction in longan plantation fields for off-season production of longan fruits in northern Thailand. Contamination of groundwater with residual chlorate in soil is a cause for concern because of its toxicity to human health. Based on our previous finding that the addition of glucose or sucrose to soil was effective for accelerating the disappearance of residual chlorate in soil, the effect of the addition of molasses, which contains a high concentration of sucrose, as a substitute for glucose or sucrose was examined in laboratory and pot experiments. Under laboratory conditions, the addition of molasses to soil at the concentrations of 100 to 200 g kg−1 soil strikingly enhanced the rate of disappearance of chlorate applied at 341 mg kg−1 soil. Addition of diluted molasses was also effective for the accelerated disappearance of chlorate in soil when 33 g kg−1 soil of molasses was added repeatedly. The effect of repeated addition of diluted molasses to soil on the decontamination of residual chlorate in soil was also confirmed in an outdoor pot experiment. These results may lead to the development of a practical method of cleaning-up chlorate-polluted soil in longan plantation fields.  相似文献

The present study examined T4-type phage communities in rice straw (RS) under the composting process by analyzing the composition of the major capsid gene ( g23 ) of T4-type bacteriophages. The g23 clones were obtained from RS throughout the composting process from RS materials to composting RS in the curing stage (for 124 days). Most of the g23 clones were phylogenetically closely related to those in rice field soils and rice field floodwaters, and Paddy Groups II and III appeared to characterize the g23 genes in the composting RS. The diversity of g23 genes in the composting RS was highest in the RS material (day 0 after the onset of composting) and in the early thermophilic stage (day 7), and decreased markedly in the middle and curing stages. This change was in contrast to that of the bacterial community, which showed higher diversity in the middle and curing stages. There was no specific clone that characterized any stage during the composting process. These findings indicate that the phage community is not the major controlling agent in determining eubacterial succession and that the thermophilic stage in the composting process efficiently annihilated T4-type phages in the composting pile.  相似文献

This research was carried out to study the characteristics and the potential utilization of coal fly ash–based synthetic aggregates (CSA) with oil palm waste as an alternative container substrate for ornamental‐plant production. CSA only, oil palm waste only, and two mixing ratios of CSA with oil palm waste at the ratio of 1:5 and 1:10 (V/V) were utilized under this study. Zeolite was utilized as a standard substrate to compare characteristics of other substrates. The physical and chemical properties of all substrates were characterized. Scanning electron microscopy (SEM) of coal fly ash and CSA were conducted in order to study the structural configuration of the CSA. Developed CSA gave an alkaline pH (9.82), high electrical conductivity (96.1 mS m–1), high cation concentrations, high water‐holding capacity, and low bulk density (0.56 g cm–3) compared to zeolite. Mixing of CSA with oil palm waste at the ratio of 1:10 gave enhanced physical and chemical properties such as bulk density (0.25 g cm–3), particle density (1.76 g cm–3), air space (20.6%), total pore space (85.8%), total water‐holding capacity (652 mL L–1), pH (6.18), and electrical conductivity (42.4 mS m–1), which were in the established ideal substrate range. Moreover, SEM study revealed that CSA is a dual‐composite material, which had well enmeshed coal–fly ash particles in the fibrous paper‐waste matrix creating porous spaces within the aggregate. The growth of French marigold (Tagetes patula), which is a popular ornamental plant in Japan, was assessed using these newly developed substrates. The mixing ratio of CSA and oil palm wastes at 1:10 reported the best maximum growth and yield parameters of French marigold, with increase in shoot fresh weight, shoot dry weight, root fresh weight, root dry weight, plant height, and number of flowers per plant by 51%, 93%, 54%, 150%, 19%, and 61%, respectively, compared to the zeolite. It is revealed that a mixture of CSA and oil palm waste at the ratio of 1:10 can be successfully utilized as an alternative container substrate for French marigold production.  相似文献

The polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and PCR-denaturing gradient gel electrophoresis (DGGE) patterns of 16S rDNA were studied to elucidate the effects of the type of fertilization and the growth stage of rice plants on the community structure of the microbiota in the floodwater of a Japanese paddy field under a long-term fertilizer trial. From the mid tillering stage, a higher pH and temperature were observed in the plot without fertilization (NoF plot) than in the plots supplied with chemical fertilizers (CF plot) and with compost (CM plot). DNA fragments specific to the respective plots and common to every plot were detected after the digestion of PCR products by restriction enzymes. Cluster analysis separated the RFLP and DGGE patterns of the microbiota in the floodwater into four clusters; the microbiota in (1) the NoF plot, (2) the CF plot, (3) the CM plot, and (4) the CF and CM plots in the early growing stage. The effect of fertilizer application on the community structure was more conspicuous than that of seasonal variation.  相似文献

Vibrios are highly motile, facultatively anaerobic bacteria that are ubiquitous in aquatic environments and part of the normal intestinal microflora of healthy fish, but some species can cause vibriosis. The adherence of vibrios to host fish intestines is a significant event not only for their survival and growth, but also in terms of pathogenicity. However, the molecular mechanism underlying the adhesion of vibrios to the intestinal tract of fish is not fully understood. We report here the identification of intestinal glycosphingolipid (GSL) receptors to which pathogenic vibrios attach in typical mariculture fish. Thin-layer chromatography overlay assays using five species of 35S-labeled vibrios and intestinal glycosphingolipids of seven species of mariculture fish revealed that all of the fish tested possessed GM3 (NeuAcα2-3Galβ1-4Glcβ1-1′Cer) and/or GM4 (NeuAcα2-3Galβ1-1′Cer) as major acidic intestinal GSLs and that all of the vibrios tested specifically adhered to GM3 and/or GM4. Our results demonstrate that these GSLs were able to function as a receptor for the various vibrios tested. Analysis of the relationship between sugar structure and receptor activity for vibrios revealed that ‘NeuAcα2-3Galβ1-’ is required at the non-reducing end of glycosphingolipids for the bacteria to attach.  相似文献

We isolated two different genomic DNAs (UprbcS1 and UprbcS2) encoding the small subunit of ribulose-1,5-bisphosphate carboxylase/oxygenase and portions of the 5′- and 3′-flanking regions from sterile Ulva pertusa Kjellman. The UprbcS1 and UprbcS2 genes had three introns in the coding region. Each predicted UprbcS polypeptide was a 180-amino-acid (AA) residue including a 38-AA transit peptide, although the 104th AA residue was replaced. The nucleotide sequences of UprbcS cDNAs isolated from a cDNA library corresponded to that of the UprbcS1 gene, suggesting that the UprbcS1 gene was predominantly expressed in sterile U. pertusa compared to UprbcS2. Southern blot analysis showed that each UprbcS gene was a single-copy gene in the sterile U. pertusa genome. Northern hybridization indicated that the expression of UprbcS was induced and repressed by dark and light treatments, respectively. When sterile U. pertusa cells were transformed with an expression vector containing the UprbcS1 promoter and terminator sequences fused with the green fluorescent protein (GFP) gene, GFP fluorescence was observed in the cells transformed. These results suggest that the UprbcS1 gene promoter is light regulated and highly active in the sterile U. pertusa cells and is available for genetic transformation system in the alga.  相似文献

ABSTRACT:   Feeding activity was examined at high temperatures by using a demand feeder for thermally selected and normal rainbow trout Oncorhynchus mykiss strains. When the water temperature was raised in experiment 1 from 17.5 to 25.7°C at 0.3°C/day in 21 days, the average daily food consumption rate in the thermally selected strain rose to 7.1%, which was significantly higher than that of the normal strain (4.1%, P  < 0.05). The corresponding rate was also significantly higher in the thermally selected (0.8%) than in the normal strain (0.2%, P  < 0.05) of fish in experiment 2 where water temperature was raised to 24.1°C in 0.5°C/day increments. When water temperature was raised rapidly in experiment 3 from 16.7 to 21.7°C in one day and gradually to 24.4°C in 28 days at 0.1°C/day, the average daily food consumption rates were 1.0 and 0.1% for the thermally selected and normal strains, respectively, with significant difference ( P  < 0.01). These results suggest that the thermally selected strain has acquired thermal tolerance as a result of artificial selection.  相似文献

The dough properties and baking qualities of a novel high‐amylose wheat flour (HAWF) and a waxy wheat flour (WWF) (both Triticum aestivum L.) were investigated by comparing them with common wheat flours. HAWF and WWF had more dietary fiber than Chinese Spring flour (CSF), a nonwaxy wheat flour. Also, HAWF contained larger amounts of lipids and proteins than WWF and CSF. There were significant differences in the amylose and amylopectin contents among all samples tested. Farinograph data showed water absorptions of HAWF and WWF were significantly higher than that of CSF, and both flours showed poorer flour qualities than CSF. The dough of WWF was weaker and less stable than that of CSF, whereas HAWF produced a harder and more viscous dough than CSF. Differential scanning calorimetry data showed that starch in HAWF dough gelatinized at a lower temperature in the baking process than the starches in doughs of WWF and CSF. The starch in a WWF suspension had a larger enthalpy of gelatinization than those in HAWF and CSF suspensions. Amylograph data showed that the WWF starch gelatinized faster and had a higher viscosity than that in CSF. The loaves made from WWF and CSF were significantly larger than the loaves made from HAWF. However, the appearance of bread baked with WWF and HAWF was inferior to the appearance of bread baked with CSF. Bread made with WWF became softer than the bread made with CSF after storage, and reheating was more effective in refreshing WWF bread than CSF bread. Moreover, clear differences in dough and bread samples were revealed by scanning electron microscopy. These differences might have some effect on dough and baking qualities.  相似文献
