Pulses, the most important source of high-quality dietary proteins are prone to serious damage due to insect pests, of which 20–25% damage was inflicted by stored product bruchid pest, Callosobruchus maculatus (F.). An alternative approach to control these insect pests apart from fumigation is the use of primary metabolites involved in plant defense mechanism. In the present study, one of such molecule, an insect anti-metabolic protein, arcelin was screened from nine different wild and cultivated varieties of pulses. Insect feeding bioassay with C. maculatus revealed that seven of these seed varieties were not infested by the bruchid insect pest. Additionally, the presence of arcelin polypeptide subunits (27–42 kDa) was demonstrated in the seeds of Phaseolus limensis L. and Phaseolus lunatus L. whereas, an insignificant amount was observed in Lablab purpureus L. Besides, seed kernel extract of P. lunatus registered a highest HA activity against native human-A erythrocytes and this activity was increased to several fold when the erythrocytes were treated with proteolytic enzymes revealing the characteristic feature of arcelin. Similarly, HA activity was observed when rat and mouse erythrocyte was treated with pronase, whereas, no titer value was observed upon treatment with its native erythrocytes. Of the 24 carbohydrates tested, only acetylated amino sugar, N-acetyl-d-galactosamine inhibited the agglutinating activity of crude seed extract against human-A RBC. Among the three glycoproteins tested, none were found potential to competitively inhibit the agglutination activity against human-A erythrocytes. Overall, this study reveals the presence of arcelin, an anti-metabolic insecticidal protein in seeds of P. lunatus.  相似文献

The rice brown planthopper (BPH) Nilaparvata lugens (Stål) is one of the major pests of rice across Asia. Host-plant resistance is the most ecologically acceptable means to manage this pest. A rice breeding line RP2068-18-3-5 (RP2068) derived from the land race Velluthacheera is reported to be resistant to BPH populations across India. We identified a new R gene [Bph33(t)] in this line using advanced generation RILs derived from TN1 × RP2068 cross through phenotyping at two locations and linkage analysis with 99 polymorphic SSR markers. QTL analysis through IciMapping identified at least two major QTL on chromosome 1 influencing seedling damage score in seed box screening, honey dew excretion by adults and nymphal survival. Since no BPH R gene has been reported on chromosome 1, we designate this locus as a new gene Bph33(t) which accounted for over 20% of phenotypic variance. Scanning the region for candidate gene suggested two likely candidates a leucine rich repeat (LRR) gene and a heat shock protein (HSP) coding gene. Expression profiling of the two genes in the two contrasting parents and RILs showed induction of the HSP gene (LOC_Os01g42190.1) at 6 h after infestation while LRR gene did not show such induction. Seed size, determined by 100-seed weight, is an important yield component and trade value trait in kabuli chickpea. In the present investigation, the small seeded kabuli genotype ICC 16644 was crossed with four genotypes (JGK 2, KAK 2, KRIPA and ICC 17109) and F1, F2 and F3 populations were developed to study the gene action involved in seed size and other yield attributing traits. Scaling test and joint scaling test revealed the presence of epistasis for days to first flower, days to maturity, plant height, number of pods per plant, number of seeds per plant, number of seeds per pod, biological yield per plant, grain yield per plant and 100-seed weight. Additive, additive?×?additive and dominance?×?dominance effects were found to govern days to first flower. Days to maturity and plant height were under the control of both the main as well as interaction effects. Number of seeds per pod was predominantly under the control of additive and additive?×?additive effects. For grain yield per plant, additive and dominance?×?dominance effects were significant in the cross ICC 16644?×?KAK 2, whereas, additive?×?additive effects were important in the cross ICC 16644?×?JGK 2. Additive, dominance and epistatic effects influenced seed size. The study emphasized the existence of duplicate epistasis for most of the traits. To explore both additive and non-additive gene actions for phenological traits and yield traits, selection in later generations would be more effective.  相似文献

Two major bacterial blight(BB) resistance genes(Xa21 and xa13) and a major gene for blast resistance(Pi54) were introgressed into an Indian rice variety MTU1010 through marker-assisted backcross breeding. Improved Samba Mahsuri(possessing Xa21 and xa13) and NLR145(possessing Pi54) were used as donor parents. Marker-assisted backcrossing was continued till BC2 generation wherein PCR based functional markers specific for the resistance genes were used for foreground selection and a set of parental polymorphic microsatellite markers were used for background selection at each stage of backcrossing. Selected BC2F1 plants from both crosses, having the highest recoveries of MTU1010 genome(90% and 92%, respectively), were intercrossed to obtain intercross F1(ICF1) plants, which were then selfed to generate 880 ICF2 plants possessing different combinations of the BB and blast resistance genes. Among the ICF2 plants, seven triple homozygous plants(xa13xa13Xa21Xa21Pi54Pi54) with recurrent parent genome recovery ranging from 82% to 92% were identified. All the seven ICF2 plants showed high resistance against the bacterial blight disease with a lesion lengths of only 0.53–2.28 cm, 1%–5% disease leaf areas and disease scoring values of ‘1' or ‘3'. The seven ICF2 plants were selfed to generate ICF3, which were then screened for blast resistance, and all were observed to be highly resistant to the diseases. Several ICF3 lines possessing high level of resistance against BB and blast, coupled with yield, grain quality and plant type on par with MTU1010 were identified and advanced for further selection and evaluation.  相似文献

Utilization of shrimp head waste from the processing industry offers a solution to environmental pollution and also produces valuable bioactive peptides. The present study was aimed to optimize the alcalase assisted extraction conditions by response surface methodology (RSM) to obtain protein hydrolysates from shrimp (Metapenaeus dobsoni) head waste with maximum antioxidant activity. Based on the models derived by RSM, the optimized conditions for maximum degree of hydrolysis (DH) were pH 8.2, temperature of 45.4°C, and enzyme/substrate (E/S) of 1.8%. Meanwhile, the optimized conditions for maximum antioxidant activity of protein hydrolysates as measured by 2, 2-diphenyl-1-picrylhydrozyl (DPPH) and ferric reducing antioxidant power (FRAP) assays were pH 8.2 and 8.1, temperatures of 49.7 and 59.4°C, and E/S of 2.1 and 2.7%, respectively. The maximum predicted values for DH, DPPH, and FRAP were 42.44%, 39.64% inhibition, and 8.3 μM Fe (II)/g wet wt. of sample, respectively. The extraction of protein hydrolysates at optimal conditions derived by these models resulted in DH, DPPH, and FRAPs of 40.31%, 38.93%, and 8.21 μM Fe (II)/g of sample, respectively, demonstrating the fitness of the models. The present study demonstrates the potential of Metapenaeus dobsoni head waste as a source of protein hydrolysates with prospective antioxidant activity.  相似文献

Methylobacterium oryzae CBMB20, a promising plant growth promoting bacteria (PGPB) and a biocontrol agent, was immobilized in different formulations such as wet chitosan, dry chitosan, wet alginate and dry alginate and were tested for tomato plant growth promotion. Chitosan solution (1.5%) with pH 5.5–6.0 and 90 min contact time was found optimal for immobilization. The chitosan formulations showed better entrapment efficiency and good degradability resistance apart from slow release of cells under prolonged incubation. Survivability of bacteria (80%) was observed in wet chitosan formulation even after 90 days of storage at 4°C. The spermosphere survival of bacteria was high in both dry and wet chitosan formulations applied soils even after 21 days under greenhouse conditions. While the alginate formulation degraded fully, partial degradation of chitosan formulation was observed even after 30 days, indicating its ability to support the survival of M. oryzae CBMB20 in soil. Plants inoculated with wet chitosan formulation registered 1.3 fold increase in the shoot and root length and dry weight compared to other treatments. Hence, chitosan formulation supporting better plant growth compared to alginate will be a better carrier for taking bacteria to the plant rhizosphere and thereby promote plant growth.  相似文献

This study was conducted to determine if metabolites of mexacarbate contribute to its residual toxicity to spruce budworm (Choristoneura fumiferana) larvae. Potted white spruce (Picea glauca) trees were treated with ‘Zectran’ UCZF # 19 at 100 g a.i. ha?1 and kept in a glasshouse. Mexacarbate residues declined by 98–99% within three days and reached non-detectable levels 10 days after treatment. Mortality of larvae fed on buds from these trees declined more gradually and was still 19–27% when exposed 10 days after treatment. The very low levels of mexacarbate (<0.07 μg g?1) found after three days did not produce such mortality. Gas chromatographic analysis of metabolites in needles revealed that after three days, 4-methylformamido-3,5-xylyl N-methylcarbamate was present at levels 20–30 times higher than the parent compound. This metabolite was about 50 times less toxic than mexacarbate to larvae when applied topically but was only 7 times less toxic when ingested. Two other methylcarbamate metabolites, the amino, and methylamino analogues were detectable for one day following treatment but not at later time points. They were as toxic as mexacarbate both topically and orally. Based on these findings, the methylformamido analogue could contribute to the residual toxicity of mexacarbate treatments of spruce.  相似文献

The viscosity, surface tension and volatility of a range of ultra-low-volume (ULV) spray diluents and pesticide formulations were measured at 5°C and 20°C. For u.l.v. application of 1.0 to 1.5 litre ha?1 through conventional boom and nozzle systems or rotary (Micronair) atomisers, it is concluded that the spray medium should have a viscosity of ?30 mPa s at 20°C. The surface tension values covered only a narrow range and showed little temperature dependence. There was no clear optimum and all surface tensions within the range measured would appear to be acceptable for ULV applications. The volatility factor, 1/(A.T1/2), where A represents the percentage of non-volatile material in the spray mixture and T1/2, the half-life (minutes) of evaporation, should be <40 times; 10?5.  相似文献
