Bovine viral diarrhoea virus (BVDV) is an economically important pathogen of cattle worldwide. Infection of a pregnant animal may lead to persistent infection of the foetus and birth of a persistently infected (PI) calf that sheds the virus throughout its life. However, BVD viruses are not strictly species specific. BVDV has been isolated from many domesticated and wild ruminants. This is of practical importance as virus reservoirs in non-bovine hosts may hamper BVDV control in cattle. A goat given as a social companion to a BVDV PI calf gave birth to a PI goat kid. In order to test if goat to goat infections were possible, seronegative pregnant goats were exposed to the PI goat. In parallel, seronegative pregnant goats were kept together with the PI calf. Only the goat to goat transmission resulted in the birth of a next generation of BVDV PI kids whereas all goats kept together with the PI calf aborted. To our knowledge, this is the first report which shows that a PI goat cannot only transmit BVD virus to other goats but that such transmission may indeed lead to the birth of a second generation of PI goats. Genetic analyses indicated that establishment in the new host species may be associated with step-wise adaptations in the viral genome. Thus, goats have the potential to be a reservoir for BVDV. However, the PI goats showed growth retardation and anaemia and their survival under natural conditions remains questionable.  相似文献

High resolution Fourier transform mass spectrometry (HRFTMS) and nuclear magnetic resonance (NMR) spectroscopy were employed as complementary metabolomic tools to dereplicate the chemical profile of the new and antitrypanosomally active sponge-associated bacterium Actinokineospora sp. EG49 extract. Principal Component (PCA), hierarchical clustering (HCA), and orthogonal partial least square-discriminant analysis (OPLS-DA) were used to evaluate the HRFTMS and NMR data of crude extracts from four different fermentation approaches. Statistical analysis identified the best culture one-strain-many-compounds (OSMAC) condition and extraction procedure, which was used for the isolation of novel bioactive metabolites. As a result, two new O-glycosylated angucyclines, named actinosporins A (1) and B (2), were isolated from the broth culture of Actinokineospora sp. strain EG49, which was cultivated from the Red Sea sponge Spheciospongia vagabunda. The structures of actinosporins A and B were determined by 1D- and 2D-NMR techniques, as well as high resolution tandem mass spectrometry. Testing for antiparasitic properties showed that actinosporin A exhibited activity against Trypanosoma brucei brucei with an IC50 value of 15 µM; however no activity was detected against Leishmania major and Plasmodium falciparum, therefore suggesting its selectivity against the parasite Trypanosoma brucei brucei; the causative agent of sleeping sickness.  相似文献

Background

The tolerance of plants against abiotic stresses can be greatly influenced by their interaction with microbes. In lowland rice (Oryza sativa) production, the iron toxicity of the soils constitutes a major constraint. Although there are tolerant cultivars, the mechanisms underlying the tolerance against excess iron are not fully understood. Even less is known about the role of microbes in the response to iron toxicity.

Aim

In the study presented here, the effects of different Bacillus isolates on the accumulation and distribution of iron within the shoots of different rice cultivars grown under iron toxicity were analyzed.

Methods

Three lowland rice cultivars with contrasting tolerance to iron toxicity (IR31785-58-1-2-3-3, Sahel 108, Suakoko 8) were inoculated with three Bacillus isolates (two B. pumilus and one B. megaterium) and, after 1 week, exposed to excess iron (1,000 ppm) for 8 days. Tolerance was evaluated by leaf symptom scoring.

Results

Bacterial inoculation mitigated leaf symptoms in the sensitive cultivar IR31785-58-1-2-3-3 despite no significant differences in shoot iron concentration between inoculated and noninoculated plants. In the tolerant excluder cultivar, Suakoko 8, leaf symptoms were exacerbated when inoculated with B. pumilus Ni9MO12. While the total shoot Fe concentration was not affected in this bacteria × cultivar combination, the distribution of iron within the shoot was clearly disturbed. Tolerance to iron toxicity of the tolerant includer cultivar, Sahel 108, was not affected by Bacillus inoculation.

Conclusion

In conclusion, our results show that Bacillus inoculation can affect the tolerance of lowland rice to iron toxicity and that the effects strongly depend on the bacteria × cultivar combination.  相似文献

Pork products are a possible source of introduction of PCV2 isolates into a pig population. However, limited work has been done on the transmission through meat of porcine circovirus type 2 (PCV2), a virus associated with several disease syndromes in pigs. The objectives of this study were to determine if pork products from PCV2-infected pigs contain PCV2 DNA/antigen and to determine if the PCV2 present in the tissues is infectious by performing in vitro and in vivo studies. Skeletal muscle, bone marrow, and lymphoid tissues from pigs experimentally inoculated with PCV2 were collected 14 days post-inoculation (DPI). The tissues were tested for presence of PCV2 DNA by quantitative real-time PCR, for PCV2 antigen by immunohistochemistry (IHC), and for presence of infectious PCV2 by virus isolation and inoculation of PCV2 naïve pigs. Lymphoid tissues contained the highest amount of PCV2 (positive by PCR, IHC, and virus isolation), bone marrow contained a lower amount of PCV2 (positive by PCR and IHC but negative by virus isolation), and skeletal muscle contained the lowest amount of PCV2 (positive by PCR but negative by IHC and virus isolation). Naïve pigs fed for three consecutive days with either skeletal muscle, bone marrow, or lymphoid tissues all became PCV2 viremic as determined by quantitative real-time PCR on serum starting at 7 DPI. The pigs also seroconverted to PCV2 as determined by PCV2 IgM and IgG ELISA. In addition, PCV2 antigen was detected by IHC stains in lymphoid tissues and intestines collected from the majority of these pigs. Results from this study indicate that uncooked PCV2 DNA positive lymphoid tissues, bone marrow, and skeletal muscle from PCV2 viremic pigs contain sufficient amount of infectious PCV2 to infect naïve pigs by the oral route.  相似文献

The mechanical and viscoelastic properties of intact wheat kernels of 36 wheat cultivars differing in low molecular weight glutenin subunit (LMW‐GS) composition (loci Glu‐A3, Glu‐B3, and Glu‐D3) were evaluated using load‐compression tests. Comparison among genotypic groups representing Glu‐3 allelic variants showed that groups representing the alleles Glu‐A3 b, c, and d; Glu‐B3 d, g, and h; and Glu‐D3 a, b, and d, had harder kernel texture, higher kernel elastic work and larger gluten strength‐related parameters than those possessing alleles Glu‐A3 e; Glu‐B3 f, i and j (translocation 1B/1R); and Glu‐D3 d. Modulus of elasticity (stress to strain ratio) showed low values (111.9–168.8 MPa) for allelic groups possessing poor elastic properties (Glu‐A3 e; Glu‐B3 f, i, and j; and Glu‐D3 d), and high values (179.8–222.6 MPa) for allelic groups possessing high kernel elastic properties (Glu‐A3 b c, and d; Glu‐B3 d, g, and h; and Glu‐D3 a, b and c). The highest values for gluten strength‐related parameters (SDS‐sedimentation, dough mixing time, and dough strength [W]) corresponded to allelic groups Glu‐A3 d; Glu‐B3 d and g; and Glu‐D3 d, while the lowest corresponded to Glu‐A3 e and Glu‐B3 j. No significant differences were observed among groups with regard to gluten extensibility parameters; however, the highest P/L value (least extensibility) corresponded to Glu‐B3 j, which indicates presence of 1B/1R translocation. Except for the Glu‐B3 j (translocation 1B/1R) allele, which presented more variation within samples, a general relationship between kernel viscoelastic properties and dough viscoelastic properties was observed; samples showing higher elastic work to plastic work ratio (E/P) tended to possess better gluten strength than cultivars with low E/P ratio.  相似文献
