Zimbabwe’s Mid-Zambezi Valley is of global importance for the emblematic mega-fauna of Africa. Over the past 30 years rapid land use change in this area has substantially reduced wildlife habitat. Tsetse control operations are often blamed for this. In this study, we quantify this change for the Dande Communal Area, Mbire District, of the Mid-Zambezi Valley and analyse the contribution of three major potential drivers: (1) increase in human population; (2) increase in cattle population (and the expansion of associated plough-based agriculture), and; (3) expansion of cotton farming. Although direct effects of land use change on wildlife densities could not be proven, our study suggests that the consequences for elephant and buffalo numbers are negative. All three of the above drivers have contributed to the observed land use change. However, we found farmland to have expanded faster than the human population, and to have followed a similar rate of expansion in cattle sparse, tsetse infested areas as in tsetse free areas where cattle-drawn plough agriculture dominates. This implies the existence of a paramount driver, which we demonstrate to be cotton farming. Contrary to common belief, we argue that tsetse control was not the major trigger behind the dramatic land use change observed, but merely alleviated a constraint to cattle accumulation. We argue that without the presence of a cash crop (cotton), land use change would have been neither as extensive nor as rapid as has been observed. Therefore, conservation agencies should be as concerned by the way people farm as they are by population increase. Conserving biodiversity without jeopardising agricultural production will require the development of innovative technological and institutional options in association with policy and market interventions.  相似文献

This study aimed at developing a sensitive LC-MS/MS method for the quantification of sulforaphane (SFN) and indole-3-carbinol metabolites in plasma and urine after dietary intake of regular and selenium-fertilized broccoli using stable isotope dilution analysis. In a three-armed, placebo-controlled, randomized human intervention study with 76 healthy volunteers, 200 g of regular (485 μg of total glucosinolates and <0.01 μg of selenium per gram fresh weight) or selenium-fertilized broccoli (589 μg of total glucosinolates and 0.25 μg of selenium per gram fresh weight) was administered daily for 4 weeks. Glucoraphanin and glucobrassicin metabolites quantified in plasma and urine were SFN-glutathione, SFN-cysteine, SFN-cysteinylglycine, SFN-acetylcysteine, and indole-3-carboxaldehyde, indole-3-carboxylic acid, and ascorbigen, respectively. Dietary intake of selenium-fertilized broccoli increased serum selenium concentration analyzed by means of atomic absorption spectroscopy by up to 25% (p < 0.001), but affected neither glucosinolate concentrations in broccoli nor their metabolite concentrations in plasma and urine compared to regular broccoli.  相似文献

Pseudomonas CMR12a was previously selected as an efficient biocontrol strain producing phenazines and cyclic lipopeptides (CLPs). In this study, biocontrol capacity of Pseudomonas CMR12a against Rhizoctonia root rot of bean and the involvement of phenazines and CLPs in this ability were tested. Two different anastomosis groups (AGs) of Rhizoctonia solani, the intermediately aggressive AG 2-2 and the highly aggressive AG 4 HGI, were included in growth-chamber experiments with bean plants. The wild-type strain CMR12a dramatically reduced disease severity caused by both R. solani AGs. A CLP-deficient and a phenazine-deficient mutant of CMR12a still protected bean plants, albeit to a lesser extent compared with the wild type. Two mutants deficient in both phenazine and CLP production completely lost their biocontrol activity. Disease-suppressive capacity of CMR12a decreased after washing bacteria before application to soil and thereby removing metabolites produced during growth on plate. In addition, microscopic observations revealed pronounced branching of hyphal tips of both R. solani AGs in the presence of CMR12a. More branched and denser mycelium was also observed for the phenazine-deficient mutant; however, neither the CLP-deficient mutant nor the mutants deficient in both CLPs and phenazines influenced hyphal growth. Together, results demonstrate the involvement of phenazines and CLPs during Pseudomonas CMR12a-mediated biocontrol of Rhizoctonia root rot of bean.  相似文献

A greenhouse experiment was conducted by growing oats (Avenasativa L.) in a continuously 13CO2 labeled atmosphere. The allocation of 13C-labeled photosynthates in plants, microbial biomass in rhizosphere and root-free soil, pools of soil organic C, and CO2 emissions were examined over the plant's life cycle. To isolate rhizosphere from root-free soil, plant seedlings were placed into bags made of nylon monofilament screen tissue (16 μm mesh) filled with soil. Two peaks of 13C in rhizosphere pools of microbial biomass and dissolved organic carbon (DOC), as well as in CO2 emissions at the earing and ripeness stages were revealed. These 13C maxima corresponded to: (i) the end of rapid root growth and (ii) beginning of root decomposition, respectively. The δ13C values of microbial biomass were higher than those of DOC and of soil organic matter (SOM). The microbial biomass C accounted for up to 56 and 39% of 13C recovered in the rhizosphere and root-free soil, respectively. Between 4 and 28% of 13C assimilated was recovered in the root-free soil. Depending on the phenological stage, the contribution of root-derived C to total CO2 emission from soil varied from 61 to 92% of total CO2 evolved, including 4-23% attributed to rhizomicrobial respiration. While 81-91% of C substrates used for microbial growth in the root-free soil and rhizosphere came from SOM, the remaining 9-19% of C substrates utilized by the microbial biomass was attributable to rhizodeposition. The use of continuous isotopic labelling and physical separation of root-free and rhizosphere soil, combined with natural 13C abundance were effective in gaining new insight on soil and rhizosphere C-cycling.  相似文献

A method for the determination of total N-nitroso compounds (NOC) by chemical denitrosation and subsequent chemiluminescence detection of evolved NO is described. Denitrosation was accomplished with CuCl in HCl at 70 degrees C. The detection limit for N-nitrosoproline (NPRO) was 1 pmol. NO formation from NPRO was linear (R(2) = 0.999) from 4 pmol to 2 nmol. Among the possible interfering compounds tested, only S-nitroso compounds contribute any significant interference. This method had several advantages over other similar methods: (1) A commercially available one-piece reaction vessel and a NO analyzer with software were used. (2) NO release occurred rapidly and was easily measured and quantified. (3) Compared to HBr or HI, CuCl was more convenient to work with and safe. (4) CuCl was suitable for samples in aqueous and most organic solvents. The application of this method to food, personal care products, and human body fluids demonstrates its utility.  相似文献

Pectin methylesterase (PME) from green bell peppers (Capsicum annuum) was extracted and purified by affinity chromatography on a CNBr-Sepharose-PMEI column. A single protein peak with pectin methylesterase activity was observed. For the pepper PME, a biochemical characterization in terms of molar mass (MM), isoelectric points (pI), and kinetic parameters for activity and thermostability was performed. The optimum pH for PME activity at 22 degrees C was 7.5, and its optimum temperature at neutral pH was between 52.5 and 55.0 degrees C. The purified pepper PME required the presence of 0.13 M NaCl for optimum activity. Isothermal inactivation of purified pepper PME in 20 mM Tris buffer (pH 7.5) could be described by a fractional conversion model for lower temperatures (55-57 degrees C) and a biphasic model for higher temperatures (58-70 degrees C). The enzyme showed a stable behavior toward high-pressure/temperature treatments.  相似文献

This article deals with the potential of Fourier transform (FT) Raman spectroscopy in discrimination of botanical species of green and roasted coffees. There are two species of commercial importance: Coffea arabica (arabica) and Coffea canephora (robusta). It is recognized that they differ in their lipid fraction, especially in the content of the diterpene kahweol, which is present at 0.1-0.3% dry matter basis in arabica beans and only in traces (<0.01%) in robusta. The visual examination of the Raman spectra of the lipid fraction extracted from arabica, robusta and liberica samples shows differences in the mid-wavenumbers region: arabica spectra have two characteristic scattering bands at 1567 and 1478 cm(-1). The spectrum of the pure kahweol shows the same bands. Principal component analysis is applied to the spectra and reveals clustering according to the coffee species. The first principal component (PC1) explains 93% of the spectral variation and corresponds to the kahweol concentration. Using the PC1 score plot, two groups of arabica can be distinguished as follows: one group with high kahweol content and another group with low kahweol content. The first group includes samples coming from Kenya and Jamaica; the second group includes samples from Australia. The main difference between these coffees is that those from Kenya and Jamaica are well-known for growing at a high altitude whereas those ones from Australia are grown at a low altitude. To our knowledge, the application of Raman spectroscopy has never been used in coffee analysis.  相似文献

Glyphosate-tolerant (Roundup Ready) corn line GA21 has been developed by genetic modification to tolerate glyphosate, the active ingredient in Roundup herbicide. The purpose of this study was to evaluate the compositional and nutritional safety of corn line GA21 compared to that of conventional corn. Compositional analyses were conducted to measure proximate, fiber, amino acid, fatty acid, and mineral contents of grain and proximate, fiber, and mineral contents of forage collected from 16 field sites over two growing seasons. The nutritional safety of corn line GA21 was evaluated in a poultry feeding study conducted with 2-day old, rapidly growing broiler chickens, at a dietary concentration of 50-60% w/w. Compositional analysis results showed that, except for a few minor differences that are unlikely to be of biological significance, the grain and forage of GA21 corn were comparable in their composition to that of the control corn line and to conventional corn. Results from the poultry feeding study showed that there were no differences in growth, feed efficiency, adjusted feed efficiency, and fat pad weights between chickens fed with GA21 grain or with parental control grain. These data taken together demonstrate that Roundup Ready corn is as safe and nutritious as conventional corn for food and feed use.  相似文献

Particle transport often plays an important role in trace metal cycling in aquatic systems. In this study, we propose a simple method to estimate total particulate Hg in water using combustion as a treatment prior to analysis as opposed to conventional chemical digestion. We also present a field validation of this method in a stream. We sampled water from a stream 24 and 48 h after a heavy storm to obtain two sets of water samples with contrasting particulate Hg levels. Measurements of particulate and dissolved Hg using our new direct combustion method compared favorably with those of total Hg in unfiltered samples (112 ± 15 %). U.S. E.P.A. standard method 1631 gave similar dissolved Hg levels but different particulate Hg levels (calculated as the difference between total and dissolved Hg concentrations) than those obtained with our new method. We suggest that this discrepancy is explained by the propagation of errors resulting from the several calculation steps used in the standard method. Our new method gives reliable results, without labor intensive and costly chemical digestion and analysis of filters.  相似文献

The European chicken meat market is characterized by numerous quality marks: "Label de Qualité Wallon" in Belgium, "Label Rouge" in France, denominations of geographical origin, organic agriculture, etc. Most of those certified productions have specifications requiring the use of slow-growing chicken strains. The amplified fragment length polymorphism (AFLP) technique has been used to search molecular markers able to discriminate slow-growing chicken strains from fast-growing ones and to authenticate certified products. Two pairs of restriction enzymes (EcoRI/MseI and EcoRI/TaqI) and 121 selective primer combinations were tested on individual DNA samples from chicken products essentially in carcass form that were ascribed as belonging to either slow- or fast-growing strains. Within the resulting fingerprints, two fragments were identified as type-strains specific markers. One primer combination gives a band (333 bp) that is specific for slow-growing chickens, and another primer pair generates a band (372 bp) that was found to be characteristic of fast-growing chickens. The two markers were isolated, cloned, and sequenced. The effectiveness and the specificity of the two interesting determinants were assessed on individuals of two well-known strains (ISA 657 and Cobb 500) and on commercialized products coming from various origins.  相似文献
