The purpose of the current investigation is to elucidate the pharmacokinetic profiles of orbifloxacin (OBFX) in lactating ewes (n = 6) following intravenous (i.v.) and intramuscular (i.m.) administrations of 2.5 mg/kg W. In a crossover study, frequent blood, milk, and urine samples were drawn for up to 48 h after the end of administration, and were then assayed to determine their respective drug concentrations through microbiological assay using Klebsiella pneumoniae as the test micro‐organism. Plasma pharmacokinetic parameters were derived from plasma concentration–time data using a compartmental and noncompartmental analysis, and validated a relatively rapid elimination from the blood compartment, with a slope of the terminal phase of 0.21 ± 0.02 and 0.19 ± 0.06 per hour and a half‐life of 3.16 ± 0.43 and 3.84 ± 0.59 h, for i.v. and i.m. dosing, respectively. OBFX was widely distributed with a volume of distribution V(d(ss)) of 1.31 ± 0.12 L/kg, as suggested by the low percentage of protein binding (22.5%). The systemic body clearance (ClB) was 0.32 ± 0.12 L/h·kg. Following i.m. administration, the maximum plasma concentration (Cmax) of 1.53 ± 0.34 μg/mL was reached at tmax 1.25 ± 0.21 h. The drug was completely absorbed after i.m. administration, with a bioavailability of 114.63 ± 11.39%. The kinetic milk AUCmilk/AUCplasma ratio indicated a wide penetration of orbifloxacin from the bloodstream to the mammary gland. OBFX urine concentrations were higher than the concurrent plasma concentrations, and were detected up to 30 h postinjection by both routes. Taken together, these findings indicate that systemic administration of orbifloxacin could be efficacious against susceptible mammary and urinary pathogens in lactating ewes.  相似文献

Optically active 1-α-methylbenzyl-3- p -tolylurea (MBTU) shows diverse plant physiological properties. Gramineae plants exhibit enantioselective root growth inhibition by R / S -MBTU. In the present paper, the dose–growth response (DGR) towards the optically active MBTUs was examined in the tribe Triticeae plants, including Hordeum vulgare L., Secale cereale L., Triticum aestivum L. and Aegilops cylindrica Host., and compared with those of the genus Bromus ( B. inermis Leyss), which is classified into the tribe Bromeae, and the genus Oryza . In spite of their phylogenetic difference, the Triticeae plants responded homogeneously in their chiral recognition. Of particular interest, is the result of the dichotomic enantioselective response of tested Gramineae plants. Only the genus Oryza responds to R -MBTU, and the tribes Triticeae ( Triticum , Aegilops , Secale and Hordeum ) and Bromeae ( Bromus ) preferentially respond to the S -enantiomer. The tetrazolium–phenazine methosulfate–formazan (TPF) assay was demonstrated to measure redox activity of root tips of the MBTU-treated Oryza sativa and T. aestivum , which are the typical dichotomic, enantioselective-responsive plants. In Oryza and Triticum root tips, enantioselective responses of reduction activity in TPF assays were strongly related to those of root growth inhibition in DGR assays, suggesting aggressive participation of the MBTU in the generation of the reducing mechanism in mitochondria.  相似文献

The target of the present study was to investigate the plasma disposition kinetics of levofloxacin in stallions (n = 6) following a single intravenous (i.v.) bolus or intramuscular (i.m.) injection at a dose rate of 4 mg/kg bwt, using a two‐phase crossover design with 15 days as an interval period. Plasma samples were collected at appropriate times during a 48‐h administration interval, and were analyzed using a microbiological assay method. The plasma levofloxacin disposition was best fitted to a two‐compartment open model after i.v. dosing. The half‐lives of distribution and elimination were 0.21 ± 0.13 and 2.58 ± 0.51 h, respectively. The volume of distribution at steady‐state was 0.81 ± 0.26 L/kg, the total body clearance (Cltot) was 0.21 ± 0.18 L/h/kg, and the areas under the concentration–time curves (AUCs) were 18.79 ± 4.57 μg.h/mL. Following i.m. administration, the mean t1/2el and AUC values were 2.94 ± 0.78 h and 17.21 ± 4.36 μg.h/mL. The bioavailability was high (91.76% ± 12.68%), with a peak plasma mean concentration (Cmax) of 2.85 ± 0.89 μg/mL attained at 1.56 ± 0.71 h (Tmax). The in vitro protein binding percentage was 27.84%. Calculation of efficacy predictors showed that levofloxacin might have a good therapeutic profile against Gram‐negative and Gram‐positive bacteria, with an MIC ≤ 0.1 μg/mL.  相似文献

运用3种多元分析方法,对黑龙江泥鳅MIsgurnus mohoity、北方泥鳅Misgurnus bipartitus和泥鳅Mis-gurnus anguillicaudatus的11个形态参数进行了综合分析。聚类分析结果表明:北方泥鳅和泥鳅聚类,亲缘关系较近,与黑龙江泥鳅群体的亲缘关系较远。主成分分析结果表明:主成分1的总方差贡献率为50.430%,主成分2的为20.880%,主成分3的为10.381%,累积贡献率为81.691%。判别分析结果表明:建立的3种泥鳅的判别函数,判别准确率P1为71.4%~100%,判别准确率P2为71.4%~100%,综合判别率为80.8%。  相似文献

Sixty-four isolates of Bean yellow mosaic virus (BYMV) from cultivated and naturalized gladioli were divided into two pathogenic groups, necrotic spot (NS) and chlorotic spot (CS) groups on Chenopodium quinoa. NS-type isolates (S-22N and E-24N), CS-type isolates (S-22C and E-92C), and broad bean isolates (Sb-50C and Sb-12C) differed in their pathogenicity on Antirrhinum majus, Nicotiana benthamiana, Phaseolus vulgaris, Spinacia oleracea and Vigna unguiculata. The four gladiolus isolates were different from BYMV-B, -P, -O and C1YVV-N in their pathogenicity on these plants, while the two broad bean isolates were similar to BYMV-B, originally from broad bean. The nucleotide (nt) sequences of the 3′-terminal region of the BYMV RNA genome of the two NS-type isolates, the two CS-type isolates, the two broad bean isolates and BYMV-B, -P and -O were determined. In a phylogenetic tree based on the CP amino acid (aa) sequences, the two NS-type isolates clustered together (identity 98.4% and 98.2% at the nt and aa level, respectively). The two CS-type isolates clustered with BYMV-O (93.2 to 99.3% nt identity and 95.6 to 98.5% aa identity). The two broad bean isolates clustered with BYMV-B (99.0 to 99.5% nt identity and 98.9 to 99.6% aa identity). BYMV-P clustered with BYMV-CS (identity 97.7% and 99.3% at the nt and aa level, respectively). The obtained sequences were compared with those of the 3′-terminal regions of seven published BYMV isolates. 以中国洪湖自然二倍体、三倍体、四倍体泥鳅Misgurnus anguillicaudatus为研究对象,运用经典的形态测量观察方法、染色体制片技术对不同倍性泥鳅的染色体组型、形态特征进行比较分析。结果表明:1)二倍体泥鳅2n=50,核型公式为10m+4sm+36t,NF=64;三倍体泥鳅3n=75,核型公式为15m+6sm+54t,NF=96;四倍体泥鳅4n=100,核型公式为20m+8sm+72t,NF=128;2)5个比例性状测量结果显示,二倍体与四倍体很接近,没有表现出明显的差异,但两者与三倍体在眼后头长/体长(PL/SL)、后头部到鳃的距离/体长(LOOSR/SL)、背鳍前端到尾鳍的距离/体长(LODO-CB/SL)3个比例性状存在显著差异(P〈0.05);3)主成分分析表明,二倍体、三倍体及四倍体形态差异主要系躯体后半部差异所致;(4)聚类分析结果显示,洪湖二倍体和四倍体相聚类,亲缘关系较近,与三倍体群体的聚类关系较远。  相似文献

辣椒炭疽病Collectotrichumspp.是韩国及中国辣椒的重要病害.比较了韩国炭疽病茵3个茵株和中国炭疽病菌3个菌株的形态学特征,并进行了致病性鉴定.结果如下:(1)根据形态学特征,K1和C1,K2和C2及K3和C3分别鉴定为ColletotrichumgloeosporloideG类型,C.gloeosporioidesR类型和C.coccodes;(2)韩国茵株与中国菌株的致病力之间没有明显的差异;(3)RAPD-PCR分析结果表明K1和C1,K2和C2,以及K3和C3的相似率分别为85.71%,71.43%,和50%.  相似文献

To assess factors for canine skin extensibility, our study investigated associations between the dogs’ skin extension index and the following factors, gender, age, neuter status, weight, coat color and six coat color related gene polymorphisms. Swab samples were collected from 69 toy poodles to extract DNA. The skin extension indices of the lower back and the neck were measured using the following formula: vertical height of the skin fold divided by body length multiplied by 100. The dogs’ age, weight, gender, neuter status and coat color were also recorded, as well as polymorphisms of the following six selected coat color related genes, Melanocortin 1 receptor, Tyrosinase-related protein 1, Melanophilin, Canine β-defensin-1, Major Facilitator Superfamily Domain Containing 12 and Agouti-signaling protein (ASIP). Univariable analysis showed there was a meaningful association between the lower back skin extension index and both gender and age (P<0.001 and P=0.048, respectively). Also, there was a possible association between the lower back skin extension index and ASIP Single nucleotide polymorphism (SNP) (R96C) (P=0.078). Linear model analysis showed there was a significant association between the lower back skin extension index and gender (P<0.001), and there was a tendency of the association between the lower back skin extension index and ASIP SNP (R96C) (P=0.098). In addition, there was an association between gender and age for the skin extension index. (P=0.048). Therefore, these results suggest that a greater risk of skin extensibility in toy poodle could be related to being female and the ASIP SNP (R96C), because these factors were associated with higher lower back skin extension index.  相似文献
