Climate warming and associated increases in nutrient mineralization may increase the availability of soil nitrogen (N) in high latitude ecosystems, such as boreal forests. These changes in N availability could feed back to affect the decomposition of litter and organic matter by soil microbes. Since fungi are important decomposers in boreal forest ecosystems, we conducted a 69-day incubation study to examine N constraints on fungal decomposition of organic substrates common in boreal ecosystems, including cellulose, lignin, spruce wood, spruce needle litter, and moss litter. We added 0, 20, or 200 μg N to vials containing 200 mg substrate in factorial combination with five fungal species isolated from boreal soil, including an Ascomycete, a Zygomycete, and three Basidiomycetes. We hypothesized that N addition would increase CO2 mineralization from the substrates, particularly those with low N concentrations. In addition we predicted that Basidiomycetes would be more effective decomposers than the other fungi, but would respond weakly or negatively to N additions. In support of the first hypothesis, cumulative CO2 mineralization increased from 635 ± 117 to 806 + 108 μg C across all fungal species and substrates in response to 20 μg added N; however, there was no significant increase at the highest level of N addition. The positive effect of N addition was only significant on cellulose and wood substrates which contained very little N. We also observed clear differences in the substrate preferences of the fungal species. The Zygomycete mineralized little CO2 from any of the substrates, while the Basidiomycetes mineralized all of the substrates except spruce needles. However, the Ascomycete (Penicillium) was surprisingly efficient at mineralizing spruce wood and was the only species that substantially mineralized spruce litter. The activities of β-glucosidase and N-acetyl-glucosaminidase were strongly correlated with cumulative respiration (r = 0.78 and 0.74, respectively), and Penicillium was particularly effective at producing these enzymes. On moss litter, the different fungal species produced enzymes that targeted different chemical components. Overall, our results suggest that fungal species specialize on different organic substrates, and only respond to N addition on low N substrates, such as wood. Furthermore, the response to N addition is non-linear, with the greatest substrate mineralization at intermediate N levels. 相似文献

Fourteen different silages were prepared using mixtures of Moringa (Moringa oleifera), Elephant grass (Pennisetum purpureum cv Taiwan) or sugar cane (Saccharum officinarum). Molasses from sugar cane was used in the amounts of either 10 or 50 g kg?1 fresh matter (FM) in treatments without sugar cane. A completely randomized design with three replicates of each treatment was used. The silages were prepared in 1800 mL micro silos and opened after 120 d. The presence of Moringa and Elephant grass in the silage changed the pH by ?0·8 and +0·7, respectively (P < 0·001), indicating a favourable effect of Moringa on silage pH. Overall differences were found among treatments for dry matter content, crude protein and acetic acid concentrations, weight loss, CO2 production and silage pH after spoilage (P < 0·001). Weight loss was proportionately 0·034 and 0·014 in silages with and without sugar cane respectively (P < 0·001). Overall, differences (P < 0·05) were also found for neutral‐detergent fibre and lactic acid concentrations, lactic acid bacteria counts, clostridial counts and time to spoilage of the silages. Treatments containing Moringa had higher lactic acid concentrations (+16 g kg?1 DM; P < 0·01) compared to treatments without but the presence of Moringa decreased time to spoilage by 67 h (P < 0·05). No differences were found in propionic acid concentration or fungal growth of the silages. The main objective of this work is to analyze the spatial and temporal response of Q. pyrenaica community to fire. In order to do so, an ecological characterization of 15 forests in different succession stages (5 Initial: 0–20 years, 5 Medium: 20–60 years and 5 Mature: over 60 years), paying special attention to structural and diversity features, was carried out. The results show that temporal changes are very important, and particularly affect the structure and relative abundance of the vegetation. The main differences consist in the change from a homogenous structure, with high cover and overlayering of woody species, at Initial stage, to a more pluristratified, complex and organized structure, with lower cover and overlayering of woody species at Mature stage. The changes in abundances showed a decrease in shrubby species, such as the Erica australis, Genista florida and Erica arborea at Initial stage, and the incorporation of tree species, such as Crataegus monogyna, Acer pseudoplatanus and Alnus glutinosa at Mature stage. However, there are no significative differences between stages for specific richness and diversity values. Spatial heterogeneity is high and similar in all three defined stages. 相似文献

Clonally propagated crop species are less adaptable to environmental changes than those propagating sexually. DNA studies have shown that in all countries where taro (Colocasia esculenta (L.) Schott) has been introduced clonally its genetic base is narrow. As genetic variation is the most important source of adaptive potential, it appears interesting to attempt to increase genetic and phenotypic diversity to strengthen smallholders’ capacity to adapt to climatic changes. A global experiment, involving 14 countries from America, Africa, Asia and the Pacific was conducted to test this approach. Every country received a set of 50 indexed genotypes in vitro assembling significant genetic diversity. After on-station agronomic evaluation trials, the best genotypes were distributed to farmers for participatory on-farm evaluation. Results indicated that hybrids tolerant to taro leaf blight (TLB, Phytophthora colocasiae Raciborski), developed by Hawaii, Papua New Guinea and Samoa breeding programmes outperformed local cultivars in most locations. However, several elite cultivars from SE Asia, also tolerant to TLB, outperformed improved hybrids in four countries and in one country none of the introductions performed better than the local cultivars. Introduced genotypes were successfully crossed (controlled crossing) with local cultivars and new hybrids were produced. For the first time in the history of Aroids research, seeds were exchanged internationally injecting tremendous allelic diversity in different countries. If climatic changes are going to cause the problems envisaged, then breeding crops with wide genetic diversity appears to be an appropriate approach to overcome the disasters that will otherwise ensue. 相似文献

In Chile, the species Choromytilus chorus stands out for its ecological and economic importance. This mussel has a very particular condition in the colour of the gonad, being cream‐yellow colour the male gonad and dark brown the female which is hardly desired by the consumers. In this context, the aim of this research is to determine the role that sex steroids have in gonadal differentiation of the mussel C. chorus. For this, juveniles with sizes 15–22 mm were selected, which were subjected to acute exposure in the form of bath for 60 days to four treatments: T1: dihydrotestosterone (DHT); T2: 17β‐estradiol (E2), T3: fadrozole (F) and T4: DHT‐F (DF), plus a control without chemicals. Each treatment as well as control included three replicates with 90 individuals each. The mussels undergoing treatment with E2 had a sex ratio (male:female) of 0.47 compared to 1:1 in the control group. In contrast, in the groups treated with DHT, F and DF the sex ratio changed to 2.0, 1.60 and 1.70 respectively. In the fertilization trials, all the mussels produced functional gametes, as they were able to generate gametes that were developed to morula, and veliger larva. Nevertheless, it should be noted that the size of oocytes from the group exposed to estradiol (61.12 ± 2.40 μm) was significantly higher than in other groups. These results support the hypothesis that sex steroids would be involved in sexual differentiation of marine bivalves. 相似文献

Cultivated beets (Beta vulgaris ssp. vulgaris) are unable to form reproductive shoots during the first year of their life cycle. Flowering only occurs if plants get vernalized, that is, pass through the winter, and are subsequently exposed to an increasing day length (photoperiod) in spring. Here, we show that the regulation of flowering time in beets is controlled by the interplay of two paralogs of the FLOWERING LOCUS T (FT) gene in Arabidopsis that have evolved antagonistic functions. BvFT2 is functionally conserved with FT and essential for flowering. In contrast, BvFT1 represses flowering and its down-regulation is crucial for the vernalization response in beets. These data suggest that the beet has evolved a different strategy relative to Arabidopsis and cereals to regulate vernalization. 相似文献

Browning reactions in fruits and vegetables are a serious problem for the food industry. In mushrooms, the principal enzyme responsible for the browning reaction is polyphenoloxidase (PPO). A microwave applicator has been designed and used for studying mushroom PPO inactivation. The effects of microwaves and conventional heating on the kinetics of the monophenolase and diphenolase activities of PPO were studied. Conventional and microwave treatments produce different enzyme intermediates with different stability and kinetic properties. We describe how considerable time can be saved during microwave inactivation of the enzyme compared with the time needed when conventional hot-water treatment is used, resulting in greater profitability and enhanced quality. The short exposure time required for samples irradiated with microwaves is very important for maintaining the quality of mushrooms. The fast microwave treatment used resulted in an increase in antioxidant content and a considerable decrease in browning. 相似文献

Flavobacterium psychrophilum is the causative agent of bacterial cold-water disease and rainbow trout syndrome in freshwater salmonid fish worldwide, generating injuries and high mortality rates. Despite several studies on this bacterium, the infection mechanism remains unknown due to limitations in the employed animal models. In this work, we propose using zebrafish (Danio rerio) as a model for studying bacterial pathogenicity. To substantiate this proposal, zebrafish infection by F. psychrophilum strain JIP 02/86 was characterized. Zebrafish larvae were infected using the bath method, and morphological changes and innate immune system activation were monitored using transgenic fish. Salmonid-like infection phenotypes were observed in 4.74% of treated larvae, as manifested by fin, muscle and caudal peduncle damage. Symptomatic and dead larvae accounted for 1.35% of all challenged larvae. Interestingly, infected larvae with no infection phenotypes showed stronger innate immune system activation than specimens with phenotypes. A failure of function assay for myeloid factor pu.1 resulted in more infected larvae (up to 43.5%), suggesting that low infection rates by F. psychrophilum would be due to the protective actions of the innate immune system against this bacterium in zebrafish larvae. Our results support the use of zebrafish as an infection model for studying F. psychrophilum. Furthermore, the percentage of infected fish can be modulated by disturbing, to varying extents, the differentiation of myeloid cells. Using this evidence as a starting point, different aspects of the infection mechanism of F. psychrophilum could be studied in vivo. 相似文献
