The pituitary gland, occupying a central position in the hypothalamo-pituitary thyroidal axis, produces thyrotropin (TSH), which is known to stimulate the thyroid gland to synthetize and release its products, thyroid hormones. TSH is produced by a specific cell population in the pituitary, the so-called thyrotropes. Their secretory activity is controlled by the hypothalamus, releasing both stimulatory and inhibitory factors that reach the pituitary through a portal system of blood vessels. Based on early experiments in mammals, thyrotropin-releasing hormone (TRH) is generally mentioned as the main stimulator of the thyrotropes. During the past few decades, it has become clear that the hypophysiotropic function of the hypothalamus is more complex, with different hormonal axes interacting with each other. In the chicken, it was found that not only TRH, but also corticotropin-releasing hormone (CRH), the main stimulator of corticotropin release, is a potent stimulator of TSH secretion. Somatostatin (SRIH), a hypothalamic factor known for its inhibitory effect on growth hormone secretion, was demonstrated to blunt the TSH response to TRH and CRH. In this review we summarize the latest studies concerning the "interaxial" hypothalamic control of TSH release in the chicken, with a special emphasis on the molecular components of these control mechanisms. It remains to be demonstrated if these findings could also be extrapolated to other species or classes of vertebrates.  相似文献

[14C]Diflubenzuron is readily degraded in various agricultural soils and in hydro-soil; 50% of the applied dose of 1 mg kg−1 was metabolised in 2 days or less. The chief products of hydrolysis were identified as 4-chlorophenylurea and 2, 6-difluorobenzoic acid. A part of the radioactivity, increasing with incubation time, could not be extracted. Release from the soil of [14C]carbon dioxide, derived from both labelled phenyl rings, points to the ultimate mineralisation of diflubenzuron.  相似文献

The susceptibility to stem rot and wilt caused by Phoma clematidina was tested for a range of clematis varieties and species widely grown in Europe, and compared with their susceptibility to wilt in practice as judged by amateur and commercial growers in two postal surveys. Most small flowering clematis species did not die back when unwounded stems were inoculated with spores of P. clematidina, and were also reported in the surveys as being very resistant to wilt. In contrast, large flowering hybrids were found to be highly susceptible both to stem infection by P. clematidina and to wilt in practice according to growers. A significant positive linear relationship existed between the susceptibility scores for disease in the stem inoculation test and those for wilt from the surveys. Fungal isolations were made from the stems of naturally wilted clematis plants from British nurseries and private gardens. Unlike the many other fungal species isolated, P. clematidina occurred very frequently, especially in stems of large flowering varieties. Together with the results from the grower surveys and stem inoculation trial, this indicates that this fungus is a regular cause of clematis wilt. The stem inoculation test with P. clematidina described in this study would be useful for European breeders and growers to determine the susceptibility of new clematis varieties to wilt before they are marketed.  相似文献

The prevalence of Cryptosporidium and Giardia was studied on 10 intensively reared sheep and goat farms in the province of East Flanders, Belgium. Random faecal samples were collected and examined using the Merifluor((R)) immunofluorescence assay. Cryptosporidium positive samples were withheld for molecular identification using primers targeting the 18S rDNA, 70 kDa heat shock protein and 60 kDa glycoprotein gene. For the molecular identification of Giardia the beta-giardin gene and a recently developed assemblage specific PCR based on the triose phosphate isomerase gene were used. The prevalence of Cryptosporidium in lambs was 13.1% (18/137), on 4 out of 10 farms. In goat kids the Cryptosporidium prevalence was 9.5% (14/148), on 6 out of 10 farms. The molecular characterisation of Cryptosporidium positive isolates indicated that in lambs (n=10) the cervine genotype was predominant, whereas in the goat kids (n=11) only C. parvum was identified, with subgenotypes IIaA15G2R1 and IIdA22G1. The Giardia prevalence was 25.5% (35/137) in lambs with all 10 farms being positive, and 35.8% (53/148) in goat kids with 8 out of the 10 farms being positive. Both in the goat kids and in the lambs the host specific assemblage E was most commonly identified. However, the zoonotic assemblage A was identified in 6 out of 28 goat kids and in 2 out of 8 lambs, based on the beta-giardin sequence alignment. Using the assemblage specific PCR, mixed assemblage A and E infections were additionally identified in 2 lambs and in 5 goat kids. The results of the present study indicate that both Cryptosporidium and Giardia are common parasites on intensively reared sheep and goat farms in the province of East Flanders, Belgium, and that they are a potential source for zoonotic infections.  相似文献

Fusarium head blight (FHB) of wheat and other small-grain cereals is a disease complex caused by several fungal species. To monitor and quantify the major species in the FHB complex during the growing season, real-time PCR was developed. TaqMan primers and probes were designed that showed high specificity for Fusarium avenaceum, F. culmorum, F. graminearum, F. poae and Microdochium nivale var. majus. Inclusion of an internal PCR control and serial dilutions of pure genomic DNAs allowed accurate determination of the concentration of fungal DNA for each of these species in leaves, ears as well as harvested grains of winter wheat. The DNA concentration of F. graminearum in grain samples correlated (r 2= 0.7917) with the incidence of this species on the grain as determined by isolation from individual kernels. Application of the TaqMan technology to field samples collected in 40 wheat crops in the Netherlands during the growing season of 2001 revealed that M. nivale var. majus predominated on leaves early in the season (GS 45-65). Ears and harvested grains from the same fields, however, showed F. graminearum as the major species. In 2002, grain samples from 40 Dutch fields showed a much wider range of species, whereas in ears from 29 wheat crops in France, F. graminearum was the predominant species. The concentration of DON correlated equally well with the incidence of the DON-producing species F. culmorum and F. graminearum in the grain samples (r 2= 0.8232) as well as with total DNA of both these species (r 2= 0.8259). The Fusarium TaqMan technology is an important tool to quantify and monitor the dynamics of individual species of the complex causing FHB in cereals during the growing season. This versatile tool has been applied in a comparison of different genotypes, but can also be applied to other disease management systems, e.g. fungicide treatments.  相似文献

Smallholder farmers in sub-Saharan Africa are confronted by low productivity and limited investment capacity in nutrient inputs. Integrated soil fertility management (ISFM) aims at increased productivity through the combined use of improved germplasm, judicious fertilizer application and organic matter management, adapted to the local farming conditions. We hypothesize that the application of these different ISFM components can result in significant increases in productivity and economic benefits of cassava–legume intercropping systems. Participatory demonstration trials were conducted in the highlands of Sud-Kivu, DR Congo with 12 farmer groups during 3 seasons. Treatments included the farmers’ common practice (local common bean and cassava varieties, seed broadcast and manure addition) and sequentially added ISFM components: improved bean and cassava germplasm, modified crop arrangements, compound NPK fertilizer application and alternative legume species (groundnut or soybean). The use of improved germplasm did not result in yield increases without simultaneous implementation of other ISFM components. Modifying the crop arrangement by planting cassava at 2 m between rows and 0.5 m within the row, intercropped with four legume lines, increased bean yields during the first season and permits a second bean intercrop, which can increase total legume production by up to 1 t ha−1 and result in an additional revenue of almost 1000 USD ha−1. Crop arrangement or a second legume intercrop did not affect cassava storage root yields. Fertilizer application increased both legume and cassava yield, and net revenue by 400–700 USD ha−1 with a marginal rate of return of 1.6–2.7. Replacing the common bean intercrop by groundnut increased net revenue by 200–400 USD ha−1 partly because of the higher market value of the grains, but mostly due to a positive effect on cassava storage root yield. Soybean affected cassava yields negatively because of its high biomass production and long maturity period; modifications are needed to integrate a soybean intercrop into the system. The findings demonstrate the large potential of ISFM to increase productivity in cassava–legume systems in the Central-African highlands. Benefits were, however, not observed in all study sites. In poor soils, productivity increases were variable or absent, and soil amendments are required. A better understanding of the conditions under which positive effects occur can enable better targeting and local adaptation of the technologies.  相似文献

The hydrological probability-distributed model (PDM) is widely used all over the world and its applicability has also been investigated in Flanders, Belgium. Rainfall–discharge relations for all gauging stations installed on non-navigable watercourses were modelled over a long succession of monitoring years. In all, 1456 years were modelled. Typical characteristics (peak flow, volumes) of modelled series are compared with observations. Based on the relatively long time series, reliable discharge values can be generated with the PDM. Water volumes and peak characteristics are very close to the observed values. The set of 98 PDMs was analysed and clustered. Three cluster approaches were considered: a single-parameter approach, a parameter set approach and an approach with known cluster zones, delineated on hydrological flow characteristics. The single-parameter approach, the parameter set approach and the combination of both gave less detailed regional information than the clustering on hydrological characteristics.  相似文献

Tare soil is soil attached to harvested products like potato tubers. Tare soil becomes a considerable waste stream after storage, washing and processing of harvested products. There is a high risk on contamination of tare soils with (quarantine) phytopathogens, because of import of harvested products from different regions of the world. Disinfestation is necessary to make tare soils applicable for agricultural production. Anaerobic (non-chemical) soil disinfestation (ASD) and inundation are suitable methods to disinfest tare soils. Two different soils (marine loam and glacial sand) were either treated with five (2011) or four (2012) treatments and these were: (i) no treatment (control), (ii) freshly cut grass, (iii) Herbie 7025, (iv) inundation (five cm water on top of the soil surface), and (v) combination of Herbie 7025 and inundation (2011 only). Containers with treated and untreated soils were inoculated with two quarantine phytopathogens, Ralstonia solanacearum biovar 2 and Globodera pallida. After soil inoculation, the containers were airtight closed and only opened for destructive sampling after 84 days. Then, soils were analysed for the presence of R. solanacearum biovar 2, using immunofluorescence colony staining, and for the presence of viable G. pallida eggs, determined by the number of juveniles hatched from eggs and lured to potato root exudate. Strong (> 99.4 %) declines of both pathogens in treated versus the appropriate control soils were observed. Repetition of the experiment revealed the same pattern and therefore it was concluded that ASD and inundation, as separate treatments, have promise for remediation of tare soils from contaminating quarantine pathogens.  相似文献
