Rainbow smelt (Osmerus mordax) tolerate temperatures close to the freezing point of sea water, in part, through the use of glycerol as an antifreeze. Potential mechanisms for glycerol production by liver were assessed by comparing activities of key enzym es of carbohydrate and amino acid metabolism in rainbow smelt to those in Atlantic tomcod (Microgadus tomcod) and smooth flounder (Liopsetta putmani). The latter two species inhabit the same environment but do not maintain high levels of blood glycerol. The enzyme profile of liver from rainbow smelt is substantially different from those of the other species and is poised for glycerol production. With respect to carbohydrate metabolism, glycerol-3-phosphate dehydrogenase activity in rainbow smelt liver was 156 µmoles min-1 g-1, a level which was 28 and 12-fold higher than activities in tomcod and flounder liver, respectively. Glycerol-3-phosphatase activity in smelt liver was 1.95 µmol min-1 g-1. This activity was 2.7 and 5.4-fold higher than those in tomcod and flounder liver, respectively. As such, the production of glycerol appears to be dependent upon the concerted action of glycerol-3-phosphate dehydrogenase and glycerol-3-phosphatase. The enzyme profile also suggests that amino acids are a potential source of carbon for glycerol. Aspartate aminotransferase activity in rainbow smelt was 7 to 14-fold higher in comparison to tomcod and flounder liver, respectively. Activities of alanine aminotransferase and glutamate dehydrogenase in liver were between 2 and 3-fold higher in rainbow smelt than in the other two species. Finally, it is shown that in vitro preparations of smelt liver sections produce glycerol at 0°C lending support to the concept that liver is a site of glycerol synthesis in vivo.  相似文献

Microbial insect pathogens offer an alternative means of pest control with the potential to wean us off our heavy reliance on chemical pesticides. Insect pathogenic fungi play an important natural role in controlling disease vectors and agricultural pests. Most commercial products employ Ascomycetes in the genera Metarhizium and Beauveria. However, their utilization has been limited by inconsistent field results as a consequence of sensitivity to abiotic stresses and naturally low virulence. Other naturally occurring biocontrol agents also face these hurdles to successful application, but the availability of complete genomes and recombinant DNA technologies have facilitated design of multiple fungal pathogens with enhanced virulence and stress resistance. Many natural and synthetic genes have been inserted into entomopathogen genomes. Some of the biggest gains in virulence have been obtained using genes encoding neurotoxic peptides, peptides that manipulate host physiology and proteases and chitinases that degrade the insect cuticle. Prokaryotes, particularly extremophiles, are useful sources of genes for improving entomopathogen resistance to ultraviolet (UV) radiation. These biological insecticides are environmentally friendly and cost‐effective insect pest control options. © 2017 Society of Chemical Industry  相似文献

The goal of this study was to determine the prevalence of gastric ulcers in horses with acute abdominal crisis (colic) and to examine the temporal effect of hospitalization on ulcer development in equine patients treated for colic. In addition, other factors that may be associated with gastric ulceration were also explored. The study design was a prospective original study incorporating 169 horses that presented to the George D. Widener Hospital for examination. One hundred and twelve horses presenting with the chief complaint of colic were included in the study group, and 57 horses that presented for non-colic or nonemergency complaints were evaluated and included as case controls. Gastroscopy was performed on equine patients presenting with the chief complaint of colic or horses presenting for reasons other than colic (control); mucosal changes were scored 0 to 3. Additionally, horses presenting for colic were gastroscopically evaluated twice during a 5-day period. Medical records were reviewed for history, clinical findings, laboratory results, and treatment. Seventy-six of 112 horses presenting with the chief complaint of colic had gastric ulceration compared with 41 of 57 horses in the control group. There was a significant association between age of the patient and chief complaint (ie, colic vs control) and between breed and chief complaint. There was no association between gastric ulcer score and chief complaint (colic vs control). Thirty-eight of the 112 horses presenting with colic deteriorated in ulcer score while hospitalized. Using a Pearson chi-squared test, there was no statistically significant association between gastric ulceration with age, breed, or sex. Horses with gastric ulceration in the colic group had lower packed cell volumes compared with horses presenting with colic without gastric ulcers, and this was statistically significant (P < .001). The high incidence of gastric ulceration in the study and control groups supports the reports of other investigators that gastric ulcers in horses, no matter the presenting complaint, are widespread. There was a significant association between breed and chief complaint (P = .005); however, breed and outcome of gastric ulceration were not related (Thoroughbreds were the least likely breed to present for colic). Although a trend in increasing gastric ulceration was seen in hospitalized colic patients, it was not statistically significant. This suggests that horses that are hospitalized may be at increased risk for developing gastric ulcers because of stress, feed deprivation, and administration of treatment. Thus, horses that present for colic should be gastroscopically evaluated if clinical signs raise the index of suspicion for gastric ulceration.  相似文献

Previous in situ studies of orchid population dynamics with conservation relevance have focused on one or a few populations in a limited area. Many species of orchids occur as hyperdispersed populations in ephemeral habitats (epiphytic, twig epiphytes, short lived or vulnerable host). In this contribution, we show that orchid populations that are patchily distributed and that exist in disturbance-prone environments may act somewhat like a metapopulation with high turnover and low correlation in population dynamics. We tested for evidence of metapopulation dynamics in the riparian orchid Lepanthes rupestris by sampling over 1000 sites (250 initially occupied, 750 initially unoccupied) in biannual surveys for 5 years. Extinction and colonization of groups of orchids on a single substrate or patch (either trees or boulders) was common and more or less consistent across different time periods, and asynchronous subpopulation dynamics were evident among the populations. From this we predict non-zero equilibrium values for site occupancy () of L. rupestris. Nevertheless, this study species differed from a typical Levins’ metapopulation system in that small populations were more likely to go extinct than large populations, and that colonization of previously occupied sites was more common than colonization of initially unoccupied sites suggesting that site quality may influence population persistence and colonization. A major difficulty applying the metapopulation approach to orchid conservation is identifying empty sites suitable for colonization. In spite of this limitation, our study highlights the necessity of following multiple orchid subpopulations (e.g., an entire orchid “metapopulation” in the broad sense) may provide a more accurate basis for predicting persistence in epiphytic orchids.  相似文献

The isoflavone compositions and concentrations in the leaf, flower, petiole, and stem of 13 red clover cultivars were studied using high-performance liquid chromatography coupled with a diode array and a mass spectrometric detector with negative electrospray ionization. Different cultivars showed significantly different concentrations of individual and total isoflavones. The leaf contained the highest overall concentration, followed by the stem, petiole, and flower. Biochanin A and formononetin were the predominant isoflavones in all cultivars and all parts, along with eight other minor aglycones, daidzein, genistein, glycitein, irilone, orobol, pratensein, pseudobaptigenin, and prunetin, and four minor malonylglycosides, genistein-7-glucoside-6' '-malonate, orobol-7-glucoside-6' '-malonate, formononetin-7-glucoside-6' '-malonate, and biochanin A-7-glucoside-6' '-malonate. The isoflavone compositions and concentrations were also found to be different between red clover parts harvested at the early bud stage and the late flowering stage. Sample storage and handling prior to analysis were also found to be important. Samples in this study were kept at -5 degrees C for a few days before being freeze-dried and were found to contain mainly the aglycones of isoflavones. This may actually be an advantage in that "natural" and more bioactive isoflavones can be obtained without using chemical hydrolysis. Findings in this study therefore provide important information for developing isoflavone-rich red clovers and for optimizing harvest timing and choosing the right part of the red clover plant.  相似文献

The measurement of antioxidant capacity in fruits differs from that of other biological samples due to their low pH and very low lipophilic antioxidant capacity. In this report, we present a modified 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) method for fruits and compare its performance with the other commonly used antioxidant methods of 2,2'-diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP). The antioxidant capacity and reaction kinetics of four phenolic compounds, two antioxidant standards, and five fruits were also investigated. The modified ABTS method prepared at a pH of 4.5 with sodium acetate buffer is highly stable and easily applied to fruit samples as compared to the standard (pH 7.4) version. The measured antioxidant capacity of samples varied with the assay method used, pH, and time of reaction. Traditional antioxidant standards (trolox, ascorbic acid) displayed stable, simple reaction kinetics, which allowed end point analysis with all of assays. Of the phenolic compounds examined, chlorogenic and caffeic acids exhibited the most complex reaction kinetics and reaction rates that precluded end point analysis while gallic acid and quercetin reached stable end points. All fruit extracts exhibited complex and varied kinetics and required long reaction times to approach an end point. Because the antioxidant capacity of fruit extracts is a function of the array of individual antioxidants present, accurate comparisons among fruit samples require that reaction times be standardized and of sufficient length to reach steady state conditions and that more than one assay be used to describe the total antioxidant activity of fruit samples.  相似文献

Diet is the primary source of iron (Fe) for freshwater fish, and the absorption of Fe is believed to occur via the Nramp family of divalent metal transporters (also called DMT1). Presently, the homeostatic regulation of dietary Fe absorption in fish is poorly understood. This study examined the gastrointestinal mRNA expression of two Nramp isoforms, Nramp-β and Nramp-γ, in the freshwater rainbow trout (Oncorhynchus mykiss), following exposure to elevated dietary Fe [1,256 mg Fe/kg food vs. 136 mg Fe/kg food (control)] for 14 days. The physiological performance, plasma Fe status and tissue-specific accumulation of Fe were also evaluated. In general, the mRNA expression level of Nramp was higher in the intestine relative to the stomach. Interestingly, fish fed on a high-Fe diet exhibited a significant induction in Nramp expression after 7 days, followed by a decrease to the level observed in control fish on day 14. The increase in Nramp expression correlated with the elevated gastrointestinal and plasma Fe concentrations. However, the hepatic Fe concentration remained unchanged during the entire exposure period, indicating strong homeostatic regulation of hepatic Fe level in fish. Fish appeared to handle increased systemic Fe level by elevating the plasma transferrin level, thereby enhancing the Fe-binding capacity in the plasma. Overall, our study provides new interesting insights into the homeostatic regulation of dietary Fe uptake and handling in freshwater fish.  相似文献

Agriculture intensification has deeply modified agroecosystems from field to landscape scales. To achieve successful pest control using natural enemies, understanding species interactions over all scales remains a challenge. Using the cabbage root fly as a model, we studied whether field and landscape characteristics influenced colonization and infestation of broccoli fields by the pest and its control by natural enemies. We also determined whether species of different trophic level or host specialization would respond to environmental characteristics at the same spatial extent. During a multiple-species and multiple-spatial extent study in northwestern France, we recorded pest colonization and infestation in 68 fields, collected associated natural enemies and assessed crop damages. In each field, we considered management practices and characterized the surrounding landscape in 50–500 m-wide buffers. Our main findings are that Delia radicum and its main natural enemies respond to both field and landscape characteristics. Semi-natural areas supported both crop colonization by pests and natural enemy action. The pest and its enemies differed in their responses to field or landscape variables. Landscape elements such as field banks favored the movement of the pest while impeding the movement of some natural enemies. Pest pressure did not increase with the neighboring density of Brassica crops. The presence of natural enemies did not reduce crop damage but reduced pest emerging rates. Finally, specialist parasitoids responded to the landscape at larger spatial extents than generalists. These results outline the complexity of improving pest control through landscape management.  相似文献

Many populations are spatially structured with frequent extinction–colonization events. A clear understanding of these processes is necessary for making informed and effective management decisions. Due to the spatially and temporally dynamic nature of many systems, population connectivity and local extinction–colonization processes can be difficult to assess, but graph theoretic and occupancy modeling approaches are increasingly being utilized to answer such vital ecological questions. In our study, we used 6 years of egg mass counts from 34 ponds for Rana sylvatica to parameterize spatially explicit demographic network models. Our models revealed that the studied populations have spatial structure with strong source–sink dynamics. We also assessed the colonization and persistence probability of each pond using multi-season occupancy modeling. We observed extreme fluctuation in reproductive effort among years, resulting in variable levels of connectivity across the landscape. Pond colonization and persistence were most influenced by local population dynamics, but colonization was also affected by precipitation. Our demographic network model had moderate ability to predict reproductive effort, but accuracy was hindered by variation in annual precipitation. Source populations had higher colonization and persistence rates as well as a greater proportion of ravine habitat within 1,000 m than sink populations. By linking a spatially explicit connectivity model with a temporal occupancy/persistence model, we provide a framework for interpreting patterns of occupancy and dispersal that can serve as an initial guide for future habitat management and restoration.  相似文献

The productive potential of potatoes (Solanum tuberosum L. cvs. Norland, Superior, Norchip, and Kennebec) was assessed for life support systems being proposed for space stations and/or lunar colonies. Plants were grown in walk-in growth rooms for 15 weeks at 20 C under 12-, 16- and 20-h photoperiods of 400 μmol mt-2st-1 photosynthetic photon flux (PPF). Norland yielded the greatest tuber fresh weight, producing 2.3, 2.4, and 2.9 kg/plant under 12-, 16-, and 20-h photoperiods, respectively. The respective yields for the other cultivars under 12-, 16-, and 20-h were: Superior, 1.9, 1.5, and 1.8 kg/plant; Norchip, 1.8, 1.4, and 2.0 kg/plant; and Kennebec, 2.3, 0.2, and 0.8 kg/plant. Shoot and total plant biomass increased with lengthening photoperiods except for Kennebec, which showed increased shoot growth but no change in total growth with the longer photoperiods. Kennebec shoot growth under the 20-h photoperiod, and to some extent under 16-h, was noticeably stunted with shortened internodes. In addition, leaves of these plants showed mild chlorosis with rusty “flecking” of the surfaces. The harvest index (ratio of tuber yield/total biomass) was highest for all cultivars under the 12-h photoperiod, with a maximum of 0.69 for Norland. Similarly, the tuber yield per input of irradiant energy also was highest under 12-h for all cultivars. The tuber yield expressed on an area basis for the highest yielding treatment (Norland under 20-h) equaled 2.2 kg dry matter mt-2. Over 15 weeks this equates to a productivity of 20.7 g tuber dry matter mt-2 dayt-1. Assuming 3.73 kcal per g tuber dry matter and a daily human dietary requirement of 2800 kcal, then 36 m2 of potatoes could supply the daily energy requirement for one human.
