For centuries, Old World chameleons (Chamaeleonidae family) have been collected and studied for their unusual biology and features, which are unique among lizards and other vertebrates. They have advanced mechanisms for capturing prey with their tongue, but have a primitive mechanism for hearing. Chameleons have the most studied ocular system because of their highly adapted yet primitive biology. Two approaches to optimise AFLP fingerprinting forthe rapid detection of genetic polymorphisms, i.e.reduced primer selectivity and bulked DNA analysiswere examined. The efficiency of reduced primerselectivity to increase the detection frequency ofgenetic polymorphisms and to obtain more informativefingerprinting profiles was tested in six differentcrops. The number of selective nucleotides was reducedto six in onion, to five in barley, potato, lettuceand cabbage, and to four in flax. This allowed therapid identification of several primer pairs that wereable to discriminate between closely relatedgermplasm. Reproducibility tests on replicate DNAsamples indicated no major negative effects on thereliability of the fingerprinting profiles due to theuse of less selective primers, although for onionpurified DNA was needed to avoid irreproducibleresults. In barley, flax and onion, a less thanfourfold increase in the number of fragments wasobserved when primer pairs were reduced by oneselective nucleotide. This result was attributed todifferent tolerance levels for amplificationmismatches between primer pairs of differentselectivity.The efficiency of bulked DNA analysis to detectgenetic polymorphisms was investigated in differentmixtures of two barley DNA samples. AFLP's of varyingintensity could still be recovered when the two DNA'swere mixed in a 1:1 ratio. However, the frequency ofrecovered bands quickly dropped when in the mixturesthe presence of the DNA carrying the fragments wasdecreased below 50%.The usefulness of the two approaches is discussed inrelation to various aspects of genetic resourcesmanagement.  相似文献

Despite its small area and intensively cropped landscape, East Java accounts for 30% of Indonesia's cattle population. About two million households draw on family labour to raise cattle in backyard sheds and small enclosures, largely for cash income. In this paper, we examine the opportunity for such small‐scale producers to benefit from Indonesia's economic transformation, given the rising urban demand for beef. The paper reports on a study in two contrasting sites in East Java – irrigated lowlands and rainfed uplands – to explore the constraints facing small‐scale cattle producers in these environments, the means by which they have adapted to these constraints (especially by going beyond the farm household to access feed supplies) and possible means to enhance their production systems and incomes. The findings suggest that such cattle production systems can provide a viable source of livelihood, even for resource‐poor households; hence, appropriately adapted cattle improvement programmes are a sensible component of a pro‐poor development strategy.  相似文献

Acidification of surface waters and forest soils is severe in large parts of southern Sweden. The shallow groundwaters are also affected. Large scale liming of surface waters and streams is in operation, often combined with wetland liming to limit the effects of acid episodes, e.g. at snow melt. Acid episodes are perhaps the most severe problem in limed surface waters and in many as yet well buffered waters, because of temperature-layered acid inflow, often superficial. As a result of some investigations, a large scale forest liming programme covering 6.500–10.000 km2 was recently suggested. The main objectives of this forest liming programme are to retard cation depletion and to prevent nutrient imbalance and forest decline in acidified areas. This paper deals with the effects of forest soil liming on streams and surface waters. The response of water chemistry is very dependent on hydrological and soil properties. Although pH itself may be little affected by liming, the acidity (or negative ANC) decreases, inorganic Al-species decrease and the Al/BC-ratio increases in the runoff water. Especially interesting is that this is also true during acid episodes. This means that toxicity for acid sensitive biota decreases. These results indicate that large scale liming may have beneficial effects on surface water chemistry. Furthermore, as surface waters are expected to respond to smaller decreases in acid deposition than do forests soils, forest soil liming may allow less frequent liming of lakes. Consequently, forest soil liming in combination with the anticipated emission reductions may have very beneficial results on surface waters in certain areas of Sweden.  相似文献

N20 emissions and denitrification N-losses. precipitation, air temperature, soil moisture, bulk density and content of mineral N were monitored in 9 different agricultural soils in 6 European countries throughout the vegetation period (April to September) 1992 and 1993. N2O emissions and denitrification N-losses were log-normal distributed, reflecting high temporal changes. While small flux rates (< 2 g N ha?1 d?1) were detectable every day, high rates (> 10 g N ha?1 d?1) were measured after fertilization. An attempt to relate the emission variables to climate and soil variables was made through the use of correlation analysis. The mean N20 emissions from soil were significantly correlated with the soil properties clay, organic C and mineral N content and the amount of applied mineral N fertilizer. The best prediction of the N2O emission rates (r2 = 0.734) was achieved by multiple linear regression using the soil parameter clay and mineral N. Only 50% of the observed variation could be explained by the factors Corg and mineral N, which describe the substrate availability for microbial processes. No successful statistical model was found for the prediction of denitrification N-losses.  相似文献

 以具有和不具有无毒基因Avr1表现型的马铃薯晚疫病菌株80029 (AVR1) 和88133 (avr1)以及其杂交F1代50个菌株群体为试材,限制性内切酶PstI和HhaI为酶切组合,采用荧光AFLP技术和混合分组分析法(BSA),通过256对引物筛选得到了5个与Avr1连锁的候选AFLP标记并进行了菌株个体验证,遗传分析表明5个标记中有2个标记与Avr1连锁且在F1代中共分离,无交换重组发生。  相似文献
