Fusarium verticillioides and Aspergillus flavus cause Fusarium ear rot (FER) and Aspergillus ear rot (AER) of maize, respectively. Both pathogens are of concern to producers as they reduce grain yield and affect quality. F. verticillioides and A. flavus also contaminate maize grain with the mycotoxins fumonisins and aflatoxins, respectively, which has been associated with mycotoxicosis in humans and animals. The occurrence of common resistance mechanisms to FER and AER has been reported. Hence, ten Kenyan inbred lines resistant to AER and aflatoxin accumulation were evaluated for resistance to FER, F. verticillioides colonisation and fumonisin accumulation; and compared to nine South African lines resistant to FER and fumonisin accumulation. Field trials were conducted at three localities in South Africa and two localities in Kenya. FER severity was determined by visual assessment, while F. verticillioides colonisation and fumonisin content were quantified by real-time PCR and liquid chromatography tandem mass spectrometry, respectively. Significant genotype x environment interactions was determined at each location (P ≤ 0.05). Kenyan inbred CML495 was most resistant to FER and F. verticillioides colonisation, and accumulated the lowest concentration of fumonisins across localities. It was, however, not significantly more resistant than Kenyan lines CML264 and CKL05015, and the South African line RO549 W, which also exhibited low FER severity (≤5%), fungal target DNA (≤0.025 ng μL?1) and fumonisin levels (≤2.5 mg kg?1). Inbred lines resistant to AER and aflatoxin accumulation appear to be promising sources of resistance to F. verticillioides and fumonisin contamination. 相似文献

Isolate 840905 of Cladosporium cucumerinum, when grown on agar or in liqiud medium, was sensitive to triadimenol, HWG 1608 (tebuconazole), fenpropimorph and pimaricin but relatively resistant to terbinafine. Conversely, isolate 49628 was sensitive to terbinafine but relatively resistant to the other fungicides. Changes in sterol composition following treatment with the fungicides reflected the known modes of action of each fungicide. When individual enantiomers of triadimenol were tested against isolate 840905 the order of activity in reducing mycelial growth was 1 S, 2R > 1R, 2R > 1R, 2S ≈? 1S, 2S, and this was paralleled by the depletion of ergosterol and the appearance of 14α-methyl sterols. Isolate 49628 had a greater saturated:unsaturated fatty acid ratio than did isolate 840905 but no major changes in fatty acid composition of either isolate were induced by fungicide treatment. 1993~1994年大麦云纹斑病菌(Rhynchosporium secalis)对杀菌剂多菌灵、三唑醇的抗药性监测表明,该菌对两种杀菌剂的敏感程度发生了变化,并且首次在田间发现了大麦云纹斑病菌的多抗菌株。这类菌株对多菌灵、乙霉威及三唑醇都具有抗药性,且抗性程度很高。与1990~1991年得到的资料相比,随着杀菌剂选择压力的增加,大田群体中三唑醇高抗菌株比例逐年增加。对该菌同工酶分析结果表明,α—酯酶有7种表现型,磷酸葡萄糖变位酶有2种表现型,过氧化氢酶有3种表现型,无论对去甲基抑制剂类(DMI)表现抗性或敏感菌株,无论调节酶还是非调节酶,其酶谱表现型的变异基本相似。这表明大麦云纹斑病菌对DMI类抗药性突变存在于多个菌系的基因型背景群体中,而不是只起源于单一的广泛分布的菌系。 相似文献

Plasma fentanyl concentrations in awake cats and cats undergoing anesthesia and ovariohysterectomy using transdermal administration 总被引:1,自引:0,他引:1
Objective To measure the plasma fentanyl concentrations achieved over time with transdermal fentanyl patches in awake cats and cats undergoing anesthesia and ovariohysterectomy. Study design Randomized prospective experimental study. Animals Twenty‐four purpose‐bred cats. Methods Cats were randomly assigned to three groups for Part I of a larger concurrent study. Group P received only a 25 μg hour?1 transdermal fentanyl patch. Group P/A received the patch and anesthesia. Group A received only anesthesia. After a minimum 1‐week washout period, the cats were randomly reassigned to two groups for Part II of the larger study. Group P/A/O received the patch, anesthesia and ovariohysterectomy. Group A/O received anesthesia and ovariohysterectomy. Patches were left in place for 72 hours and plasma samples were obtained for fentanyl analysis while the patches were in place, and for 8 hours after patch removal for cats in Group P, P/A, and P/A/O. Results The 25 μg hour?1 transdermal fentanyl patches were well tolerated by the cats in this study (mean body weight of 3.0 kg) and no overt adverse effects were noted. Mean plasma fentanyl concentrations over time, mean plasma fentanyl concentrations at specific times (8, 25, 49, and 73 hours after patch placement), time to first detectable plasma fentanyl concentration, time to reach maximum plasma fentanyl concentration, maximum plasma fentanyl concentration, mean plasma fentanyl concentration from 8 to 73 hours, elimination half‐life, and total area under concentration (AUC) were not statistically different among the groups. Conclusions Halothane anesthesia and anesthesia/ovariohysterectomy did not significantly alter the plasma fentanyl concentrations achieved or pharmacokinetic parameters measured, when compared with awake cats. There was a high degree of individual variability observed both within and between groups of cats in parameters measured. Clinical significance The high degree of variability observed suggests that careful observation of cats with fentanyl patches in place is required to assess efficacy and any potential adverse effects. Anesthesia and anesthesia/ovariohysterectomy do not appear to alter plasma fentanyl concentrations achieved by placement of a 25 μg hour?1 transdermal fentanyl patch when compared to cats not undergoing these procedures. 相似文献

Thirty-eight dogs with orthopedic disorders received etodolac, an NSAID, at 10.0 to 13.3 mg/kg PO once daily for 14 to 19 days. Mean total thyroxine (T4), free thyroxine (fT4), and canine thyrotropin (cTSH) values before and after etodolac administration were compared using paired t-tests. A significant (P <.05) decrease in T4 values occurred after etodolac administration with 21% of these values falling below the reference range. A significant (P <.05) increase in cTSH following etodolac administration, but none of the values was above the reference range. No significant changes occurred in mean fT4 values; however, 10% of the values fell below the reference range. In conclusion, T4 and fT4 test results should be interpreted with caution in dogs receiving etodolac. 相似文献

Photoperiod manipulation, specifically a long-day photoperiod (LDPP), increases milk production in lactating cattle. We have previously reported that the galactopoietic effect of LDPP is associated with an increase in circulating IGF-I, which seems to occur independently of changes in concentrations of GH, IGFBP-2, and IGFBP-3. This study tested the hypothesis that LDPP increases the expression of GH receptor (GHR) 1A messenger RNA (mRNA) in the liver. Two groups of Holstein steer calves (98 +/- 4 d old) were maintained indoors and exposed to LDPP (16-h light: 8-h dark; n = 6) or short-day photoperiod (SDPP; 8-h light: 16-h dark; n = 6) for 60 d. Calves were individually fed a grain- and alfalfa-based diet. Jugular blood samples were collected weekly and via cannula at 15-min intervals for a 4-h period on d 1, 26, and 55 of the study to monitor pulsatile hormone secretion. Serum was harvested and assayed for IGF-I, prolactin (PRL), and GH using RIA. Liver biopsies were obtained at 3-wk intervals to quantify changes in hepatic IGF-I and GHR 1A mRNA using real-time PCR. Steer BW increased during the study but did not differ between treatments. No differences in ADG or total DMI were observed. Relative to SDPP, calves on LDPP had higher (P < 0.05) serum IGF-I concentrations. Concentrations of PRL increased (P < 0.01) in calves exposed to LDPP compared with calves exposed to SDPP. Differences (P < 0.05) in pulsatile GH secretion were also detected. Hepatic IGF-I and GHR 1A mRNA were positively correlated with circulating IGF-I concentrations, and although both increased with time, they were not affected by photoperiod treatment. These results confirm that LDPP increases circulating concentrations of IGF-I, but this occurs independently of changes in IGF-I synthesis and GHR 1A mRNA expression in the liver. Salmonella gastroenteritis and septicemia were diagnosed in two cats presented for necropsy. Both cats resided in the same household and were fed a home-prepared, raw meat-based diet. Salmonella was isolated from multiple organs in both cats and from samples of raw beef incorporated into the diet fed to one of the cats. Subtyping of the bacterial isolates yielded Salmonella newport from one cat and from the diet it had been fed. Research was conducted in northern Colorado in 2011 to estimate the crop water stress index (CWSI) and actual transpiration (T a) of maize under a range of irrigation regimes. The main goal was to obtain these parameters with minimum instrumentation and measurements. The results confirmed that empirical baselines required for CWSI calculation are transferable within regions with similar climatic conditions, eliminating the need to develop them for each irrigation scheme. This means that maize CWSI can be determined using only two instruments: an infrared thermometer and an air temperature/relative humidity sensor. Reference evapotranspiration data obtained from a modified atmometer were similar to those estimated at a standard weather station, suggesting that maize T a can be calculated based on CWSI and by adding one additional instrument: a modified atmometer. Estimated CWSI during four hourly periods centered on solar noon was largest during the 2 h after solar noon. Hence, this time window is recommended for once-a-day data acquisition if the goal is to capture maximum stress level. Maize T a based on CWSI during the first hourly period (10:00–11:00) was closest to T a estimates from a widely used crop coefficient model. Thus, this time window is recommended if the goal is to monitor maize water use. Average CWSI over the 2 h after solar noon and during the study period (early August to late September, 2011) was 0.19, 0.57, and 0.20 for plots under full, low-frequency deficit, and high-frequency deficit irrigation regimes, respectively. During the same period (50 days), total maize T a based on the 10:00–11:00 CWSI was 218, 141, and 208 mm for the same treatments, respectively. These values were within 3 % of the results of the crop coefficient approach. 相似文献
