As a result of the high lability and slow growth of Campylobacter fetus subspecies, the laboratory diagnosis of bovine genital campylobacteriosis has always been difficult. This is especially true under South African conditions, where farms are far apart, laboratories are only present in major centres and there are high ambient temperatures. In order to overcome the shortcomings associated with traditional diagnostic methods, the implementation of a molecular assay was sought. This work describes how a previously published PCR assay (MG3F/ MG4R primers) was adapted, optimised and applied in the diagnostic laboratory to test preputial samples directly for the presence of Campylobacter fetus. Field evaluation of the assay revealed an analytical sensitivity and specificity of 85.7% and 99%, respectively. Subsequent genotyping and phenotyping of a diverse collection of South African field isolates revealed that South Africa has an unexpected and previously unreported high incidence of Campylobacter fetus subsp. venerealis biovar intermedius strains. These strains were not identified correctly by the subspecies-specific primer set evaluated. Until such time that cost- effective genotyping methods are available to diagnostic laboratories in South Africa, and other countries with these atypical Campylobacter fetus subsp. venerealis strains, the need for bacterial culture will persist. Identification to subspecies level of isolates at present remains dependent upon a single phenotypic criterion, namely tolerance to 1% glycine.  相似文献

Objective: Mechanical evaluation of 2 suture constructs for extracapsular stifle stabilization. Study Design: In vitro study. Sample Population: Crimped interlocking loop constructs (ILC) of 45 kg nylon leader line (NLL) and Orthofiber® (OF). Methods: ILC were tightened to 100 N, then crimp secured. Ramp to failure (n=10/group)—Data were derived from force/displacement plots. Stress–relaxation testing (n=10/group)—ILC's were nondestructively loaded and held at resultant displacement as force data were recorded. Incremental, cyclic loading (n=10/group)—ILC's were loaded (5 cycles/set) starting at 100 N and incrementally increased by 50 N (1 and 3 Hz protocols). Loop tension and elongation were recorded after each set. Results: Ramp to failure—initial loop tension was similar (NLL 75.5 ± 9.5 N; OF 68.7 ± 10.4 N, P=.140). Tested OF constructs were stiffer (NLL 125.7 ± 4.0; OF 234.6 ± 25.0 N/mm, P≤.001), had lower yield load (NLL 193.6 ± 13.8; OF 137.3 ± 94.3 N, P≤.001), lower peak load (NLL 873.7 ± 68.6; OF 653.6 ± 80.2 N, P≤.001), and lower elongation at failure (NLL 19.1 ± 1.4; OF 5.2 ± 1.0 mm, P≤.001) and at yield (NLL 1.52 ± 0.2; OF 0.3 ± 0.6 mm, P=.003) than NLL constructs. Yield in NLL ILC's was variable knot tightening/crimp slippage, but only crimp‐suture slippage in OF. Stress–relaxation testing—OF demonstrated greater relaxation. Incremental, cyclic loading—induced ILC elongation and tension loss in both groups, independent of loading frequency. NLL lost tension at lower rate, but elongated more than OF. Conclusions: NLL construct is mechanically superior to OF construct.  相似文献

A flock of sheep, known to be infected with the "FSA" mutant of Brucella melitensis Rev. 1, was examined serologically and bacteriologically to determine whether any relationship existed which would help in the control of this infection in the field. An attempt was also made to determine whether vertical transmission occurred. Twenty-one out of 62 sheep were bacteriologically positive. The best organs for isolation were the udder, supramammary lymphnodes and uterus. No significant relationship could be shown between the complement fixation test and bacterial isolation. The absence of any relationship between serological and bacteriological results agrees with a short-lived infection. None of the 24 lambs sacrificed at 5 months showed either serological reactions or were bacteriologically positive, thus no vertical transmission could be shown.  相似文献

Context

This study synthesizes evidence from three separate surveys of American Black Duck and Mallard breeding habitat usage to quantify the effects of agriculture at the landscape scale.

Objectives

To assess duck breeding activity in agricultural landscapes within the Canadian maritimes in order to measure the overall impact of agricultural land use, the response to particular agricultural activities, and the influence of landscape configuration.

Methods

Models, constructed using a long-term census (SURVEY1), were used to predict habitat selection for two other independent surveys (SURVEY2, SURVEY3). Predictions incorporated information about wetland area and diversity, as well as anthropogenic factors, allowing subsequent analyses to focus on the remaining residual error attributable to agricultural effects.

Results

SURVEY2 results demonstrated that the proportion of active agriculture is an important indicator of the severity of human disturbance, yielding threshold estimates of 39% for Mallards and 60% for Black Ducks, with an overall average of 50%. Agricultural conversion beyond these thresholds deterred breeding ducks independently of other factors. SURVEY3 land cover information demonstrated that the presence of cropland intensified this deterrence effect, even at levels as low as 10%. Woodland cover (in excess of 30%) was important for both species, but its configuration was not.

Conclusions

In addition to quantifying threshold effects, this study reaffirms that woodland is an important part of the maritime landscape matrix, and contributes positively to habitat diversity in mixed use, moderate intensity agricultural regions. Wetland restoration in agricultural landscapes should monitor and promote less crop-intensive, mixed-use practices.

Objective: To estimate the relative accuracy of a thoracic focused assessment with sonography for trauma (TFAST) protocol for rapid diagnosis of pneumothorax (PTX) and other thoracic injury in traumatized dogs. Design: Prospective case series. Setting: Private veterinary emergency center. Animals: One hundred and forty‐five client‐owned dogs evaluated within 24‐hours of injury. Interventions: Thoracic focused assessment with sonography for trauma protocol. Measurements and Main Results: Traumatized dogs were evaluated with a conventional ultrasound (US) machine using a standardized 4‐point thoracic FAST protocol before thoracic radiography (CXR) and thoracocentesis. PTX was diagnosed by the absence of the ‘glide sign,’ defined as the lack of the normal dynamic interface between lung margins gliding along the thoracic wall during respiration. Concurrent thoracic trauma was diagnosed by the presence of pleural or pericardial fluid or the presence of a ‘step sign,’ defined as an abnormal glide sign. Accuracy of TFAST was calculated relative to CXR findings. Results: Overall sensitivity (Se), specificity (Sp), and accuracy of TFAST relative to CXR were 78.1% (95% CI; 61.5, 89.9), 93.4% (95% CI; 87.4, 97.1), and 90.0%, respectively. Se and Sp were higher in dogs with penetrating trauma (93.3%, 96.0%) and for the evaluator with the most clinical experience (95.2%, 96.0%); only Se between the most experienced compared with others was statistically significant (P<0.05). TFAST documented other concurrent thoracic injury. Median time for TFAST was 3 minutes. Conclusions and Clinical Relevance: TFAST has the potential to rapidly diagnose PTX and other thoracic injury and guide therapy, including potentially life‐saving interventions, in traumatized dogs.  相似文献

Important drivers of woody cover in African savannas are rainfall, soil and nutrients, and disturbance factors, such as fire and herbivory. However, very little is known about how these drivers influence woody cover at specific height classes. The main aim of this study was to identify which of these drivers are the best predictors of woody cover at three height classes: shrub (0.75–2.5 m), brush (2.5–5.5 m) and tree (>5.5 m). Percent woody cover estimates were collected using a rapid monitoring technique over 1 700 sites in Kruger National Park, South Africa. Geology (basalt and granite), mean annual rainfall, fire frequency and elephant density were analysed as potential drivers of woody cover. Results indicate that mean annual rainfall was negatively associated with shrub cover, fire frequency was negatively associated with brush cover, and elephant density was negatively associated with tree cover. Patterns of woody cover show that while geology influences the spatial distribution of horizontal woody cover, variation in vertical vegetation structure is created and maintained by top-down disturbance. This provides the first documentation of drivers associated with structurally differentiated woody cover at regional scales. Future studies on woody cover in disturbance-driven environments should not ignore vegetation structure.  相似文献

Endothia gyrosa is a canker pathogen best known as the causal agent of pin oak blight in North America, and causes cankers on other woody hosts such as Castanea spp. and Liquidambar spp. In South Africa, Australia and Tasmania, a fungus identified as E. gyrosa has been recorded on Eucalyptus spp. Some morphological differences exist between the North American fungus and the isolates from Eucalyptus . Phylogenetic relationships between E. gyrosa from North America and E. gyrosa from South Africa and Australia, as well as that of the related fungi Cryphonectria parasitica and C. cubensis , were studied using PCR-based restriction fragment length polymorphism (RFLP) and sequences of the internal transcribed spacer (ITS) region of the rRNA operon. Endothia gyrosa isolates from South Africa produced the same RFLP banding patterns as those from Australia, which differed markedly from North American isolates of E. gyrosa . In a phylogram based on the DNA sequences, the Australian and South African isolates of E. gyrosa resided in a single, well resolved clade, distinct from North American isolates. Isolates of C. parasitica grouped in the same clade as the South African and Australian isolates of E. gyrosa , but C. cubensis was distantly related to them. The molecular data suggest that the E. gyrosa isolates from South Africa and Australia represent a distinct taxon, and probably belong to the genus Cryphonectria .  相似文献

Abundances of Culicoides imicola, the insect vector of several livestock viruses, including bluetongue and African horse sickness, were recently published for 34 sites in southern Africa, together with associated climate data. Here, these data are analysed statistically in combination with certain satellite-derived variables, with the aim of developing predictive models of C. imicola abundance. Satellite-derived variables were the land surface temperature (LST, a measure of temperature at the earth's surface) and the normalised difference vegetation index (NDVI, a measure of photosynthetic activity). Two models were developed: (1) climatic variables only and (2) satellite-derived and climatic variables. For model I, the best model used a single predictor variable (the mean daily minimum temperature) only, and accounted for nearly 34% of the variance in C. imicola abundance. Two variable climatic models did not perform significantly better. For model II, the best 1-variable model used the annual minimum LST as a predictor of C. imicola abundance, and accounted for nearly 40% of the variance in C. imicola abundance. The best 2-variable model, which gave a significantly better fit than the 1-variable model, combined the minimum LST and minimum NDVI as predictors of C. imicola abundance, and accounted for nearly 67% of variance. A map of predicted C. imicola abundances is produced on the basis of this 2nd model which, despite some anomalies, agrees largely with what is currently known of the prevalence of C. imicola in the region.  相似文献

Lumpy skin disease (LSD) is a disease of cattle, primarily in Africa and Madagascar and rarely in the Middle East. It is caused by a capripoxvirus that belongs to the family Poxviridae. The disease is of economic importance in endemic areas. Effective control of LSD requires accurate and rapid laboratory techniques to confirm a tentative clinical diagnosis. Comparative studies on different diagnostic tests used at different stages of the disease have not been done. The aim of this study was to compare several of these tests. Six seronegative bulls, between 11 and 20 months of age, were infected intravenously and kept in an insect-free facility. The course of the infection was monitored. During a 3-month period blood samples and skin biopsies were collected for virus isolation and polymerase chain reaction (PCR). Skin biopsies were also examined using transmission electron microscopy (TEM). The incubation period in infected animals varied from 4-5 days. The length of the viraemic period did not correlate with the severity of clinical disease. Viraemia was detected from 1-12 days using virus isolation and from 4-11 days using the PCR, which is longer than has previously been reported. Virus was isolated from skin biopsies until Day 39 post infection (p.i.) and PCR could demonstrate viral DNA until Day 92 p.i. Transmission electron microscopy of negatively stained skin biopsies detected LSD virus only in one of the four bulls that developed skin lesions until Day 33 p.i. The PCR was a fast and sensitive method to demonstrate viral DNA in blood and skin samples. It could detect viral nucleic acid in skin lesions 53 days longer than virus isolation. Virus isolation from blood and skin samples was sensitive and reliable, but as a single test it may be too time-consuming to use although this depends on how rapidly the diagnosis must be confirmed. In conclusion, this study showed the PCR to be superior in detecting LSD virus from blood and skin samples. However, virus isolation is still required when the infectivity of the LSD virus is to be determined.  相似文献
